The aim of the present study was to investigate the suitability of insoluble Eudragit® water dispersions (NE, NM, RL, and RS) for direct high-shear granulation of very soluble levetiracetam in order to decrease its burst effect from HPMC K100M matrices. The process characteristics, ss-NMR analysis, in vitro dissolution behavior, drug release mechanism and kinetics, texture profile analysis of the gel layer, and PCA analysis were explored. An application of water dispersions directly on levetiracetam was feasible only in a multistep process. All prepared formulations exhibited a 12-hour sustained release profile characterized by a reduced burst effect in a concentration-dependent manner. No effect on swelling extent of HPMC K100M was observed in the presence of Eudragit®. Contrary, higher rigidity of formed gel layer was observed using combination of HPMC and Eudragit®. Not only the type and concentration of Eudragit®, but also the presence of the surfactant in water dispersions played a key role in the dissolution characteristics. The dissolution profile close to zero-order kinetic was achieved from the sample containing levetiracetam directly granulated by the water dispersion of Eudragit® NE (5% of solid polymer per tablet) with a relatively high amount of surfactant nonoxynol 100 (1.5%). The initial burst release of drug was reduced to 8.04% in 30 min (a 64.2% decrease) while the total amount of the released drug was retained (97.02%).

Swellable-matrix tablets based on hydrophilic polymers are simple dosage forms with controlled release, which are currently used in pharmacotherapy of many diseases. The most commonly used polymer is (hydroxypropyl)methylcellulose (hypromelose). The drug release from these systems through a gel layer can be modified using additives changing the drug dissolution profile. These excipients can be divided into several groups such as indifferent fillers (soluble or insoluble), substances increasing drug solubility (ß-cyclodextrins), excipients forming interactive products (polymers, ionexchange resins, surfactants) and substances influencing pH in matrix microenvironment (acid and basic modifiers).

BACKGROUND: We hypothesised that different solutions for submucosal injection may influence early healing of endoscopic mucosal resection (EMR). The aim of this study was to evaluate histological and immunological changes after EMR in experimental pigs. MATERIALS AND METHODS: Two parallel EMRs on the anterior and posterior wall of the gastric body were performed by means of the cap technique in 21 female pigs. A glycerol-based solution (anterior EMR) and hydroxypropyl methylcellulose solution (posterior EMR) were applied for submucosal injection. The animals were sacrificed 7 days later, and tissue sections of all EMRs were stained using combined trichrome. Computer image analysis was used for objective evaluation of elastic and collagen fibres content. Two-colour indirect immunophenotyping of blood and gastric samples were performed using mouse anti-pig monoclonal antibodies. RESULTS: The values of collagen fibre content 7 days after EMR were significantly higher in lesions after the use of solution A in comparison with solution B (2.10 +/- 0.25% versus 1.57 +/- 0.25%, p = 0.009). Concordant results were found in elastic fibres (3.23 +/- 0.49% versus 2.93 +/- 0.61%, p = 0.018). No systemic changes in major leukocyte subpopulations were found. In gastric tissue, lymphocyte subsets exhibited only minor changes. CD4(+) T-lymphocytes were increased in the healing tissue after EMR using solution A (17.08 +/- 9.24% versus 9.76 +/- 7.97%, p = 0.011). Significant increase of SWC3(+) leukocytes was observed after EMR using solution B (47.70 +/- 25.41% versus 18.70 +/- 12.16%, p = 0.001). CONCLUSIONS: The use of glycerol-based solution for submucosal injection was associated with more pronounced histological signs of early healing of EMRs compared with hydroxypropyl methylcellulose.

V oblasti technologie léků nachází rozsáhlé uplatnění gelové systémy vysokomolekulárních látek, které mají řadu výhod, jako jsou nízká toxicita, dostupnost, ojedinělé fyzikální vlastnosti, biokompatibilita, mukoadhezivita a další. Gelové systémy se používají v oblasti lokální i celkové terapie, v tvarově specifických i tvarově nespecifických lékových formách, v lécích první, druhé i třetí generace. Významnou skupinu gelů, které se používají v oblasti farmacie, tvoří gely hydrofilní, neboli hydrogely, tvořené nejčastěji hydrofilními polymery přírodního, polosyntetického a syntetického původu. Přestože celulosové deriváty jako představitelé polymerů polosyntetického původu se uplatňují ve farmaceutické technologii po dlouhou dobu, v jejich výzkumu se neustává a hledají se další možnosti jejich uplatnění. Mezi jejich přednosti patří zejména bezpečnost, snadná dostupnost a relativně nízká cena. Přehledový článek popisuje vybrané celulosové deriváty, jejich vlastnosti a využití ve farmaceutické technologii se zaměřením na uplatnění v oblasti tvorby gelových systémů.
The field of drug technology widely ulilizes gel systems of high-molecular substances, which have a number of advantages, such as low toxicity, availability, unique physical properties, biocompatibility, mucoadhesivity, and others. Gel systems are used in the field of local as well as general therapy, in both shape-specific and shape-non-specific dosage forms, in medicaments of the first, second, and third generations. An important group of gels employed in pharmacy are hydrophilic gels or hydrogels, most frequently composed of hydrophilic polymers of natural, semisynthetic and synthetic origin. Though cellulose derivatives as the representatives of polymers of semisynthetic origin are used in pharmaceutical technology for a long time, their research continues and their other possible uses are being searched for. Their advantages include especially safety, easy availability, and a relatively low price. The review paper describes selected cellulose derivatives, their properties and uses in pharmaceutical technology with regard to their use in the field of production of gel systems.

