Lactobacilli in the vaginal tract are essential to protect against microbial infections. We therefore focused on isolating vaginal lactobacilli from pregnant women and testing their functional properties. Lactobacilli were isolated from 50 vaginal swabs and the purified isolates were identified by MALDI-TOF MS. Functional properties (antimicrobial activity, organic acids and hydrogen peroxide production, antibiotic susceptibility, auto-aggregation, and hydrophobicity) of selected isolates were tested. Lactobacilli (41 strains) were identified in 58% of swabs with a predominance of Lactobacillus crispatus (48%) followed by L. jensenii (21%), L. rhamnosus (14%), L. fermentum (10%), and L. gasseri (7%). The highest antibacterial activity was determined for L. fermentum and L. rhamnosus. Strong anti-Candida activity was observed for strains L. crispatus, L. fermentum, and L. rhamnosus. Strain L. jensenii 58C possessed the highest production of hydrogen peroxide (6.32 ± 0.60 mg/l). The best lactic acid producer was strain L. rhamnosus 72A (11.6 ± 0.2 g/l). All strains were resistant to fluconazole and metronidazole. The highest auto-aggregation was observed for strain L. crispatus 51A (98.8 ± 0.1% after 24 h). Strain L. rhamnosus 68A showed the highest hydrophobicity (69.1 ± 1.4%). Strains L. fermentum and L. rhamnosus showed high antibacterial activity and hydrophobicity, and strains L. crispatus possessed high auto-aggregation and anti-Candida activity. Thus, these strains alone or in a mix could be used for the preparation of probiotic products for treatment and prevention of vulvovaginal infections of pregnant and non-pregnant women.

Nosocomial infections are an important cause of morbi-mortality worldwide. The increase in the rate of resistance to conventional drugs in these microorganisms has stimulated the search for new therapeutic options. The nitro moiety (NO2) is an important pharmacophore of molecules with high anti-infective activity. We aimed to synthesize new nitro-derivates and to evaluate their antibacterial and anti-Candida potential in vitro. Five compounds [3-nitro-2-phenylchroman-4-ol (3); 3-nitro-2-phenyl-2H-chromene (4a); 3-nitro-2-(4-chlorophenyl)-2H-chromene (4b); 3-nitro-2-(4-fluorophenyl)-2H-chromene (4c), and 3-Nitro-2-(2,3-dichlorophenyl)-2H-chromene (4d)] were efficiently synthesized by Michael-aldol reaction of 2-hydroxybenzaldehyde with nitrostyrene, resulting in one β-nitro-alcohol (3) and four nitro-olefins (4a-4d). The antibacterial and anti-Candida potentials were evaluated by assaying minimal inhibitory concentration (MIC), minimum fungicidal concentration (MFC), and minimum bactericidal concentration (MBC). Mono-halogenated nitro-compounds (4b and 4c) showed anti-staphylococcal activity with MIC values of 15.6-62.5 μg/mL and MBC of 62.5 μg/mL. However, the activity against Gram-negative strains was showed to be considerably lower and our data suggests that this effect was associated with the outer membrane. Furthermore, nitro-compounds 4c and 4d presented activity against Candida spp. with MIC values ranging from 7.8-31.25 μg/mL and MFC of 15.6-500 μg/mL. In addition, these compounds were able to induce damage in fungal cells increasing the release of intracellular material, which was associated with actions on the cell wall independent of quantitative changes in chitin and β-glucan. Together, these findings show that nitro-compounds can be exploited as anti-staphylococcal and anti-Candida prototypes.

The genus Candida is well known for its significant pathogenesis with which it radically affects the medical practice nowadays. C. albicans, above all, belongs to severe pathogens and it developed high resistance to a variety of antimicrobials. Undesirable effects of this species cast a shadow upon the entire group of these microorganisms, favourable metabolic qualities of which are thus often omitted. These properties find their usage in various fields of biotechnology. The present contribution is specifically focused on the application of genus Candida in traditional or recombinant biotechnology. Its pathogenesis is presented in contrast to the potential use of these yeasts in the industry.
