Spermatozoon represents a very special cell type in human body, and glycosylation plays essential roles in its whole life including spermatogenesis, maturation, capacitation, sperm-egg recognition, and fertilization. In this study, by mapping the most comprehensive N-glycoproteome of human spermatozoa using our recently developed site-specific glycoproteomic approaches, we show that spermatozoa contain a number of distinctive glycoproteins, which are mainly involved in spermatogenesis, acrosome reaction and sperm:oocyte membrane binding, and fertilization. Heavy fucosylation is observed on 14 glycoproteins mostly located at extracellular and cell surface regions in spermatozoa but not in other tissues. Sialylation and Lewis epitopes are enriched in the biological process of immune response in spermatozoa, while bisected core structures and LacdiNAc structures are highly expressed in acrosome. These data deepen our knowledge about glycosylation in spermatozoa and lay the foundation for functional study of glycosylation and glycan structures in male infertility.
Glycosylation is one of the most important post-translational modifications of proteins and plays an essential role in spermatogenesis, maturation, extracellular quality control, capacitation, sperm-egg recognition, and final fertilization. Spermatozoa are synthesized in the testes inactively with a thick glycocalyx and passed through the epididymis for further modification by glycosylation, deglycosylation, and integration to reach maturation. Subsequently, sperm capacitation and further fertilization require redistribution of glycoconjugates and dramatic glycocalyx modification of the spermatozoa surface. Furthermore, glycoproteins and glycans in seminal plasma are functional in maintaining spermatozoa structure and stability. Therefore, aberrant glycosylation may cause alteration of semen function and even infertility. Currently, mass spectrometry-based technologies have allowed large-scale profiling of glycans and glycoproteins in human semen. Quantitative analysis of semen glycosylation has also indicated many involved glycoproteome issues in male infertility and the potential biomarkers for diagnosis of male infertility in clinical. This review summarizes the role of glycosylation during spermatozoa development, the large-scale profiling of glycome and glycoproteome in human semen, as well as the association of aberrant glycosylation with infertility.

Avian brood parasites lay their eggs in other birds' nests, and hosts can mitigate the fitness cost of raising unrelated offspring by rejecting parasitic eggs. A visually-based cognitive mechanism often thought to be used by hosts to discriminate the foreign egg is to compare it against the hosts' own eggshell by size, shape, maculation, and/or ground coloration (i.e., absolute chromatic contrast). However, hosts may instead discriminate eggs based on their colors along a scale of natural avian eggshell coloration (i.e., directional chromatic contrast). In support of this latter visual process, recent research has found that directional chromatic contrasts can explain some host species' rejection behavior better than absolute chromatic or achromatic contrasts. Here, for the first time, we conducted an experiment in a cavity-nesting host species to test the predictions of these different visual mechanisms. We experimentally parasitized nests of the Common Redstart Phoenicurus phoenicurus, a regular host of a mimetic-egg laying Common Cuckoo Cuculus canorus host-race, using painted, immaculate 3D-printed model eggs in two geographically distant areas (Finland and Czech Republic). We found that directional chromatic contrasts better explained rejection behaviors in both parasitized (Finland) and non-parasitized (Czech Republic) host populations, as hosts rejected eggs that were noticeably browner, but not eggs that were noticeably bluer, than redstart eggs. These results support the paradigm of a single rejection threshold predicted by the directional chromatic contrast model and contribute to a growing generality of these patterns across diverse avian host-brood parasite systems.

The optimal acceptance threshold hypothesis provides a general predictive framework for testing behavioural responses to discrimination challenges. Decision-makers should respond to a stimulus when the perceived difference between that stimulus and a comparison template surpasses an acceptance threshold. We tested how individual components of a relevant recognition cue (experimental eggs) contributed to behavioural responses of chalk-browed mockingbirds, Mimus saturninus, a frequent host of the parasitic shiny cowbird, Molothrus bonariensis. To do this, we recorded responses to eggs that varied with respect to two components: colour, ranging from bluer to browner than the hosts' own eggs, and spotting, either spotted like their own or unspotted. Although tests of this hypothesis typically assume that decisions are based on perceived colour dissimilarity between own and foreign eggs, we found that decisions were biased toward rejecting browner eggs. However, as predicted, hosts tolerated spotted eggs more than unspotted eggs, irrespective of colour. These results uncover how a single component of a multicomponent cue can shift a host's discrimination threshold and illustrate how the optimal acceptance threshold hypothesis can be used as a framework to quantify the direction and amount of the shift (in avian perceptual units) of the response curve across relevant phenotypic ranges. Brood parasites lay their eggs in other females' nests, leaving the host parents to hatch and rear their young. Studying how brood parasites manipulate hosts into raising their young and how hosts detect parasitism provide important insights in the field of coevolutionary biology. Brood parasites, such as cuckoos and cowbirds, gain an evolutionary advantage because they do not have to pay the costs of rearing their own young. However, these costs select for host defenses against all developmental stages of parasites, including eggs, their young, and adults. Egg rejection experiments are the most common method used to study host defenses. During these experiments, a researcher places an experimental egg in a host nest and monitors how hosts respond. Color is often manipulated, and the expectation is that the likelihood of egg discrimination and the degree of dissimilarity between the host and experimental egg are positively related. This paper serves as a guide for conducting egg rejection experiments from describing methods for creating consistent egg colors to analyzing the findings of such experiments. Special attention is given to a new method involving uniquely colored eggs along color gradients that has the potential to explore color biases in host recognition. Without standardization, it is not possible to compare findings between studies in a meaningful way; a standard protocol within this field will allow for increasingly accurate and comparable results for further experiments.

