BACKGROUND: The ventricular trabeculae play a role, among others, in the impulse spreading in ectothermic hearts. Despite the morphological similarity with the early developing hearts of endotherms, this trabecular function in mammalian and avian embryos was poorly addressed. RESULTS: We simulated impulse propagation inside the looping ventricle and revealed delayed apical activation in the heart with inhibited trabecular growth. This finding was corroborated by direct imaging of the endocardial surface showing early activation within the trabeculae implying preferential spreading of depolarization along with them. Targeting two crucial pathways of trabecular formation (Neuregulin/ErbB and Nkx2.5), we showed that trabecular development is also essential for proper conduction patterning. Persistence of the slow isotropic conduction likely contributed to the pumping failure in the trabeculae-deficient hearts. CONCLUSIONS: Our results showed the essential role of trabeculae in intraventricular impulse spreading and conduction patterning in the early endothermic heart. Lack of trabeculae leads to the failure of conduction parameters differentiation resulting in primitive ventricular activation with consequent impact on the cardiac pumping function.

Zvukové fenomény patří mezi čtyři nejčastější příznaky onemocnění temporomandibulárního kloubu. Rozdělují se na lupání a krepitace. Základním objektivním vyšetřením těchto fenoménů je auskultace kvalitním fonendoskopem, který by dnes měl být součástí standardního vybavení každé stomatologické i fyzioterapeutické ordinace.
Acoustic phenomena remain one out of four major signs of temporomandibular joint disorders. They are divided into clicking and crepitation. Basic examination method of these phenomena is an auscultation with high quality stethoscope, which should be a standard part of armamentarium in every dental or physiotherapeutic office.
Left ventricular noncompaction cardiomyopathy is associated with heart failure, arrhythmia, and sudden cardiac death. The developmental mechanism underpinning noncompaction in the adult heart is still not fully understood, with lack of trabeculae compaction, hypertrabeculation, and loss of proliferation cited as possible causes. To study this, we utilised a mouse model of aberrant Rho kinase (ROCK) signalling in cardiomyocytes, which led to a noncompaction phenotype during embryogenesis, and monitored how this progressed after birth and into adulthood. The cause of the early noncompaction at E15.5 was attributed to a decrease in proliferation in the developing ventricular wall. By E18.5, the phenotype became patchy, with regions of noncompaction interspersed with thick compacted areas of ventricular wall. To study how this altered myoarchitecture of the heart influenced impulse propagation in the developing and adult heart, we used histology with immunohistochemistry for gap junction protein expression, optical mapping, and electrocardiography. At the prenatal stages, a clear reduction in left ventricular wall thickness, accompanied by abnormal conduction of the ectopically paced beat in that area, was observed in mutant hearts. This correlated with increased expression of connexin-40 and connexin-43 in noncompacted trabeculae. In postnatal stages, left ventricular noncompaction was resolved, but the right ventricular wall remained structurally abnormal through to adulthood with cardiomyocyte hypertrophy and retention of myocardial crypts. Thus, this is a novel model of self-correcting embryonic hypertrabeculation cardiomyopathy, but it highlights that remodelling potential differs between the left and right ventricles. We conclude that disruption of ROCK signalling induces both morphological and electrophysiological changes that evolve over time, highlighting the link between myocyte proliferation and noncompaction phenotypes and electrophysiological differentiation.

