Photosystem II (PSII) is an intrinsic membrane protein complex that functions as a light-driven water:plastoquinone oxidoreductase in oxygenic photosynthesis. Electron transport in PSII is associated with formation of reactive oxygen species (ROS) responsible for oxidative modifications of PSII proteins. In this study, oxidative modifications of the D1 and D2 proteins by the superoxide anion (O2•-) and the hydroxyl (HO•) radicals were studied in WT and a tocopherol cyclase (vte1) mutant, which is deficient in the lipid-soluble antioxidant α-tocopherol. In the absence of this antioxidant, high-resolution tandem mass spectrometry was used to identify oxidation of D1:130E to hydroxyglutamic acid by O2•- at the PheoD1 site. Additionally, D1:246Y was modified to either tyrosine hydroperoxide or dihydroxyphenylalanine by O2•- and HO•, respectively, in the vicinity of the nonheme iron. We propose that α-tocopherol is localized near PheoD1 and the nonheme iron, with its chromanol head exposed to the lipid-water interface. This helps to prevent oxidative modification of the amino acid's hydrogen that is bonded to PheoD1 and the nonheme iron (via bicarbonate), and thus protects electron transport in PSII from ROS damage.

Potato (Solanum tuberosum) mutant (ST) lacking one isoform of manganese-stabilizing protein (MSPI) of photosystem II exhibited besides spontaneous tuberization also growth changes with strongly impaired root system development. Previous studies revealed marked changes in carbohydrate levels and allocation within ST plant body. To verify causal relationship between changed carbohydrate balance and root growth restriction we engaged dark grown sucrose-supplied root organ-cultures of ST plants to exclude/confirm shoot effects. Unexpectedly, in ST root cultures we observed large alterations in growth and architecture as well as saccharide status similar to those found in the intact plant roots. The gene expression analysis, however, proved PsbO1 transcript (coding MSPI protein) neither in ST nor in WT root-organ cultures. Therefore, the results point to indirect effects of PsbO1 allele absence connected possibly with some epigenetic modulations.

Chromera velia is a marine photosynthetic relative of human apicomplexan parasites. It has been isolated from coral reefs and is indicted for being involved in symbioses with hermatypic corals. C. velia has been subject to intensive research, but still very little is known of its response to light quality and quantity. Here, we have studied the growth and compositional responses of C. velia to culture under monochromatic light (blue, green or red), at two photon flux densities (PFD, 20 and 100 μmol photons m-2 s-1). Our results show that C. velia growth rate is unaffected by the quality of light, whereas it responds to PFD. However, light quality influenced cell size, which was smaller for cells exposed to blue monochromatic light, regardless of PFD. PFD strongly influenced carbon allocation: at 20 μmol photons m-2 s-1, carbon was mainly allocated into proteins while at 100 μmol photons m-2 s-1, carbon was allocated mainly into carbohydrate and lipid pools. The blue light treatment caused a decrease in the lipids and carbohydrates to proteins and thus suggested to affect nitrogen metabolism in acclimated cells. Whole-cell absorption spectra revealed the existence of red-shifted chlorophyll a antenna not only under red light but in all low PFD treatments. These findings show the ability of C. velia to successfully adapt and thrive in spectrally very different environments of coral reefs.
This study presents a mathematical model, which expresses the absorbance of a photosynthetic sample as a non-linear polynomial of selected reference absorbance. The non-linearity is explained by inhomogeneities of a product of pigment concentration and light path length in the sample. The quadratic term of the polynomial reflects the extent of inhomogeneities, and the cubic term is related to deviation of the product distribution from a symmetric one. The model was tested by measurements of suspension of unstacked tobacco thylakoid membranes of different chlorophyll concentrations in cuvettes of different thicknesses. The absorbance was calculated from the diffuse transmittance and reflectance of sample, illuminated by perpendicular collimated light. The evaluated quantity was a sensitivity defined as the relative difference between the sample absorbance and the reference absorbance to the reference absorbance. The non-linearity of sample absorbance was demonstrated by a characteristic deviation of the sensitivity spectrum from a constant value. The absorbance non-linearity decreased on an increase of the product of pigment concentration and cuvette thickness. The model suggests that the sieve and detour effects influence the absorbance in a similar way. The model may be of interest in modeling of leaf or canopy optics including light absorption and scattering.

