Sox3/SOX3 gene is considered to be one of the earliest neural markers in vertebrates. Despite the mounting evidence that Sox3/SOX3 is one of the key players in the development of the nervous system, limited data are available regarding the transcriptional regulation of its expression. This review is focused on the retinoic acid induced regulation of SOX3 gene expression, with particular emphasis on the involvement of retinoid receptors. Experiments with human embryonal carcinoma cells identified two response elements involved in retinoic acid/retinoid X receptor-dependent activation of the SOX3 gene expression: distal atypical retinoic acid-response element, consisting of two unique G-rich boxes separated by 49 bp, and proximal element comprising DR-3-like motif, composed of two imperfect hexameric half-sites. Importantly, the retinoic acid-induced SOX3 gene expression could be significantly down-regulated by a synthetic antagonist of retinoid receptors. This cell model provides a solid base for further studies on mechanism(s) underlying regulation of expression of SOX3 gene, which could improve the understanding of molecular signals that induce neurogenesis in the stem/progenitor cells both during development and in adulthood.

Project is aimed at analysis of molecular cytogenetic markers with prognostic value in selected childhood tumors of embryonal origin (medulloblastoma/PNET, neuroblastoma, retinoblastoma, and other extracranial solid tumors). Bioptic material will be examined using CGH, HR-CGH and FISH techniques; tumor tissue will be also cultivated for metaphase analysis using SKY technique and G-banding. Histological type of the cell culture will be assessed by immunocytochemical methods. Relevance of molecular cytogenetic techniques (CGH, HR-CGH, SKY) for detection of specific aberrations and correlation between detected cytogenetic markers and clinical course of the disease will be evaluated. The final outcome of the project will be the assessment of the importanceof cumulative cytogenetic changes for both subclasification of patients and prediction of the course of disease.
Projekt je zaměřen na analýzu molekulárně cytogenetických markerů s prognostickým významem u vybraných embryonálních nádorů dětského věku (medulloblastom/PNET, neuroblastom, retinoblastom a další extrakraniální solidní nádory). Materiál z biopsií bude přímo vyšetřován metodami CGH, HR-CGH a FISH; nádorová tkáň bude rovněž kultivována v podmínkách in vitro za účelem vyšetření metafází metodami G-banding a SKY. Ověření typu kultivovaných buněk bude prováděno imunocytochemicky. Dalším cílem projektu je posoudit přínos zvolených technik (CGH, HR-CGH, SKY) pro zvýšení záchytu specifických aberací a zhodnotit korelaci mezi detekovanými molekulárně cytogenetickými markery a klinickým průběhem onemocnění. Finálním výstupem projektu bude zhodnocení významu kumulovaných cytogenetických změn pro subklasifikaci nemocných a predikci průběhu onemocnění.

The aim of our study was to characterize mouse embryonal carcinoma (EC) cells P19 in different stages of retinoic acid induced neurodifferentiation by two methods, immunocytochemistry and RT qPCR. The characterization of the cells is crucial before any transplantation into any model, e.g. in our case into the mouse brain with the aim to treat a neurodegenerative disease. Specific protein markers (MAP-2, OCT-4, FORSE-1) were detected by immunocytochemistry in the cell cultures. The mRNA expression levels of PAX-6, MASH-1, Brachyury, GATA-4 and AFP were determined by RT qPCR method. HPRT was used as a housekeeping gene. The degree of differentiation can be characterized by expression of analyzed genes. The presence of OCT-4 and FORSE-1 proteins in undifferentiated pluripotent cells and the presence of dendrite specific MAP-2 in neuroprogenitors was detected. The expression levels of PAX-6 and MASH-1 increased and expression of Brachyury decreased during the neurodifferentiation process. The expression levels of GATA-4 and AFP were the highest after induction of differentiation with retinoic acid. Detailed characterization of cells before transplantation experiments can contribute to better understanding of their effect.

OBJECTIVES: Intracellularly generated reactive oxygen species (ROS) are thought to modulate redox sensitive signaling pathways and thus regulate cell physiology including proliferation and differentiation. However, the role of ROS in neuronal differentiation of embryonic pluripotent cells is unknown. For this reason, the modification of retinoic acid (RA) induced neuronal differentiation of mouse embryonal carcinoma cells P19 by selected ROS scavengers and flavoprotein inhibitor was evaluated. METHODS: Intracellular ROS was evaluated by flowcytometry. Cellular redox status was evaluated based on total levels of reduced thiol groups in cells. The activity of the RA responsive element (RARE) was evaluated by luciferase reporter assay. The RA-induced neuronal differentiation was determined based on changes in the expression of protein markers characteristic for undifferentiated (Oct-4) and neuron-like cell differentiated cells (N-cadherin and III-beta tubulin). RESULTS: RA increased the intracellular ROS production that was accompanied by a decrease in thiol groups in cells. The ROS scavengers and flavoprotein inhibitor reduced RA-induced ROS production, RA-induced activity of RARE, and it decreased the RA-induced expression of N-cadherin and III-beta tubulin. CONCLUSIONS: Our data outline a role of ROS as important molecules in the transduction of an intracellular signal during the neuronal differentiation of ES cells