Obligate avian brood parasitic species impose the costs of incubating foreign eggs and raising young upon their unrelated hosts. The most common host defence is the rejection of parasitic eggs from the nest. Both egg colours and spot patterns influence egg rejection decisions in many host species, yet no studies have explicitly examined the role of variation in spot coloration. We studied the American robin Turdus migratorius, a blue-green unspotted egg-laying host of the brown-headed cowbird Molothrus ater, a brood parasite that lays non-mimetic spotted eggs. We examined host responses to model eggs with variable spot coloration against a constant robin-mimetic ground colour to identify patterns of rejection associated with perceived contrast between spot and ground colours. By using avian visual modelling, we found that robins were more likely to reject eggs whose spots had greater chromatic (hue) but not achromatic (brightness) contrast. Therefore, egg rejection decision rules in the American robin may depend on the colour contrast between parasite eggshell spot and host ground coloration. Our study also suggests that egg recognition in relation to spot coloration, like ground colour recognition, is tuned to the natural variation of avian eggshell spot colours but not to unnatural spot colours.

Polystomatid monogeneans have a wide diversity of life cycles correlated with the varied ecology and behaviour of their aquatic vertebrate hosts. Typically, transmission involves a swimming infective larva but most hosts are amphibious and invasion is interrupted when hosts leave water. A key life cycle adaptation involves a uterus that, in the most specialised cases, may contain several hundred fully-developed larvae prepared for instant host-to-host transmission. By contrast, one subfamily of the Polystomatidae - the Polystomoidinae, specific to chelonians (freshwater turtles) - has a simplified reproductive system without a uterus. Recently, Polystomoides nelsoni Du Preez et Van Rooyen, 2015 has been described with a uterus containing multiple eggs. The present study explores the exceptional interest of this parasite - for the functional biology of egg production, for the evolution of a reproductive system unique amongst ca 60 species in the subfamily, and for systematic relationships. A new genus is proposed, Uteropolystomoides gen. n., separate from the four currently-recognised genera Polystomoides Ward, 1917, Uropolystomoides Tinsley et Tinsley, 2016, Neopolystoma Price, 1939 and Polystomoidella Price, 1939 which lack a uterus. In addition, U. nelsoni (Du Preez et Van Rooyen, 2015) comb. n. has a suite of distinctive copulatory stuctures: a massive genital bulb with an exceptionally large number of very long genital spines and hyper-development of the vaginal openings. These characters set U. nelsoni apart from all other polystomoidines worldwide except Polystomoides multifalx Stunkard, 1924 and P. stunkardi Harwood, 1931. Missing data for these latter species preclude definitive assessment of inter-relationships but the distinguishing characters of U. nelsoni, especially the unique occurrence of the uterus, suggest a novel evolutionary pathway isolated from other lineages of polystomatids infecting chelonians.