Úvod a cíl: Autotransplantaci zubu můžeme v současné době pokládat za jeden z možných způsobů ošetření ztráty zubu. Tento způsob terapie se rozvíjí již více než 30 let a dostává se postupně do širšího povědomí zubních lékařů. Cílem této retrospektivní klinické studie bylo zpracovat a vyhodnotit výsledky zubní autotransplantace u dětí a dospělých pacientů s určením míry přežití autotransplantovaného zubu a míry úspěšnosti tohoto typu ošetření. Dalším cílem bylo určit specifika autotransplantací u jednotlivých věkových skupin se závěry důležitými pro klinickou praxi. Metodika: Zkoumaným souborem byli pacienti odeslaní konsekutivně k autotransplantaci zubů na specializované pracoviště v letech 2016–2021. Byla zaznamenána odbornost ošetřujícího lékaře, který pacienta k autotransplantaci indikoval, vhodnost a proveditelnost transplantace a donorová a příjmová oblast zubního transplantátu. Počátkem roku 2022 probíhalo klinické a rentgenologické hodnocení autotransplantovaných zubů. Pacienti byli rozděleni na dvě skupiny podle věku v době autotransplantace, a to do 18 let a více než 18 let. Výsledky: Celkově bylo k autotransplantaci delegováno 73 pacientů ve věkovém rozmezí 10–59 let. Věkový průměr byl 21,4 let s mediánem 17 let. U 12 pacientů nebyla autotransplantace doporučena. Celkově bylo provedeno 68 autotransplantací zubů, 12 jich však nesplňovalo minimální období sledování (šest měsíců po autotransplantaci). Celkově bylo tedy zhodnoceno 56 dentálních autotransplantátů. Ve skupině dětí se jednalo o 27 jedinců ve věku 10–17 let, u kterých byla provedena autotransplantace alespoň jednoho stálého zubu. Celkem bylo transplantováno 34 zubů. Vyhodnocení transplantovaných zubů proběhlo po 6 až 50 měsících od transplantace s průměrem 24 měsíců. Míra přežití dosahovala 100 %, míra úspěšnosti 91 %. Ve skupině dospělých se jednalo o 22 jedinců ve věku 18–59 let a celkem bylo transplantováno 22 zubů. Vyhodnocení transplantovaných zubů proběhlo po 6 až 72 měsících od transplantace s průměrem 33 měsíců. Míra přežití dosahovala 95 %, míra úspěšnosti 77 %. Závěr: Využití autotransplantátu k náhradě nezaloženého nebo ztraceného zubu nejčastěji indikovali ortodontisté. U pacientů ve věku do 18 let převažují jako donorová i příjmová oblast premoláry. U pacientů nad 18 let věku převažují jako donorová oblast třetí moláry a jako příjmová oblast moláry v dolní čelisti. U obou věkových skupin je vysoká míra přežití i úspěšnosti autotransplantátu, a proto lze tento postup považovat za spolehlivou metodu náhrady zubu.
Introduction, aim: At present, tooth autotransplantation is considered one of the therapeutic methods for the replacement of lost teeth. In the last 30 years, the method of tooth autotransplantation has been developed and refined and has become a basic knowledge of dental practitioners. The aim of this clinical retrospective study was to examine children and adult patients with tooth autotransplantation and obtain survival and success rates. Another aim was to determine other specifics of autotransplantation in each group with conclusions relevant for clinical practice. Methods: The study population consisted of the patients referred consecutively for tooth autotransplantation to a specialist department between years 2016 and 2021. The specialization of the reffering dentist, the suitability and feasibility of the transplantation, and the donor and recipient area of the tooth graft were recorded. In 2022, clinical and radiological evaluation of the autotransplanted teeth was performed. Patients were divided into two groups according to age in time of autotransplantation, namely under 18 years and over 18 years. Results: Overall, 73 patients in the age range of 10–59 years were referred for autotransplantation. The mean age was 21.43 years with a median age of 17 years. Autotransplantation was not recommended in 12 patients. A total of 68 autotransplantations were performed, but at the time of examination, 12 did not meet the minimum 6-month follow up, so they were excluded from the evaluation. A total of 56 autotransplants were evaluated. In the group of children, there were 27 patients aged 10–17 years who underwent autotransplantation of at least one permanent tooth. A total of 34 teeth were transplanted. The evaluation of the transplanted teeth was performed 6–50 months after the transplantation with a mean follow-up time of 24 months. The survival rate was 100% and the success rate was 91%. The adult group consisted of 22 patients aged 18–59 years and a total of 22 teeth were transplanted. The evaluation of the transplanted teeth was performed 6–72 months after transplantation with a mean of 33 months. The survival rate was 95%, the success rate 77%. Conclusion: The use of autograft to replace undeveloped or lost teeth was most often indicated by orthodontists. In the patients under 18 years of age, the premolars are the predominant donor area and recipient area. In patients over 18 years of age, the third molars are the predominant donor area and the mandibular molars the predominant recipient area. Both age groups have high survival and success rates of autograft and this procedure can be considered as a predictable method of tooth replacement.

That the highly trabeculated ventricular walls of the developing embryos transform to the arrangement during the fetal stages, when the mural architecture is dominated by the thickness of the compact myocardium, has been explained by the coalescence of trabeculations, often erroneously described as 'compaction'. Recent data, however, support differential rates of growth of the trabecular and compact layers as the major driver of change. Here, these processes were assessed quantitatively and visualized in standardized views. We used a larger dataset than has previously been available of mouse hearts, covering the period from embryonic day 10.5 to postnatal day 3, supported by images from human hearts. The volume of the trabecular layer increased throughout development, in contrast to what would be expected had there been 'compaction'. During the transition from embryonic to fetal life, the rapid growth of the compact layer diminished the proportion of trabeculations. Similarly, great expansion of the central cavity reduced the proportion of the total cavity made up of intertrabecular recesses. Illustrations of the hearts with the median value of left ventricular trabeculation confirm a pronounced growth of the compact wall, with prominence of the central cavity. This corresponds, in morphological terms, to a reduction in the extent of the trabecular layer. Similar observations were made in the human hearts. We conclude that it is a period of comparatively slow growth of the trabecular layer, rather than so-called compaction, that is the major determinant of the changing morphology of the ventricular walls of both mouse and human hearts.