Candida krusei is a pathogenic yeast species that is phylogenetically outside both of the well-studied yeast groups, whole genome duplication and CUG. Like all other yeast species, it needs to accumulate high amounts of potassium cations, which are needed for proliferation and many other cell functions. A search in the sequenced genomes of nine C. krusei strains revealed the existence of two highly conserved genes encoding putative potassium uptake systems. Both of them belong to the TRK family, whose members have been found in all the sequenced genomes of species from the Saccharomycetales subclade. Analysis and comparison of the two C. krusei Trk sequences revealed all the typical features of yeast Trk proteins but also an unusual extension of the CkTrk2 hydrophilic N-terminus. The expression of both putative CkTRK genes in Saccharomyces cerevisiae lacking its own potassium importers showed that only CkTrk1 is able to complement the absence of S. cerevisiae's own transporters and provide cells with a sufficient amount of potassium. Interestingly, a portion of the CkTrk1 molecules were localized to the vacuolar membrane. The presence of CkTrk2 had no evident phenotype, due to the fact that this protein was not correctly targeted to the S. cerevisiae plasma membrane. Thus, CkTrk2 is the first studied yeast Trk protein to date that was not properly recognized and targeted to the plasma membrane upon heterologous expression in S. cerevisiae.

Yeasts, like other microorganisms, create numerous types of multicellular communities, which differ in their complexity, cell differentiation and in the occupation of different niches. Some of the communities, such as colonies and some types of biofilms, develop by division and subsequent differentiation of cells growing on semisolid or solid surfaces to which they are attached or which they can penetrate. Aggregation of individual cells is important for formation of other community types, such as multicellular flocs, which sediment to the bottom or float to the surface of liquid cultures forming flor biofilms, organized at the border between liquid and air under specific circumstances. These examples together with the existence of more obscure communities, such as stalks, demonstrate that multicellularity is widespread in yeast. Despite this fact, identification of mechanisms and regulations involved in complex multicellular behavior still remains one of the challenges of microbiology. Here, we briefly discuss metabolic differences between particular yeast communities as well as the presence and functions of various differentiated cells and provide examples of the ability of these cells to develop different ways to cope with stress during community development and aging.
Problematika vzniku bakteriálních a kvasinkových infekcí s komplikovaným průběhem léčby, způsobeným rezistencí původců těchto onemocnění, je častým tématem výzkumu na poli humánní i veterinární medicíny. V této studii jsme se zaměřili na studium účinku selenových nanočástic ve formě gelovitého komplexu s polymerní látkou Cekolem (SeNPs), jako možné alternativy antibiotických léčiv při eliminaci bakteriálních a kvasinkových infekcí. Testování komplexu probíhalo na bakteriálních a kvasinkových izolátech získaných z kohorty 450 pa-cientů Oddělení traumatologie Úrazové nemocnice v Brně, od kterých byl odebrán vždy jeden vzorek stěru z povrchové kožní infekce, a doposud se podařilo identifikovat celkem 74 poddruhů bakteriálních kmenů nebo kvasinek. K identifikaci kmenů a testování SeNPs bylo využito mikrobiologických a molekulárně-biologických metod a metody hmotnostní spektrometrie. Studie prokázala výrazný vliv tohoto přípravku na bakteriální kmeny a kvasinky bez ohledu na jejich morfologii. Růst většiny kmenů ze souboru se podařilo aplikací nanočástic zcela eliminovat, ve všech případech však byl tento růst alespoň velkou měrou potlačen. Velikosti inhibičních zón, které se dají považovat za efektivní (1), se ve většině případů pohybovaly v rozmezí 5–20 mm (79 %). Výsledky popisují významné antimikrobiální účinky na bázi nanotechnologií selenových nanočástic, které mohou být použity ke snížení rizika vzniku nekontrolovatelných infekcí. Issue of bacterial and yeast infections with complicated course of treatment due to resistance of agents of these diseases are a frequent topic of research in the field of human and veterinary medicine. In this study, we focused on the study of selenium nanoparticles effect in the form of a gel-like complex with a polymer substance Cekol (SeNPs) as possible alternative to antibiotic