Statin use is associated with increased calorie intake and consequent weight gain. It is speculated that statin-dependent improvements in lipid profile may undermine the perceived need to follow lipid-lowering and other dietary recommendations leading consequently to increased calorie intake. However, increases in calorie intake in statin users may also be related to statin-dependent decreases in satiety factors such as leptin, an adipocyte-derived adipokine. The objective of our study was to examine the direct effects of statins on leptin expression. Adipocytes are the main source of circulating leptin. Therefore, we examined the effects of atorvastatin and simvastatin on leptin expression in cultured human white adipocytes. We show that treatment of white adipocytes with simvastatin and atorvastatin decreases leptin mRNA expression (simvastatin: P = 0.008, atorvastatin: P = 0.03) and leptin secretion (simvastatin: P = 0.0001, atorvastatin: P = 0.0001). Both simvastatin and atorvastatin mediate decreases in leptin expression via extracellular-signal-regulated kinases 1/2 and peroxisome proliferator-activated receptor gamma pathways (simvastatin: P = 0.01, atorvastatin: P = 0.026). Additionally, statin treatment also induced expected increases in adiponectin, while decreasing monocyte chemoattractant protein 1 (MCP1) mRNA. Furthermore, statins increased secretion of both total as well as high molecular weight adiponectin while decreasing MCP1 secretion. To conclude, statins act directly on human white adipocytes to regulate adipokine secretion and decrease leptin expression. Leptin is an important satiety factor. Hence, statin-dependent decreases in leptin may contribute, at least in part, to increases in food intake in statin users.

Because of the increasing prevalence worldwide, there has been an increase in the amount of studies dealing with reciprocal associations between asthma and obesity.

Agonisté ß3-adrenergních receptorů jsou látky, které stimulují lipolýzu v bílé tukové tkáni a termogenezi v hnědé tukové tkáni, čehož by mohlo být využito při léčbě obezity a s ní spojeného metabolického syndromu. Cílem této práce bylo zhodnotit vliv šestitýdenního p.o. podávání látky B496 – ethylesteru (4-{2-[(2hydroxy-3-(4- -ethylkarbamoyl)fenoxypropyl)amino]ethyl}fenoxy) octové kyseliny ve formě ve vodě rozpustného hydrochloridu laboratorním potkanům kmene Wistar samčího pohlaví krmených hypertučnou dietou na koncentrace glukosy, triacylglyceridů, celkového cholesterolu a leptinu v séru a na váhový přírůstek. Zvířata byla rozdělena na skupinu léčenou látkou B496 a na skupinu kontrolní. Léčené skupině byla látka B496 rozpuštěna v destilované vodě v koncentraci 5 mg/1 kg vody. Testovaná látka B496 při dlouhodobém podávání laboratorním potkanům významně snižuje sérové koncentrace glukosy (-26 %, p<0,01), triacylglyceridů (-21 %, p<0,05) a leptinu (-43 %, p<0,01). Dále byl zkoumán vliv jednorázového i.p. podání (1 mg/kg hmotnosti zvířete) nově syntetizované látky B496 a látky BRL-37344 na koncentrace sérového leptinu u inbredních myší C57Bl/6J. Látka BRL-37344 vedla ke statisticky významnému snížení koncentrace leptinu v séru (-55 %, p<0,001). Takové snížení se nepodařilo prokázat u testované látky B496.
ß3-adrenoreceptor agonists can stimulate lipolysis in the white adipose tissue and thermogenesis in the brown adipose tissue. These activities could be useful in the treatment of obesity and the associated metabolic syndrome. The effects of six-week oral administration of the newly synthesized substance B496 (methyl-4-[2- -[2-hydroxy-3-(4-ethylcarbamoyl)phenoxyprophyl]amino]etyl)-phenoxyacetate hydrochloride) on serum glucose, triglycerides, total cholesterol, and leptin levels were studied in male Wistar rats fed with a high-fat diet. The animals were divided into a group treated with B496 (5 mg dissolved in 1 litre of water) and a control group. The results indicated a significant reduction in serum glucose levels (-26 %, p<0,01), triacylglyceride levels (-21 %, p<0,05) and leptin levels (-43 %, p<0,01). Further the effect of a single intraperitoneal dose (1 mg/kg) of B496 and BRL-37344 on serum leptin levels in the C57Bl/6J mouse was investigated. Administration of BRL-37344 resulted in a significant decrease in serum leptin levels (-55 %, p<0,001). This reduction was not demonstrated by newly synthesized substance B496.

Leptin is a circulating pleiotropic hormone that play an important role in appetite control, fat metabolism, regulation of body weight, fetus growth, growth and aging of adults and hematopoiesis. It is expressed abundantly and specifically in the adipose tissue. A liver cell with developed steatosis represents a cell metabolism similar to metabolism of cells of adipose tissue. Analyses of serum leptin and free leptin receptor in the serum of patients with steatosis showed significant variations from reference limits of normal values. However in liver tissue with verified steatosis detection of mRNA gene for leptin was not proven. Such expression of ob gene for leptin was not found even in the liver tissue without steatosis. With respect to the absence of ob gene expression, the direct effect of ob gene expression on other parameters of leptin metabolism could not be evaluated. The RT-PCR method with verified specificity and satisfying sensitivity was developed. The results obtained from analysis of serum leptin and free leptin receptor in the serum are presented and evaluated. The used methods were verified and reference limits for Czech population were defined in dependence on age and other clinical parameters.