PURPOSE: Osteoporosis is a severe health problem with social and economic impacts on society. The standard treatment consists of the systemic administration of drugs such as bisphosphonates, with alendronate (ALN) being one of the most common. Nevertheless, complications of systemic administration occur with this drug. Therefore, it is necessary to develop new strategies, such as local administration. METHODS: In this study, emulsion/dispersion scaffolds based on W/O emulsion of PCL and PF68 with ALN, containing hydroxyapatite (HA) nanoparticles as the dispersion phase were prepared using electrospinning. Scaffolds with different release kinetics were tested in vitro on the co-cultures of osteoblasts and osteoclast-like cells, isolated from adult osteoporotic and control rats. Cell viability, proliferation, ALP, TRAP and CA II activity were examined. A scaffold with a gradual release of ALN was tested in vivo in the bone defects of osteoporotic and control rats. RESULTS: The release kinetics were dependent on the scaffold composition and the used system of the poloxamers. The ALN was released from the scaffolds for more than 22 days. The behavior of cells cultured in vitro on scaffolds with different release kinetics was comparable. The difference was evident between cell co-cultures isolated from osteoporotic and control animals. The PCL/HA scaffold show slow degradation in vivo and residual scaffold limited new bone formation inside the defects. Nevertheless, the released ALN supported bone formation in the areas surrounding the residual scaffold. Interestingly, a positive effect of systemic administration of ALN was not proved. CONCLUSION: The prepared scaffolds enabled tunable control release of ALN. The effect of ALN was proved in vitro and in in vivo study supported peri-implant bone formation.

Bruxismus je vůbec nejčastější funkční poruchou orofaciálního systému a jako takový působí mimo rámec fyziologických funkčních aktivit stomatognátního systému, přičemž tento systém významným způsobem poškozuje. Jedná se o multifaktoriální onemocnění, kde můžeme jediný etiologický faktor identifikovat jen velmi zřídka, zpravidla se jedná o kombinaci predisponujících, vyvolávajících a udržovacích faktorů. Pro úspěšnou léčbu bruxismu je pochopení komplexní etiopatogeneze zcela klíčové. Vzhledem ke komplikované etiopatogenezi bruxismu je zásadní multioborová spolupráce jak v diagnostice, tak v léčbě, a to zejména mezi stomatologem a fyzioterapeutem, případně s účastí dalších odborníků.
Bruxism represents the most common functional disorder of the orofacial system and thus works out of the range of physiologic functional activities of the stomatognathic system, causing its damage. It is a multifactorial disorder where the possibility of identification of only one particular etiologic factor is rare; usually it represents a combination of predisposing, precipitating and perpetuating factors. Understanding of the complex etiopathogenesis is crucial for successful treatment of bruxism. Due to complicated etiopathogenesis, the interdisciplinary cooperation in diagnostics and treatment is crucial, mainly between dentist and physiotherapeutist, alternatively in cooperation with other specialists.

Considerable amount of research has been focused on dentin mineralization, odontoblast differentiation, and their application in dental tissue engineering. However, very little is known about the differential role of functionally and spatially distinct types of dental epithelium during odontoblast development. Here we show morphological and functional differences in dentin located in the crown and roots of mouse molar and analogous parts of continuously growing incisors. Using a reporter (DSPP-cerulean/DMP1-cherry) mouse strain and mice with ectopic enamel (Spry2+/- ;Spry4-/- ), we show that the different microstructure of dentin is initiated in the very beginning of dentin matrix production and is maintained throughout the whole duration of dentin growth. This phenomenon is regulated by the different inductive role of the adjacent epithelium. Thus, based on the type of interacting epithelium, we introduce more generalized terms for two distinct types of dentins: cementum versus enamel-facing dentin. In the odontoblasts, which produce enamel-facing dentin, we identified uniquely expressed genes (Dkk1, Wisp1, and Sall1) that were either absent or downregulated in odontoblasts, which form cementum-facing dentin. This suggests the potential role of Wnt signalling on the dentin structure patterning. Finally, we show the distribution of calcium and magnesium composition in the two developmentally different types of dentins by utilizing spatial element composition analysis (LIBS). Therefore, variations in dentin inner structure and element composition are the outcome of different developmental history initiated from the very beginning of tooth development. Taken together, our results elucidate the different effects of dental epithelium, during crown and root formation on adjacent odontoblasts and the possible role of Wnt signalling which together results in formation of dentin of different quality. © 2021 The Authors.