In the model cyanobacterium Synechocystis sp. PCC 6803, the terminal enzyme of chlorophyll biosynthesis, chlorophyll synthase (ChlG), forms a complex with high light-inducible proteins, the photosystem II assembly factor Ycf39 and the YidC/Alb3/OxaI membrane insertase, co-ordinating chlorophyll delivery with cotranslational insertion of nascent photosystem polypeptides into the membrane. To gain insight into the ubiquity of this assembly complex in higher photosynthetic organisms, we produced functional foreign chlorophyll synthases in a cyanobacterial host. Synthesis of algal and plant chlorophyll synthases allowed deletion of the otherwise essential native cyanobacterial gene. Analysis of purified protein complexes shows that the interaction with YidC is maintained for both eukaryotic enzymes, indicating that a ChlG-YidC/Alb3 complex may be evolutionarily conserved in algae and plants.

We studied the resistance of Parmotrema austrosinense to UV-B stress. We focused on the effects of a high dose UV-B radiation on the content of chlorophylls, carotenoids and UV-B screening compounds. Photosynthetic parameters were measured by chlorophyll fluorescence (potential and effective quantum yields, photochemical and non-photochemical quenching) and evaluated in control and UV-B-treated lichens. Lichens from two different locations in Cordoba, Argentina, were selected: (i) high altitude and dry plots at (Los Gigantes) and (ii) lowland high salinity plots (Salinas Grandes). UV-B treatment led to a decrease in the content of photosynthetic pigments and UV-B screens (absorbance decrease in 220-350 nm) in the samples from Salinas Grandes, while in Los Gigantes samples, an increase in UV-B screen content was observed. Chlorophyll fluorescence parameters showed a UV-B-induced decline in FV /FM , ΦPSII and qP indicating limitation of primary photosynthetic processes in photosystem II (PSII) of symbiotic alga, more pronounced in Salinas Grandes samples. Protective mechanism of PSII were activated by the UV-B treatment to a higher extent in samples from Salinas Grandes (NPQ 0.48) than in Los Gigantes samples (NPQ 0.26). We concluded that site-related characteristics, and in particular different UV-B radiation regimen, had a strong effect on resistance of the photosynthetic apparatus of P. austrosinense to UV-B radiation.

It has previously been shown that the long-term treatment of Arabidopsis thaliana with the chloroplast inhibitor lincomycin leads to photosynthetic membranes enriched in antennas, strongly reduced in photosystem II reaction centers (PSII) and with enhanced nonphotochemical quenching (NPQ) (Belgio et al. Biophys J 102:2761-2771, 2012). Here, a similar physiological response was found in the microalga Chromera velia grown under high light (HL). In comparison to cells acclimated to low light, HL cells displayed a severe re-organization of the photosynthetic membrane characterized by (1) a reduction of PSII but similar antenna content; (2) partial uncoupling of antennas from PSII; (3) enhanced NPQ. The decrease in the number of PSII represents a rather unusual acclimation response compared to other phototrophs, where a smaller PSII antenna size is more commonly found under high light. Despite the diminished PSII content, no net damage could be detected on the basis of the Photosynthesis versus irradiance curve and electron transport rates pointing at the excess capacity of PSII. We therefore concluded that the photoinhibition is minimized under high light by a lower PSII content and that cells are protected by NPQ in the antennas.

