Many studies observed that increased consumption of cereal grains is associated with a reduced incidence of lifestyle diseases. Cereals are a rich source of functional compounds including dietary fibers, ?-glucans, resistant starches, vitamins, minerals and other bioactive phytochemicals, therefore they are suitable and increasingly used raw materials for production of functional foods.

Kurkumín (diferuloylmetan) je oranžovo-žlto sfarbená obsahová látka – sekundárny metabolit z rizomy (Curcuma longa L.), korenia (turmeric) často prítomného v prášku curry. Je to jeden z hlavných kurkuminoidov turmeric Curcuma longa L. Po stáročia bol kurkumín používaný v niektorých liečivých preparátoch, alebo ako korigens farby v potravinách. Ukázalo sa, že viaceré enzýmy, ktoré sú úzko spojené so zápalovým a nádorovým procesom, sú modulované kurkumínom. V tejto práci sú sumarizované výsledky inhibičného efektu kurkumínu a etanolického extraktu z Curcuma longa L. na aktivitu lipoxygenázy (LOX) z cytozolovej frakcie pľúc potkana. Stanovenie polohovej špecifity dioxygenácie kyseliny arachidónovej RP- and SP-HPLC metódou ukázalo, že v purifikovanom enzýmovom preparáte z cytozolovej frakcie pľúc potkana je prítomná špecifická forma lipoxygenázy preukazujúca 12/15-LOX duálnu špecifitu (s prevažujúcou 15-LOX aktivitou). Inhibičná aktivita kurkumínu a extraktu z Curcuma longa na aktivitu lipoxygenázy z cytozolovej frakcie pľúc potkana bola vyjadrená v percentách inhibície a hodnotách IC50. Vyjadrením kinetiky enzýmovej reakcie podľa Lineweaver-Burk za prítomnosti inhibítora sa zistilo, že kurkumín je kompetitívny inhibítor 12/15- LOX z cytozolovej frakcie pľúc potkana.
Curcumin (diferuloylmethane) is an orange-yellow secondary metabolic compound from the rhizome of turmeric (Curcuma longa L.), a spice often found in curry powder. It is one of the major curcuminoids of turmeric. For centuries, curcumin has been used in some medicinal preparations or as a food colouring agent. A variety of enzymes that are closely associated with inflammation and cancer were found to be modulated by curcumin. This paper summarized the results of the inhibitory effect of curcumin and a Curcuma longa L. ethanolic extract on lipoxygenase from the rat lung cytosolic fraction. The positional specificity determination of arachidonic acid dioxygenation by RP- and SP-HPLC methods showed that in a purified enzyme preparation from the rat lung cytosol the specific form of lipoxygenase (LOX) is present exhibiting 12/15-LOX dual specificity (with predominant 15-LOX activity). The inhibitory activity of curcumin and Curcuma longa extract on LOX from cytosolic fraction of rat lung was expressed in the percentage of inhibition and as IC50. Lineweaver-Burk plot analysis has indicated that curcumin is the competitive inhibitor of 12/15 LOX from the rat lung cytosolic fraction.

The article is focused on principal aspects of monitoring fermentation processes. Problems of sampling associated with the separation of biomass are explained mainly on the basis of various types of filtration and flow dialysis methods. The analytical flow methods FIA and SIA are briefly described with respect to their use in on-line monitoring. Some applications of biosensors in practical measurements are presented.
