Agrimonia eupatoria L. (AE) has a rich tradition of use in wound healing improvement across various cultures worldwide. In previous studies, we revealed that Agrimonia eupatoria L. water extract (AE) possesses a rich polyphenolic composition, displaying remarkable antioxidant properties. Our investigations also demonstrated that lipophosphonoxin (LPPO) exhibited antibacterial efficacy in vitro while preserving the proliferation and differentiation of fibroblasts and keratinocytes. Building upon our prior findings, in this study, we intended to examine whether a combination of AE and LPPO could enhance skin wound healing while retaining antibacterial attributes. The antibacterial activity of AE/LPPO against Staphylococcus aureus was evaluated, alongside its effects on fibroblast-to-myofibroblast transition, the formation of extracellular matrix (ECM), and endothelial cells and keratinocyte proliferation/phenotype. We also investigated AE/LPPO's impact on TGF-β1 and VEGF-A signaling in keratinocytes/fibroblasts and endothelial cells, respectively. Additionally, wound healing progression in rats was examined through macroscopic observation and histological analysis. Our results indicate that AE/LPPO promotes myofibroblast-like phenotypic changes and augments ECM deposition. Clinically relevant, the AE/LPPO did not disrupt TGF-β1 and VEGF-A signaling and accelerated wound closure in rats. Notably, while AE and LPPO individually exhibited antibacterial activity, their combination did not lead to synergism, rather decreasing antibacterial activity, warranting further examination. These findings underscore substantial wound healing improvement facilitated by AE/LPPO, requiring further exploration in animal models closer to human physiology.

Hlboká mozgová stimulácia (deep brain stimulation; DBS) je súčasťou pokročilej liečby vybraných neurologických ochorení, pri ktorých nie je dostatočne efektívna konzervatívna terapia. Etablovanými indikáciami sú Parkinsonova choroba, rôzne formy dystónie a esenciálny tras. V posledných rokoch sa rozšírilo indikačné spektrum DBS v rámci neurológie o vybrané formy epilepsie a v experimentálnej rovine prebiehajú klinické štúdie s využitím DBS pri chronickej klastrovej bolesti hlavy a ďalších extrapyramídových ochoreniach (Tourettov syndróm, Huntingtonova choroba). Ďalším posunom sú technologické inovácie v oblasti hardvéru. Prakticky všetky neurostimulátory sú kompatibilné s MR a životnosť batérie sa výrazne predĺžila (výrobca garantuje 13 rokov, ale experimentálne dáta predikujú životnosť pri nabíjateľných neurostimulátoroch až do 25 rokov). V rutinnej praxi sa začali používať tzv. smerovateľné elektródy, ktoré umožňujú presnejšie zacielenie elektrického poľa a minimalizáciu stimuláciou indukovaných nežiaducich účinkov. DBS je teda bezpečnou a efektívnou liečbou medicínsky refraktérnych neurologických ochorení a rýchlosť pokroku na tomto poli predpokladá ešte ďalšie zdokonalenie (automatizácia programovania, zmenšovanie a ďalšie predlžovanie životnosti batérie, jednoduchšia obslužnosť).
Deep brain stimulation (DBS) is an established advanced treatment option for selected neurological disorders with failed conservative therapy. Present indications of DBS are Parkinson‘s disease, various types of dystonia and essential tremor. In recent years, the DBS indication spectrum of neurological disorders has broadened with epilepsy and other more experimental indications such as chronic cluster headache and other movement disorders (Tourette’s syndrome, Huntington’s disease). Technological hardware innovations are another important step. Almost all currently manufactured neurostimulators are MRI compatible and the longevity of batteries has significantly improved (manufacturer guarantees a longevity of 13 years and the experimental data predict up to 25 years). Directional leads became a standard practice enabling the shaping of the electrical fields and minimalization of stimulation-induced side effects. Deep brain stimulation is a safe and effective therapy option for medically refractory neurological disorders and we may predict further substantial advances in this field (time-saving automatic programming, minimalization and longevity of batteries, user-friendly patient programmers).

Understanding the molecular and cellular processes in skin wound healing can pave the way for devising innovative concepts by turning the identified natural effectors into therapeutic tools. Based on the concept of broad‐scale engagement of members of the family of galactoside‐binding lectins (galectins) in pathophysiological processes, such as cancer or tissue repair/regeneration, the present study investigated the potential of galectins‐1 (Gal‐1) and ‐3 (Gal‐3) in wound healing. Human dermal fibroblasts, which are key cells involved in skin wound healing, responded to galectin exposure (Gal‐1 at 300 or Gal‐3 at 600 ng/ml) with selective changes in gene expression among a panel of 84 wound‐healing‐related genes, as well as remodeling of the extracellular matrix. In the case of Gal‐3, positive expression of Ki67 and cell number increased when using a decellularized matrix produced by Gal‐3‐treated fibroblasts as substrate for culture of interfollicular keratinocytes. In vivo wounds were topically treated with 20 ng/ml Gal‐1 or ‐3, and collagen score was found to be elevated in excisional wound repair in rats treated with Gal‐3. The tensile strength measured in incisions was significantly increased from 79.5±17.5 g/mm2 in controls to 103.1±21.4 g/mm2 after 21 days of healing. These data warrant further testing mixtures of galectins and other types of compounds, for example a combination of galectins and TGF‐β1.

