The antimicrobial effect of OKCEL® H-D, a topical, absorbable hemostatic textile prepared from oxidized cellulose, was tested. Testing by dilution and diffusion methods was conducted on a spectrum of 27 select microorganisms, including also antibiotic-resistant strains. OKCEL® H-D showed inhibitory effects on nearly all tested bacteria. In testing using the dilution suspension method, the majority of bacteria showed decrease in cell density by 7-8 orders of magnitude after just 6 h of exposure. For clinical isolates of antibiotic-resistant strains, a reduction occurred after 24 h of exposure. In testing the antimicrobial effects of OKCEL® H-D by the dilution method was least effective on spore-forming Bacillus subtilis, for which no antimicrobial effect was detected after 48 h, and on Mycobacterium smegmatis, for which the number of cells decreased by four orders of magnitude only after 24 h. By the diffusion method, inhibition zones were recorded for nearly all test microorganisms except for Staphylococcus aureus, M. smegmatis, and Listeria monocytogenes. No growth beneath the tested OKCEL® H-D material was recorded, however, even for the latter-named bacteria strains, which attests to its good inhibitory effect.

Novel hydrolytically stable gelatin nanofibers modified with sodium or calcium salt of oxycellulose were prepared by electrospinning method. The unique inhibitory effect of these nanofibers against Escherichia coli bacteria was examined by luminometric method. Biocompatibility of these gelatin/oxycellulose nanofibers with eukaryotic cells was tested using human lung adenocarcinoma cell line NCI-H441. Cells firmly adhered to nanofiber surface, as determined by scanning electron microscopy, and no signs of cell dying were detected by fluorescent live/dead assay. We propose that the newly developed gelatin/oxycellulose nanofibers could be used as promising scaffold for lung disease modeling and anti-cancer drug testing.

Celulózové materiály nebo jejich deriváty nabízejí pro hojení ran řadu cenných vlastností. Karboxylované celulózy zastupuje pře-devším karboxymethylovaná celulóza a oxidovaná celulóza. Hcel HT (karboxymethylovaná celulóza) je nový typ celulózového krytí s kyselým povrchovým pH.
V oblasti technologie léků nachází rozsáhlé uplatnění gelové systémy vysokomolekulárních látek, které mají řadu výhod, jako jsou nízká toxicita, dostupnost, ojedinělé fyzikální vlastnosti, biokompatibilita, mukoadhezivita a další. Gelové systémy se používají v oblasti lokální i celkové terapie, v tvarově specifických i tvarově nespecifických lékových formách, v lécích první, druhé i třetí generace. Významnou skupinu gelů, které se používají v oblasti farmacie, tvoří gely hydrofilní, neboli hydrogely, tvořené nejčastěji hydrofilními polymery přírodního, polosyntetického a syntetického původu. Přestože celulosové deriváty jako představitelé polymerů polosyntetického původu se uplatňují ve farmaceutické technologii po dlouhou dobu, v jejich výzkumu se neustává a hledají se další možnosti jejich uplatnění. Mezi jejich přednosti patří zejména bezpečnost, snadná dostupnost a relativně nízká cena. Přehledový článek popisuje vybrané celulosové deriváty, jejich vlastnosti a využití ve farmaceutické technologii se zaměřením na uplatnění v oblasti tvorby gelových systémů.
The field of drug technology widely ulilizes gel systems of high-molecular substances, which have a number of advantages, such as low toxicity, availability, unique physical properties, biocompatibility, mucoadhesivity, and others. Gel systems are used in the field of local as well as general therapy, in both shape-specific and shape-non-specific dosage forms, in medicaments of the first, second, and third generations. An important group of gels employed in pharmacy are hydrophilic gels or hydrogels, most frequently composed of hydrophilic polymers of natural, semisynthetic and synthetic origin. Though cellulose derivatives as the representatives of polymers of semisynthetic origin are used in pharmaceutical technology for a long time, their research continues and their other possible uses are being searched for. Their advantages include especially safety, easy availability, and a relatively low price. The review paper describes selected cellulose derivatives, their properties and uses in pharmaceutical technology with regard to their use in the field of production of gel systems.