Práca je zameraná na formuláciu antihistaminika – cetirizínu do hydrogélov. Na prípravu hydrogélov boli použité deriváty celulózy – hydroxyetylcelulóza (HEC) a metylcelulóza (MC). Pretože nevyhnutnou súčasťou hydrogélov sú pomocné látky zo skupiny humektantov, v príspevku sa sleduje ich vplyv (glycerolu – GL, propylénglykolu – PG a sorbitolu – SO) na reologické vlastnosti a farmaceutickú dostupnosť cetirizínu pri jeho formulácii do hydrogélu. Z dosiahnutých výsledkov štúdie vyplýva, že z hľadiska dermálnej aplikácie sú pre cetirizín v tomto štádiu výskumu optimálne hydrogély následovného zloženia: 3% HEC + 15% GL a 2,5% MC + 10% SO.
The paper is concerned with the formulation of the antihistamine cetirizine into hydrogels. The cellulose derivatives hydroxyethylcellulose (HEC) and methylcellulose (MC) were employed to prepare hydrogels. As auxiliary substances from the group of humectants are indispensable components of hydrogels, the paper examines their effect (glycerol – GL, propylene glycol – PG, and sorbitol – SO) on the rheological properties and pharmaceutical availability of cetirizine in its formulation into hyrogel. The obtained results show that from the viewpoint of dermal administration for cetirizine at this stage of research hydrogels of the following composition are optimal: 3 %HEC + 15% GL and 2.5% MC + 10% SO.

Hodnotenie farmaceutickej dostupnosti liečiva z topických liekov sa spravidla zameriava na posúderúe schopnosti liekovej formy uvolniť liečivo na jeho terapeutické pôsobenie. Na tieto účely sa používajú permeačné bunky, v ktorých sa uplatňujú tuhé membrány, medzi ktoré patrí aj celofán. v rámci štúdia derivátov kyseliny fenylkarbámovej s lokálne anestetickým účinkom sa v tejto práci hodnotila lieková forma pre topickú aplikáciu s obsahom potenciálneho liečiva XIX M, chemicky N-[2-(2-heptyloxyfenylkarbamoyloxy)-etyl]-morfoliniumchlorid a potenciálneho liečiva XXII B, chemicky N-[2-(2-oktyloxyfenylkarbamoyloxy)-etyl]-dimetylamoniumchlorid. Vybranou pomocnou látkou pri formulácii liekovej formy bol derivát celulózy - metylcelulóza v stúpajúcej koncentrácii od 0,5 po 1,5 %. Na základe dosiahnutých výsledkov možno uviesť, že v práci študovaný polymér metylcelulóza so stúpajúcou koncentráciou spomaľoval uvolňovanie oboch potenciálnych liečiv približne rovnako a môže byť použitý v praxi na predĺženie lokálne anestetického účinku.
Evaluation of pharmaceutical availability of drugs from topical preparations is usually aimed to evaluate the capability of the dosage form to release the drug for its therapeutic action. Permeation cells have been used for this purpose, employing solid membranes includii^ cellophane. Within a study of phenylcarbamic acid derivatives with local anaesthetic effect, the present paper evaluates the dosage form for topical administration containing potential drug XIX M, the chemical name N-[2-(2-heptyloxyphenylcarbamoyloxy)-ethyl]-morpholinium chloride and potential drug XXII B, the chemical name N-[2-(2-octyloxyphenylcarbamoyloxy)-ethyll-dimethylammonium chloride. The selected auxiliary substance in the formulation of the dosage form was a cellulose derivative - methylcellulose in increasing concentrations from 0.5 to 1.5 %. On the basis of the obtained results it can be stated that the examined polymer methylcellulose with increasii^ concentration decelerated the release of both potential drugs approximately in the same way and that it can be used in practice to prolong the local anaesthetic effect.