Intracranial epidermoid cysts are slow growing congenital avascular neoplasms that spread across the basal surface of the brain. They most commonly occur in the paramedial region in the cerebellopontine angle and the parasellar region. Despite its generally benign nature, sporadically they can be accompanied with hemorrhage or very rarely undergo malignant transformation. The authors present a case report of a patient with a hemorrhagic vermian epidermoid cyst and a review of all published similar cases.

Úvod: Endometrióza je definovaná ako prítomnosť funkčného endometriálneho tkaniva, endometriálnych žliazok alebo endometriálnej strómy mimo maternicovú dutinu, čím je indukovaná chronická zápalová odpoveď. Endometrióza brušnej steny je pomerne raritné ochorenie s prevalenciou menej ako 1%. Najčastejšie je lokalizovaná v jazve po cisárskom reze. Hmatná rezistencia v jazve a zvýraznená bolestivosť lézie počas menštruácie sú hlavnými prejavmi ochorenia. Liečbou voľby je chirurgická resekcia s negatívnym resekčným okrajom. Kazuistika: Autori prezentujú dva prípady žien v reprodukčnom veku, ktoré boli vyšetrené pre bolestivosť v jazve po sekcii. Ako príčina ťažkostí bola už predoperačne suponovaná endometrióza, čo bolo definitívne potvrdené histologickým vyšetrením. Záver: U žien v reprodukčnom veku s rezistenciou v pooperačnej jazve je potrebné v rámci diferencionálnej diagnostiky myslieť na možný výskyt endometriózy brušnej steny.
Introduction: Endometriosis is defined as the presence of endometrial tissue, endometrial glands or endometrial stroma outside the uterine cavity causing chronic inflammatory response. The prevalence of abdominal wall endometriosis is less than 1%. Cesarean scar endometriosis is the most common type of abdominal wall endometriosis. Chronic lower abdominal pain amplified during menstruation and palpable mass in the area of scar are the main symptoms. Generally, surgical resection with negative resection margins offers the best chance for definitive treatment of abdominal wall endometriosis. Case report: The authors present two female patients in fertile age with chronic pain in the area of Cesarean scar. The preoperatively assumed endometriosis was histologically confirmed after complete surgical excision. Conclusion: Abdominal wall endometriosis is rare. However, it is a possible cause of constant lower abdominal pain, impacting quality of life of the patient.

Článok pojednáva o pomerne novom lipoglykopeptidovom antibiotiku na trhu s názvom dalbavancín. Stručne popisuje spektrum a mechanizmus jeho antibakteriálneho účinku a dávkovacie schémy, ktoré je možné použiť v liečbe infekcií kože a mäkkých tkanív. V predloženej kazuistike autori popisujú prípad flegmóny ramena u pacientky, ktorej vyvolávateľom bol MRSA a na jej liečbu bolo -úspešne použité toto antibiotikum.
The article discusses dalbavancin, a relatively new lipoglycopeptide antibiotic brought to market. It briefly describes the spectrum and mechanism of its antibacterial effect and dosing regimens that can be used in the treatment of skin and soft tissue infections. The authors present a case of a patient with shoulder phlegmon caused by MRSA who was successfully treated with this antibiotic.

OBJECTIVE: Hepatitis E infection is one of the most frequent acute hepatitis in the world. Currently five human genotypes with different geographical distributions and distinct epidemiologic patterns are identified. In Slovakia, only rare cases of hepatitis E have been reported in recent years. Therefore, the aim of the study was to evaluate the prevalence of anti-HEV total antibodies and the main risk factors for HEV in the general population in Eastern Slovakia. METHODS: Detection of anti-HEV total antibodies samples was done by a commercial enzyme-linked immunosorbent assay (ELISA) kit. RESULTS: Of 175 hospitalized patients included in the study, 76 (43.5%) showed positivity for anti-HEV total antibodies. No statistically significant differences were found in anti-HEV positivity between men and women or in the groups of different living areas (town/village - urban/rural). CONCLUSION: Prevalence of anti-HEV total antibodies of hospitalised patients was high. The risk factor significantly associated with antibody positivity was eating raw meat. Other factors, such as sex, age, living area and contact with animals were not associated with antibody positivity.