drugs, in the elimination of bacterial and yeast infections. Testing of complex was carried out on the bacterial and yeast isolates obtained from a cohort of 450 patients from the Department of Traumatology at Trauma Hospital in Brno, from which it was taken always one swab sample from a superficial skin infection and to date have managed to identify in total 74 sub-species of bacterial strains or yeasts. To identify strains and tests of SeNPs microbiological, molecular-biological and mass spectrometry methods were used. The study demonstrated a significant effect of this product on the bacterial strains and yeasts, regardless of their morphology. The growth of most strains from the file was managed by application of nanoparticles completely eliminated. However, in all cases this growth was at least largely suppressed. Sizes of inhibition zones, which can be regarded as effective (1), ranged from 5–20 mm. Our results describe significant antimicrobial effects based on nanotechnologies, which can be used to reduce the risk of uncontrolled infections in comparison with conventional antibiotic therapy. A high intracellular concentration of potassium (200-300 mmol/L) is essential for many yeast cell functions, such as the regulation of cell volume and pH, maintenance of membrane potential, and enzyme activation. Thus, cells use high-affinity specific transporters and expend a lot of energy to acquire the necessary amount of potassium from their environment. In Candida genomes, genes encoding 3 types of putative potassium uptake systems were identified: Trk uniporters, Hak symporters, and Acu ATPases. Tests of the tolerance and sensitivity of C. albicans, C. dubliniensis, C. glabrata, C. krusei, C. parapsilosis, and C. tropicalis to various concentrations of potassium showed significant differences among the species, and these differences were partly dependent on external pH. The species most tolerant to potassium-limiting conditions were C. albicans and C. krusei, while C. parapsilosis tolerated the highest KCl concentrations. Also, the morphology of cells changed with the amount of potassium available, with C. krusei and C. tropicalis being the most influenced. Taken together, our results confirm potassium uptake and accumulation as important factors for Candida cell growth and suggest that the sole (and thus probably indispensable) Trk1 potassium uptake system in C. krusei and C. glabrata may serve as a target for the development of new antifungal drugs.
Cíl práce: Druh Candida dubliniensis sdílí celou řadu fenotypových charakteristik s nejčastěji izolovanou kvasinkou z klinického materiálu, kterou je Candida albicans. To značně komplikuje jeho správnou identifikaci a záchyt v klinickém materiálu. Nedostatek informací o výskytu C. dubliniensis v České republice byl motivací ke zjištění jejího zastoupení ve vzorcích vyšetřených v Mikrobiologickém ústavu LF MU a FN u sv. Anny v Brně. Součástí práce je také posouzení spolehlivosti použitých kultivačních metod. Materiál a metodika: Celkem bylo vyšetřeno 2260 izolátů kvasinek určených původně jako C. albicans. Pro odlišení C. dubliniensis a C. albicans byly použity čtyři kultivační metody – hodnocení barvy kolonií na médiu CHROMagar Candida, kultivace na médiu s 6,5 % NaCl, růst při 42 °C a charakter kolonií na Staibově agaru. K verifikaci výsledků byla použita jednak latexová aglutinace pomocí soupravy Bichro-Dubli Fumouze, jednak druhově specifická polymerázová řetězová reakce (PCR). Výsledky: Z testovaného souboru bylo pomocí fenotypových metod, latexové aglutinace a PCR identifikováno 50 kmenů C. dubliniensis. To odpovídá 2,2 % z celkového počtu izolátů. Většina izolátů C. dubliniensis pocházela z dýchacích cest (31), zbytek byl získán z moči (3), stolice (4), z cévky (1) a jeden kmen byl zachycen z krve. S výjimkou hodnocení barvy kolonií na CHROMagaru Candida, u kterého byla specificita pouhých 85,5 %, vykazovaly všechny ostatní kultivační metody vysoké procento citlivosti a specificity. Závěr: Byl potvrzen výskyt C. dubliniensis mezi izoláty C. albicans v různých klinických materiálech, nejčastěji pocházely tyto kmeny z horních cest dýchacích. Rozlišení C. dubliniensis od C. albicans na základě barvy kolonií na chromogenním médiu CHROMagar Candida lze doporučit pouze jako screeningový test pro odlišení od ostatních druhů rodu Candida. Zbylé metody poskytují vysoké procento spolehlivosti. Konečná identifikace by měla raději vždy spočívat na použití kombinací těchto metod, případně lze výsledek verifikovat pomocí druhově specifické PCR nebo latexové aglutinace.
