Úvod a cieľ práce: Exozómy, ktoré sú generované endozomálnou cestou, sa exocytózou uvoľňujú do extracelulárneho priestoru vrátane slín. Obsahujú nukleové kyseliny, proteíny a lipidy, ktoré transportujú do rôznych častí tela. Podieľajú sa na medzibunkovej komunikácii, či už podporujú alebo narúšajú rôzne fyziologické procesy. Cieľom práce bolo overiť efektívnosť ultracentrifugačnej metódy izolácie exozómov zo slín, izolovať a identifikovať proteíny v nich obsiahnuté, s dôrazom na exozomálne proteíny súvisiace s ochoreniami ústnej dutiny alebo systematickými ochoreniami s prejavmi v ústnej dutine. Metódy: Exozómy boli z plných nestimulovaných ľudských slín izolované opakovanými centrifugačnými krokmi s premývaním vo fosfátovom tlmivom roztoku s následnou metanolovo/chloroformovou precipitáciou proteínov. Proteíny boli identifikované bottom-up prístupom pomocou hmotnostnej spektrometrie s predradenou separáciou na kvapalinovom chromatografe. Výsledky: Identifikované proteíny boli klasifikované podľa proteínových tried, molekulových funkcií a biologických procesov. Najviac identifikovaných proteínov bolo z proteínových tried: cytoskeletálne proteíny, obranné/imunitné proteíny a štrukturálne proteíny, a proteínov zodpovedajúcich za katalytickú aktivitu a štrukturálnu molekulovú aktivitu. Závery: Proteíny boli klasifikované do skupín na základe ich molekulovej funkcie a biologických procesov, na ktorých sa v ľudskom organizme zúčastňujú. Boli identifikované niektoré proteíny/skupiny proteínov, ktoré môžu byť zaujímavé z hľadiska skúmania etiológie niektorých orálnych ochorení, napr. anexín A1, zymogén granulárny proteín 16 homológ B, mucín-5B, oblasti Ig lambda-3 reťazca C, oblasť Ig kapa reťazca C, oblasť Ig alfa-2 reťazca C a oblasť Ig alfa-1 reťazca C.
Introduction, aim: Exosomes generated by the endosomal pathway are released by the exocytosis into the extracellular space, including saliva. They contain nucleic acids, proteins and lipids that are transported to different parts of the body. They participate in intercellular communication, whether they support or disrupt various physiological processes. The aim of this work was to verify the efficiency of the ultracentrifugation method of isolation of exosomes from saliva, to exctract and identify proteins contained therein, with an emphasis on exosomal proteins related to diseases of the oral cavity or systemic diseases with manifestations in the oral cavity. Methods: Exosomes were isolated from full unstimulated human saliva by repeated centrifugation steps with washing in phosphate buffer followed by methanol/chloroform protein precipitation. Proteins were identified by a bottom-up approach using mass spectrometry with pre-separation by liquid chromatography. Results: The identified proteins were classified according to protein classes, molecular functions and biological processes. The most identified proteins were of the protein classes: cytoskeletal proteins, defense/immune proteins and structural proteins and proteins responsible for catalytic activity and structural molecular activity. Conclusions: Proteins have been classified into groups based on their molecular function and biological processes in which they participate in the human body. Some proteins/family of proteins have been identified that may be of interest in investigating the etiology of some oral diseases e.g. annexin A1, zymogen granular protein 16 homologue B, mucin-5B, Ig lambda-3 chain C region, Ig kappa chain C region, Ig alpha-2 chain C region and Ig alpha-1 chain C region.

