Mycobacterium marinum patří mezi pomalu rostoucí, netuberkulózní (environmentální, atypická) mykobakterie se zoonotickým potenciálem. Vyskytuje se ve vodním prostředí a vyvolává onemocnění u ryb a jiných vodních živočichů označované jako mykobakteriózy. U lidí vyvolává primárně kožní onemocnění, které bývá nejčastěji lokalizováno na horních končetinách. Onemocnění se objevuje nejčastěji v souvislosti s akvarijním prostředím, a proto také bývá označováno jako „fish tank granuloma“. Léčba je, jako v případě všech mykobakteriálních onemocnění, komplikovaná a zdlouhavá. Pro definitivní určení patogenního agens by vždy mělo být provedeno vyšetření biologického materiálu laboratoří specializujících se na diagnostiku mykobakteriálních onemocnění. Zásadním bodem pro diagnostiku je správné odebrání vzorku a také posouzení anamnézy pacienta. Základními laboratorními metodami jsou kultivace a mikroskopie. Pro zařazení do druhu jsou nejčastěji využívány moderní biologické metody, jako hmotnostní spektrofotometrie (MALDI), polymerázová řetězová reakce (PCR), hybridizační sondy nebo sekvenování.
Mycobacterium marinum is a slowly growing non-tuberculous (environmental, atypical) mycobacterium with zoonotic potential. It occurs in the aquatic environment and causes diseases in fish and other aquatic animals known as mycobacterioses. In humans, it primarily causes skin infections, which are most commonly located in the upper limbs. The disease commonly appears in connection with the aquarium environment and is thus referred to as fish tank granuloma. As with all mycobacterial diseases, treatment is complicated and lengthy. For a definitive determination of the pathogen, biological materials should always be examined in a laboratory specializing in diagnosing mycobacteria. Critical for the right diagnosis is proper sample collection and assessment of the patient’s history. To detect mycobacteria, culture and microscopy are generally used. Species are identified using modern biological methods such as mass spectrometry (MALDI), polymerase chain reaction, hybridization probes or sequencing.

Ticks are obligate hematophagous arthropods of significant importance to human and veterinary medicine. They transmit a vast array of pathogens, including bacteria, viruses, protozoa, and helminths. Most epidemiological data on ticks and tick-borne pathogens (TBPs) in the West Indies are limited to common livestock pathogens such as Ehrlichia ruminantium, Babesia spp. (i.e., B. bovis and B. bigemina), and Anaplasma marginale, and less information is available on companion animal pathogens. Of note, human tick-borne diseases (TBDs) remain almost completely uncharacterized in the West Indies. Information on TBP presence in wildlife is also missing. Herein, we provide a comprehensive review of the ticks and TBPs affecting human and animal health in the Caribbean, and introduce the challenges associated with understanding TBD epidemiology and implementing successful TBD management in this region. In particular, we stress the need for innovative and versatile surveillance tools using high-throughput pathogen detection (e.g., high-throughput real-time microfluidic PCR). The use of such tools in large epidemiological surveys will likely improve TBD prevention and control programs in the Caribbean.

Snake fungal disease (SFD) is an emerging disease of conservation concern in eastern North America. Ophidiomyces ophiodiicola, the causative agent of SFD, has been isolated from over 30 species of wild snakes from six families in North America. Whilst O. ophiodiicola has been isolated from captive snakes outside North America, the pathogen has not been reported from wild snakes elsewhere. We screened 33 carcasses and 303 moulted skins from wild snakes collected from 2010-2016 in Great Britain and the Czech Republic for the presence of macroscopic skin lesions and O. ophiodiicola. The fungus was detected using real-time PCR in 26 (8.6%) specimens across the period of collection. Follow up culture and histopathologic analyses confirmed that both O. ophiodiicola and SFD occur in wild European snakes. Although skin lesions were mild in most cases, in some snakes they were severe and were considered likely to have contributed to mortality. Culture characterisations demonstrated that European isolates grew more slowly than those from the United States, and phylogenetic analyses indicated that isolates from European wild snakes reside in a clade distinct from the North American isolates examined. These genetic and phenotypic differences indicate that the European isolates represent novel strains of O. ophiodiicola. Further work is required to understand the individual and population level impact of this pathogen in Europe.