Study objective: The species Candida dubliniensis shares a wide range of phenotypic characteristics with Candida albicans, the most common yeast species isolated from clinical specimens. This is a considerable complication for the detection and identification of Candida dubliniensis from clinical specimens. The lack of data on the incidence of C. dubliniensis in the Czech Republic was the motivation behind the efforts to detect this pathogen in specimens analyzed at the Institute for Microbiology, Faculty of Medicine Masaryk University and St. Anne's Faculty Hospital in Brno. Another aim was to test the reliability of the culture methods used. Material and methods: Altogether 2260 yeast isolates initially identified as C. albicans were analysed. To differentiate C. dubliniensis from C. albicans, four phenotypic methods were used: colour-based differentiation on CHROMagar Candida medium, culture on medium with 6.5% of NaCl, growth at 42 °C, and colony characteristics on Staib agar. To verify the results, the Bichro-Dubli Fumouze latex agglutination test and species-specific polymerase chain reactions (PCR) were used. Results: Using phenotypic methods, latex agglutination, and PCR, 50 (2.2%) strains from the study set were assigned to C. dubliniensis. Most (31) C. dubliniensis isolates were recovered from the respiratory tract and the remaining others were three urine isolates, four stool isolates, one central venous catheter isolate, and one blood isolate. With the exception of colour-based differentiation on CHROMagar Candida medium showing a specificity of 85.5%, all the culture methods used have a high sensitivity and a high specificity. Conclusion: Identification of C. dubliniensis as C. albicans was confirmed in various clinical specimens, most often from the upper respiratory tract. The colour-based differentiation of C. dubliniensis from C. albicans on chromogenic CHROMagar Candida medium can only be recommended as a screening test for the differentiation of C. dubliniensis from other species of the genus Candida. The remaining three methods are highly reliable. The final identification should be based on a combination of these methods, with the species-specific PCR or latex agglutination test used for verification.

Cíl práce: Močové katétry kolonizuje celá řada mikroorganizmů, včetně kvasinek. Na kolonizaci se obvykle podílí více mikrobiálních druhů, které zde vytváří společenstvo - mnohodruhový biofilm. Tato kolonizace katétru většinou neovlivňuje významným způsobem klinický stav pacienta. Na druhou stranu biofilmové ložisko se může stát zdrojem endogenní infekce či může ovlivňovat funkčnost katétru a tvorbu močových kamenů. Materiál a metody: Celkem bylo zpracováno 721 močových katétrů. Mikroorganizmy byly z katétrů uvolněny sonikací a následně kultivovány. Identifikace byla provedena pomocí běžných fenotypových testů, dále pomocí hmotnostní spektrometrie MALDI-TOF a nejednoznačně určené kvasinky byly sekvenovány. Tvorba biofilmu byla prokazována kultivací v mikrotitrační destičce. Výsledky a závěr: Kolonizaci kvasinkami se podařilo prokázat u 244 (33,8 %) vyšetřených močových katétrů. Celkem však bylo izolováno 274 kmenů kvasinek, které se ve většině případů vyskytovaly na katétrech spolu s dalšími kvasinkami či jinými mikroorganizmy a tvořily zde vícedruhový biofilm. Nejčastěii byla izolována Candida albicans (n = 144), následovala Candida glabrata (n = 41), Candida tropicalis (n = 41) a Candida parapsilosis sensu stricto (n = 14). Dále byly zachyceny Candida kefyr (n = 10), Candida krusei (n = 9), Candida fabianii (n = 6), Candida lusitaniae (n = 5), Candida dubliniensis (n = 3) a v jednom případě Saccharomyces cerevisiae. Většina kvasinek byla schopna poměrně snadno vytvořit pevně lpící vrstvu biofilmu na umělých površích.
Objective: Urinary catheters are colonized by a wide range of microorganisms, including numerous yeasts. The catheters are usually colonized by more microbial species forming a community - multispecies biofilm. Catheter colonization usually does not affect the patient's clinical status in any significant way. On the other hand, the biofilm can become a source of endogenous infection and its presence can affect functionality of the catheter and formation of urinary stones. Material a Methods: A total of 721 urinary catheters were studied. Microorganisms were released from catheters by sonication and subsequently cultured. Their identification was performed with the use of common phenotypic tests, as well as using MALDI TOF. Yeasts whose identification was ambiguous were recognized by sequencing. Biofilm formation was assessed by growth in a microtiter plate. Results and Conclusion: Yeast colonization was proved in 244 urinary catheters. However, a total of 274 yeast strains were isolated. Most of them occurred together with other yeast species and/or bacteria on the catheters, producing multispecies biofilm there. The most frequent species was Candida albicans (a total of 144 isolated strains), followed by Candida glabrata (41), Candida tropicalis (41) and Candida parapsilosis sensu stricto (14). Other isolated species were as follows: Candida kefyr (10), Candida krusei (9), Candida fabianii (6), Candida lusitaniae (5), Candida dubliniensis (3) and Saccharomyces cerevisiae (one case). Most of the yeasts rather readily formed a firmly adhering biofilm layer on artificial surfaces.