Accurate recognition of salient cues is critical for adaptive responses, but the underlying sensory and cognitive processes are often poorly understood. For example, hosts of avian brood parasites have long been assumed to reject foreign eggs from their nests based on the total degree of dissimilarity in colour to their own eggs, regardless of the foreign eggs' colours. We tested hosts' responses to gradients of natural (blue-green to brown) and artificial (green to purple) egg colours, and demonstrate that hosts base rejection decisions on both the direction and degree of colour dissimilarity along the natural, but not artificial, gradient of egg colours. Hosts rejected brown eggs and accepted blue-green eggs along the natural egg colour gradient, irrespective of the total perceived dissimilarity from their own egg's colour. By contrast, their responses did not vary along the artificial colour gradient. Our results demonstrate that egg recognition is specifically tuned to the natural gradient of avian eggshell colour and suggest a novel decision rule. These results highlight the importance of considering sensory reception and decision rules when studying perception, and illustrate that our understanding of recognition processes benefits from examining natural variation in phenotypes.
Pri závislosti od alkoholu môžu vznikať rôzne zdravotné problémy v dôsledku postihnutia rôznych orgánových systémov. V aktuálnom príspevku približujeme pelagru ako možnú komplikáciu nadužívania alkoholu. Pelagra je ochorenie spôsobené nedostatkom vitamínu B3 (niacínu), alebo jeho prekurzora tryptofánu. Pri závislosti od alkoholu je vznik pelagry podmienený zníženým príjmom niacínu alebo tryptofánu, malabsorbciou, hnačkami a hepatopatiou. Klasickou triádou klinických príznakov pelagry je syndróm „3D“, ktorý zahŕňa „dermatitídu“ (kožné príznaky), „diarrheu“ (gastrointestinálne príznaky) a „demenciu“ (neuropsychiatrické príznaky); k týmto príznakom sa pridáva aj „death“ (smrť) a následne sa hovorí o syndróme „4D“. Diagnostika pelagry je založená na rozpoznaní vyššie uvedených príznakov a ich správnom zhodnotení v diferenciálne-diagnostickej úvahe. Podpornými informáciami sú (v praxi psychiatra) (hetero-)anamnestické údaje o nadužívaní alkoholu. Dôležitým potvrdením diagnózy pelagry je pozitívna terapeutická odpoveď pacienta s typickými prezentovanými ťažkosťami na podanie vitamínu B3. Bez včasnej a správnej liečby zomierajú pacienti s pelagrou v priebehu niekoľkých rokov. Liečba spočíva v dlhodobom dopĺňaní niacínu a liečbe základného ochorenia, ktoré jeho deficit spôsobuje (vrátane odvykacej liečby závislosti od alkoholu). Vhodné je dopĺňať nielen vitamín B3, ale celé spektrum vitamínov zo skupiny B, a tiež horčík a zinok. V prevencii je dôležitá strava obsahujúca potraviny ako droždie, mäso, ryby, mlieko, vajcia, zelená zelenina, strukoviny a obilniny.
Various health issues may arise within the course of alcohol addiction. Here we describe pellagra as possible complication of alcohol use disorder (AUD). Pellagra is a disease caused by deficiency of vitamin B3 (niacin) or its precursor tryptophan. There are several factors contributing to exacerbation of pellagra within the course of AUD: reduced intake of niacin or tryptophan, malabsorption, diarrhea, and liver insufficiency. The classic syndrome of “3D” in pellagra consists of “dermatitis” (dermatological symptoms), “diarrhea” (gastrointestinal symptoms) and “dementia” (neuropsychiatric symptoms); “death” can be added to these symptoms, creating the syndrome of “4D”. Diagnosis of pellagra is based on the recognition of the symptoms mentioned above, while history of chronic alcohol use can be informative, too. The diagnosis can be confirmed by positive therapeutic response to niacin in a patient with typical symptoms. If untreated, pellagra leads to death in the horizon of several years. The treatment consists of long-term supplementation of niacin and management of the underlying condition that causes its deficit (including the long-term therapy of AUD). It is advisable to supplement the entire spectrum of B-vitamins as well as magnesium and zinc. A diet consisting of meals prepared from ingredients such as yeast, meat, fish, milk, eggs, green vegetables, legumes and cereals is important in the prevention of pellagra.

Three new psilostomid genera, Byrdtrema n. g., Longisaccus n. g. and Macracetabulum n. g., each with a single species, are described from ducks, Aix sponsa (L.) and Bucephala albeola (L.) in North America. Byrdtrema n. g. and Macracetabulum n. g. possess a bipartite seminal vesicle and share this character with four psilostomid genera, Grysoma Byrd, Bogitsh & Maples, 1961, Neopsilotrema Kudlai, Pulis, Kostadinova & Tkach, 2016, Psilostomum Looss, 1899 and Psilotornus Byrd & Prestwood, 1969. Byrdtrema n. g. differs from Macracetabulum n. g. in the shape of the body (elongate vs elongate-oval); the position of the ventral sucker (in first third of body vs just pre-equatorial); the shorter forebody; as well as in the smaller size of the eggs in relation to body length. Both new genera differ from (i) Grysoma by the nature of the vitellarium (large, compact follicles with small vitelline cells vs weakly defined follicles with large vitelline cells, respectively) and the smaller size of the eggs in relation to body length; (ii) Psilostomum in the posterior extend of the cirrus-sac in relation to ventral sucker (slightly posterior vs more posterior), the location of the genital pore (at the level of oesophagus vs just postbifurcal), the shorter length of uterine and longer post-testicular fields in relation to body length, and the anterior limits of vitellarium (at the level of ventral sucker vs posterior to ventral sucker); (iii) Psilotornus by the presence of a muscular pharynx (vs absent or rudimentary) and the location of the cirrus-sac (antero-dorsal to ventral sucker or more posterior vs entirely anterior to ventral sucker) and ovary (in hindbody vs in forebody). Byrdtrema n. g. differs from Neopsilotrema in the shape of the body (elongate vs subspherical to elongate-oval) and ventral sucker (elongate-oval vs subspherical to transversely oval), the shorter forebody and smaller eggs in relation to body length. Macracetabulum n. g. differs from Neopsilotrema by the shape of the ventral sucker (elongate-oval vs subspherical to transversely oval), the anterior limits of vitellarium (level of middle of ventral sucker vs level of intestinal bifurcation or anterior testis); and the slightly smaller size of eggs in relation to body length. Among the psilostomid genera, Longisaccus n. g. shows close affinities to Psilochasmus Lühe, 1909 in the presence of the long cirrus-sac and tubular internal seminal vesicle but can be clearly distinguished from the latter by the absence of the retractile tail-like process. In combination with molecular data, the above differences justify the recognition of three new genera. A key to the genera of the Psilostomidae is provided.