In this study, we focused on spreading of vancomycin-resistant enterococci (VRE) to the environment. We studied that weather crows in Canada may be carriers and potentially reservoirs of VRE with vanA gene. We have found one multi-resistant isolate of Enterococcus faecium sequence type (ST) 448 with vanA gene on Prince Edward Island. This study is the first report of VRE in Canadian wildlife.

The aim of this study was to determine the species distribution among 44 randomly selected clinical isolates (30 mecA-positive and 14 mecA-negative) of animal origin previously identified as Staphylococcus intermedius by phenotypic tests and species-specific PCR amplification of the 16S rRNA gene. For this purpose, we used a multiplex PCR for the detection of the nuc gene and restriction fragment length polymorphism analysis of pta gene amplified by PCR. Both methods allow discrimination of Staphylococcus pseudintermedius from the other closely related members of the S. intermedius group and other coagulase-positive staphylococci isolated from animals. Genetic diversity of S. pseudintermedius strains was analyzed by staphylococcal protein A-encoding gene (spa) typing. Multiplex PCR method was used to identify staphylococcal cassette chromosome mec (SCCmec) type in mecA-positive strains. All isolates previously identified as S. intermedius were shown to belong to S. pseudintermedius. According to PCR-based SCCmec typing, SCCmecIII was the most prevalent type (n = 23), and solely seven isolates were designated as non-typeable. Furthermore, the assessment of spa-typing results revealed that the majority of all strains (n = 27) harbored spa type t02, and 17 strains were classified as non-typeable.

Bartonely patří mezi méně známé původce rady lidských onemocněni. Jsou to gramnegativní bakterie pomalu rostoucí na heminem či bovinním sérem obohacených kultivačních médiích. Rod Bartonella, zahrnující v současné době více než 15 druhů, je celosvětově rozšířen. Bartonely kolují v přírodních ohniscích v závislosti na výskytu přirozeného hostitele (hlodavci, kočkovité a psovité šelmy, člověk) a hmyzího vektoru (blecha, klíště, veš). U rezervoárových živočichů zpravidla způsobují permanentní intraerytrocytární bakteriémii bez známek systémové zánětlivé aktivace. Klasickým příkladem lidského onemocnění je nemoc z kočičího škrábnutí (cat scratch dinase, CSD), způsobená Bartonella henselae a charakterizovaná regionální lymfangoitidou a lymfadenitidou. Do popředí zájmu se dostává schopnost bartonel (např. B. quintana) způsobovat, zejména u imunokompromitovaných osob, oportunní infekce, a to s rozmanitou klinickou manifestací: bacilární angiomatóza, specifická vaskulitida jater a sleziny (peliosis hepatis, splenis), endokarditida a další. Problematika infekcí způsobených bartonelami je relativně nová, a s přibývajícími poznatky v této oblasti její význam stále vzrůstá.
Bartonellae belong to less known causal agents of many human diseases. They are gramnegative bacteria growing slowly on culture media enriched with hemin or bovine serum. The genus Bartoneiia, which currently involves more than 15 species, is present worldwide. Bartonellae live in natural foci in dependence on the occurance of natural host (rodents, felines, canidae, human) and insect vector (flea, tick, louse). By reservoir animals they usually cause permanent intraerythrocytic bacteraemia without system inflammation symptoms. A classical example of a human disease is cat scratch disease (CSD) caused by Bartonella henselae and characterised by regional lymphagoitis and lymphadenitis. Increasing interest is being devoted to the ability of Bartonella sp. (e.i. B. quintana) to cause the opportune infections with diverse clinical manifestation: bacillary angiomatosis, specific liver and spleen vasculitis (pehosis hepatis, splenis), endocarditis and others. The issue of Bartonella infections is relatively new and its importance is still growing with increasing knowledge in this field.