Predmet oznámenia: Cieľom predkladanej práce je podať stručný prehľad o najnovších úspechoch pri štúdiu slinných proteínov proteomickými metódami pri parodontálnych ochoreniach. Tradičné diagnostické metódy založené na posúdení vizuálnych a rádiografických informácií o stave parodontu zlyhávajú pri identifikácii osôb ohrozených vznikom ochorenia a pri stanovení stupňa susceptibility k progresii ochorenia. V poslednom období došlo pri skúmaní telových tekutín, vrátane analýzy slín, k výraznému rastu použitia proteomických metód, ktoré sa stávajú doplnkovým nástrojom aj pri diagnostike ochorenia parodontu. Pri hľadaní potenciálnych biomarkerov sliny poskytujú rad výhod zahŕňajúcich dostupnosť vzorky, možnosť neinvazívneho odberu a komplexnosť obsahu proteínov. Je preto veľkou výzvou stanoviť kritériá pre diagnostiku ochorení parodontu, ktoré by boli založené na dôkazoch (evidence-based criteria). Tento prístup, ktorý využíva technológie na báze hmotnostnej spektrometrie, je schopný identifikovať a kvantifikovať veľké počty proteínov a ich izoforiem v jednej komplexnej vzorke orálnej tekutiny a extrahovať podrobné informácie o regulačných mechanizmoch hostiteľa na úrovni proteínu. Záver: Klinická diagnostika na báze analýzy orálnych tekutín, najmä tá, ktorá využíva proteomické metódy, umožňuje určiť vzťah medzi proteínami slín a daným ochorením. Preto je zámerom tohto článku snaha poskytnúť aktuálne informácie týkajúce sa štúdia slinných proteínov proteomickými metódami a ich vhodnosti použitia ako biomarkerov pre diagnostiku ochorení parodontu.
Background: The aim of the present work is to give a brief overview of the latest achievements in the study of salivary proteins by proteomic methods in periodontal diseases. Traditional diagnostic methods based on visual assessment and radiographic infomations about periodontal status fail to identify people at risk of developing the disease and in determining the degree of susceptibility to disease progression. Recently, there was when examining body fluids, including saliva analysis, a significant growth in the use of proteomic methods to become an additional tool in the diagnosis of periodontal disease. In the search for potential biomarkers saliva provides a number of advantages including the availability of samples, the possibility of non-invasive sampling and complexity of protein content. It is therefore a challenge to establish criteria for the diagnosis of periodontal diseases, which are based on evidence (evidence-based criteria). This approach, which uses technology based on mass spectrometry is able to identify and quantify large numbers of proteins and their isoforms in one comprehensive oral fluid samples and extract detailed information on the regulatory mechanisms of the host at the protein level. Conclusion: The clinical diagnosis based on the analysis of oral fluids, especially one that uses proteomic method allows to determine the relationship between salivary proteins and the condition. Therefore, the purpose of this article attempt to provide an update on the study of salivary proteins proteomic methods and their suitability for use as biomarkers for diagnosing periodontal diseases.

The agar working electrode as a hydrogel support for bilayer lipid membranes (BLM) was studied by electrochemical impedance spectroscopy. The aim was to study the influence of dc bias voltage on the formation of a stable supported bilayer lipid membrane (s-BLM) on the electrode tip. It was concluded that the hydrogel support is sufficiently stable in the applied range of bias voltages. The agar electrode provides satisfactory results in the frequency range 1–1?104 Hz. The electrode properties suggest applicability in deposition of s-BLM. Electric properties of s-BLM significantly depend on the dc bias voltage. The specific membrane capacitance of s-BLM at ?0.4 V was comparable with that of a conventional planar BLM. However, a shift to negative bias voltages led to a significant change of membrane resistance, possibly due to the increasing number of pores or defective structures in the lipid bilayer.
The electrical properties of the supported lipid bilayer membrane (s-BLM) of egg phosphatidylcholine (PC) self-assembled on agar surface were examined. To characterize the insulating properties of s-BLMs, electrochemical impedance spectroscopy (EIS) was used. The analysis of impedance spectra in terms of an equivalent circuit of agar/electrolyte and agar/s-BLM/electrolyte in the frequency range of 0.1 Hz-10 kHz was performed. The high-density lipoproteins (HDL)/s-BLM interaction in the concentration range from 20 microg/ml to 80 microg/ml of HDL was investigated. It is evident that treatment of s-BLM with HDL resulted in an increase of the lipid film resistance and a decrease of membrane capacitance.