This study describes the impacts of inorganic carbon limitation on the photosynthetic efficiency and operation of photosynthetic electron transport pathways in the biofuel-candidate microalga Nannochloropsis oculata. Using a combination of highly-controlled cultivation setup (photobioreactor), variable chlorophyll a fluorescence and transient spectroscopy methods (electrochromic shift (ECS) and P700 redox kinetics), we showed that net photosynthesis and effective quantum yield of Photosystem II (PSII) decreased in N. oculata under carbon limitation. This was accompanied by a transient increase in total proton motive force and energy-dependent non-photochemical quenching as well as slightly elevated respiration. On the other hand, under carbon limitation the rapid increase in proton motive force (PMF, estimated from the total ECS signal) was also accompanied by reduced conductivity of ATP synthase to protons (estimated from the rate of ECS decay in dark after actinic illumination). This indicates that the slow operation of ATP synthase results in the transient build-up of PMF, which leads to the activation of fast energy dissipation mechanisms such as energy-dependent non-photochemical quenching. N. oculata also increased content of lipids under carbon limitation, which compensated for reduced NAPDH consumption during decreased CO2 fixation. The integrated knowledge of the underlying energetic regulation of photosynthetic processes attained with a combination of biophysical methods may be used to identify photo-physiological signatures of the onset of carbon limitation in microalgal cultivation systems, as well as to potentially identify microalgal strains that can better acclimate to carbon limitation.

Low oxygen conditions occur in grass sites due to high and frequent precipitation, poor soil quality, and over-irrigation followed by slow drainage. Three warm-season and one cool-season grass were analyzed at metabolic level during a time-course experiment performed in a controlled anoxic environment. Prolonged oxygen depletion proved detrimental by leading to premature death to all the species, with the exception of seashore paspalum. Moreover, the anoxia tolerance observed in these grasses has been associated with slow use of carbohydrates, rather than with their relative abundance, which was more important than their antioxidant capacity. Further physiological characterization of eight seashore paspalum genotypes to anoxia was also performed, by examining the variation in photosystem II (PSII) efficiency and gas exchange during post-anoxia recovery. Multivariate analysis highlighted the presence of three main clusters of seashore paspalum genotypes, characterized by different ability to restore the PSII photochemistry during recovery after one day of anoxia. Taken together, our data demonstrate that the analysis of post-anoxia recovery of fluorescence and gas exchange parameters can represent a fast and reliable indicator for selecting species and cultivars more able to acclimate their photosynthetic apparatus.

MAIN CONCLUSION: The basal streptophyte Klebsormidium and the advanced Zygnema show adaptation to terrestrialization. Differences are found in photoprotection and resistance to short-term light changes, but not in CO 2 acquisition. Streptophyte green algae colonized land about 450-500 million years ago giving origin to terrestrial plants. We aim to understand how their physiological adaptations are linked to the ecological conditions (light, water and CO2) characterizing modern terrestrial habitats. A new Klebsormidium isolate from a strongly acidic environment of a former copper mine (Schwarzwand, Austria) is investigated, in comparison to Klebsormidium cf. flaccidum and Zygnema sp. We show that these genera possess different photosynthetic traits and water requirements. Particularly, the Klebsormidium species displayed a higher photoprotection capacity, concluded from non-photochemical quenching (NPQ) and higher tolerance to high light intensity than Zygnema. However, Klebsormidium suffered from photoinhibition when the light intensity in the environment increased rapidly, indicating that NPQ is involved in photoprotection against strong and stable irradiance. Klebsormidium was also highly resistant to cellular water loss (dehydration) under low light. On the other hand, exposure to relatively high light intensity during dehydration caused a harmful over-reduction of the electron transport chain, leading to PSII damages and impairing the ability to recover after rehydration. Thus, we suggest that dehydration is a selective force shaping the adaptation of this species towards low light. Contrary to the photosynthetic characteristics, the inorganic carbon (C i ) acquisition was equivalent between Klebsormidium and Zygnema. Despite their different habitats and restriction to hydro-terrestrial environment, the three organisms showed similar use of CO2 and HCO3- as source of Ci for photosynthesis, pointing out a similar adaptation of their CO2-concentrating mechanisms to terrestrial life.