Oxidovaná celulosa patří mezi netoxické a biokompatibilní biopolymery. Oxidovaná regenerovaná celulosa (ORC) se vyrábí z regenerované celulosy původem z dřevní buničiny, obsahující okolo 50 % celulosy. Pro dosažení čištěné celulosy je potřeba ji chemicky rozložit a následně spojit v „regenerovanou“ celulosu. Díky svým dobrým hemostatickým účinkům, vysoké biosolubilitě a biodegradabilitě, antioxidačním a rány-hojícím vlastnostem představuje oxidovaná celulosa vhodný prostředek při terapii krvácivých stavů v různých oblastech medicíny. Navíc, potvrzené baktericidní účinky oxidované celulosy vůči širokému spektru aerobních i anaerobních patogenů zvyšují terapeutický profil této látky pro použití v klinické praxi. V současnosti dochází k novému zájmu o její širší využití v klinické praxi a k prohlubování znalostí o mechanismech účinků, které jsou testovány in vitro, na zvířecích modelech i v klinických studiích s humánními subjekty hodnocení. Tato práce si klade za cíl shrnout dosavadní poznatky o hemostatických vlastnostech oxidované celulosy a charakterizovat také její další biologické účinky
Oxidized cellulose ranks among nontoxic and biocompatible biopolymers. Oxidized regenerated cellulose (ORC) is manufactured from regenerated cellulose derived from wood pulp containing about 50% of cellulose. To obtain purified cellulose, it is necessary to decompose it in a chemical way and subsequently put it together to make “regenerated” cellulose. Thanks to its good hemostatic effects, high biosolubility and biodegradability, antioxidant and wound-healing properties, oxidized cellulose represents a suitable means for the therapy of bleeding conditions in various fields of medicine. In addition, the confirmed bactericidal effects of oxidized cellulose towards a wide spectrum of aerobic and anaerobic pathogens increase the therapeutic potential of this agent for use in clinical practice. At present there is a renewed interest in its wider use in clinical practice and in an improvement of the knowledge of its mechanisms of effects, which are tested in vitro, on animal models as well as in clinical studies. The present paper attempts to summarize the hitherto knowledge of hemostatic properties of oxidized cellulose and also to characterize other possible biological effects.
Antimicrobial properties of oxidized cellulose and its salts in linters (-L) and microsphere (-M) form (OKCEL H-L, OKCEL Zn-M, OKCEL ZnNa-L, OKCEL ZnNa-M and OKCEL Ag-L) were tested by a dilution method against a spectrum of microbial strains: Escherichia coli, Pseudomonas aeruginosa, Staphylococcus epidermidis, Bacillus licheniformis, Aspergillus niger, Penicillium chrysogenum, Rhizopus oryzae, Scopulariopsis brevicaulis, Candida albicans and Candida tropicalis. OKCEL Ag-L exhibited antimicrobial activity in the range 0.1-3.5% w/v against all the bacteria and fungi involved in this study. Strong inhibition by OKCEL ZnNa-M was observed for Staphylococcus epidermidis, Bacillus licheniformis, Rhizopus oryzae, Candida albicans and Candida tropicalis in the range 0.5-2.0% w/v. Antimicrobial effects of oxidized cellulose and its salts in textile form were investigated by a diffusion and dilution method against the spectrum of above-cited microbial strains extended by Clostridium perfringens. Generally, OKCEL Ag-T, OKCEL Zn-T and OKCEL H-T showed high antimicrobial activity against populations of Pseudomonas aeruginosa, Bacillus licheniformis and Staphylococcus epidermidis. OKCEL Zn-T was the only sample suppressing the growth of species.
Přehledový článek je zaměřen na další z řady farmaceuticky a medicínsky využívaných celulosových derivátů – oxycelulosu. Tato látka připravená oxidací celulosy se v praxi na rozdíl od celulosových esterů a etherů neuplatňuje dosud tak široce, ale nové studie odhalují její potenciál pro využití v moderních terapeutických systémech ať už jako pomocné látky modifikující uvolňování léčiva, nosiče léčiva, nebo i jako účinné látky se schopností zastavovat krvácení, zabraňovat srůstům tkání po chirurgických výkonech a v neposlední řadě jako látky s protimikrobní a imunomodulační aktivitou, jejíž unikátní vlastnost biodegradace, podpořená hypoalergenitou a nedráždivostí, nevykazuje žádný z dosud rozsáhle farmaceuticky uplatňovaných celulosových derivátů. Článek podává základní přehled o historii používání tohoto derivátu, jeho fyzikálně-chemických vlastnostech, přes popis schopnosti biodegradace, až po již realizované a plánované využití.
The review article is focused on one of the wide range of pharmaceutically and medicinally employed cellulose derivates – oxycellulose. This substance, prepared by oxidation of cellulose, in contrast to cellulose esters and ethers, has not been not employed in practice in a wide extent yet. However, recent studies reveal its potential use in modern therapeutical systems, whether as a release-modifying excipient, a drug carrier, or also an active substance with ability of hemostasis, prevention of tissue adhesion after surgical operations, and a substance with antimicrobial and immunomodulating activity, whose unique property of biodegradability, promoted by hypoallergenicity and non-irritability, has not been observed in any of the hitherto used cellulose derivates. The article presents a comprehensive overview about the history of this derivate, from its physico-chemical properties through the description of the ability of biodegradability to its already realized and planned utilization.
