Študovala sa možnost tvorby primárnej membrány ako i sekundárnej retardačnej bariéry po zhustení mikročástic obalených vodnou disperziou etylcelulózy systémom Surelease fluidnou technikou (Aeromatic). Ukázalo sa, že na transformáciu práškovitého liečiva na dobre listovateľné aglomeráty je potrebný pomerne vysoký obsah polyméru (30 %). Významnejšia retardácia sa u matríc z obalených peliet dosiahla pri 20 a 30 % obsaJiu polyméru. Matrice formované z mikrodražé vykazujú výrazné spomalenie rýchlosti liberácie liečiva už pri 10 % obsahu prípravku Surelease a potvrdzujú retardačnú schopnosť etylcelulózového latexu.
The paper studies the possible formation of the primary membrane and the secondary retarding barrier after thickening the micropartides coated with an aqueous dispersion of ethylcellulose using the system Surelease with the fluid technique (Aeromatic). It has been found that for the transformation of the powdered drug into well-compressible agglomerates a relatively high content of the polymer (30 %) is needed. More significant retardation in the matrices made of coated pellets was achieved with contents of 20 % and 30 % of the polymer. Matrices formed from coated microtablets show marked deceleration of drug liberation rate already with a content of 10 % of the preparation Surelease and they confirm the retardii^ ability of ethylcellulose latex.

Obalené pelety si v nedávné době získaly značnou pozornost farmaceutického průmyslu, neboť představují relativně snadný způsob, jak dosáhnout řízeného uvolňování léčivých látek. Pelety vhodných jakostních parametrů s obsahem 40 % diklofenaku sodné soli se připravily rotační aglomerací. Jako obalový materiál se vybraly dvě odlišné vodné polymerové disperze – Surelease® a Eudragit®RS 30 D. Rychlost uvolňování léčivé látky se obecně s rostoucím množstvím naneseného obalu zpomalovala, nicméně disoluční profily pelet obalených ekvivalentním množstvím Surelease ® nebo Eudragitu® RS 30 D se výrazně odlišovaly. Zatímco obalování Eudragitem® RS 30 D vedlo k tvorbě membrán účinně řídících uvolňování léčivé látky po dobu 24 hodin, při obalování vodnou disperzí ethylcelulosy vznikal nekvalitní film. Rychlejší uvolňování diklofenaku sodné soli z pelet opatřených ethylcelulosovým obalem by mohla vysvětlovat vyšší rozpustnost diklofenaku sodné soli v zásadité vodné disperzi ethylcelulosy a následná migrace účinné látky do formujícího se obalu a/nebo nedostatečné tepelné ošetření ethylcelulosového filmu. Výsledky této studie naznačují, že je vždy nutné zvážit možné interakce mezi obalem a obsaženou účinnou látkou a složení obalu se musí velmi pečlivě navrhnout.
Recently, coated pellets have gained attention of the pharmaceutical industry as they represent a relatively easy way leading to controlled drug release. Pellets of appropriate properties containing approx. 40 % of diclofenac sodium were prepared by roto-agglomeration. For the coating, two different aqueous dispersions (Surelease® and Eudragit®RS 30 D) were selected. Generally, the drug release rate slowed down as the coating load increased from 10 to 22 %. However, big differences between diclofenac sodium release from pellets coated with Surelease® and Eudragit®RS30D at equivalent coating loads were observed. Although Eudragit®RS 30 D provided membranes successfully controlling drug release over an extended period of 24 hours, the coating process with Surelease® led to a film of a very poor quality. Faster release from ethyl cellulose coated pellets could be explained in terms of a higher solubility of diclofenac sodium in alkaline aqueous dispersion of ethyl cellulose and its migration into the coat during the coating process and/or the unsatisfactory curing of the ethyl cellulose film. Therefore possible interactions between the coating and the drug should be always considered as suggested in this study.

Patients tend to evade the occurrence of hypoglycemic episodes by excessive carbohydrate intake. Glucose pellets with delayed release in the time of the maximum effect of insulin can not only prevent hypoglycemia but also eliminate the preventive carbohydrate intake. The pellets can be administered in a mixture with semisolid food. The cores containing glucose in combination with osmotically active agents (croscarmellose sodium, carmellose sodium, polyethylene glycol, or carboxymethyl starch) were prepared by extrusion-spheronization and coated with 15% water ethylcellulose dispersion (Surelease® B NF) in Wurster column (Medipo, Havlíčkův Brod, Czech Republic) into four coating levels (12.5, 25, 35, and 50%). Mean particle size is 0.63-0.73 for cores and 0.82-0.98 for coated pellets. Cores and coated pellets have excellent or good flow properties according to Hausner ratio and Carr index. Aspect ratio ranges from 1.78 to 2.17 for cores and from 1.73 to 2.31 for coated pellets. Dissolution was performed using pH-independent method and method with continual change of pH. The suitable pH-independent release was achieved in the samples containing carboxymethyl starch or polyethylene glycol. Glucose release is enabled by a membrane rupture caused by core swelling. It can be, therefore, assumed that the glucose release profile will not be affected by food or transit time.
