Changes of amino acid concentrations (glutamic acid, glutamine, asparagine, aspartate, proline, tryptophan, alanine, glycine, valine and serine), gas-exchange parameters (net photosynthetic rate, transpiration rate, stomatal conductance and intercellular CO(2) concentration) and nitrate levels in Lactuca serriola L. under airborne particulate matter (PM) contamination reported here reveal their role in plant chronic stress adaptation. Results of the pot experiment confirmed the toxic effect of trace elements present in PM for lettuce. PM applied to soil or on the lettuce leaves were associated with the strong inhibition of above-ground biomass and with the enhancement of plant trace element contents. The significant changes of amino acid levels and leaf gas-exchange parameters of the plants showed strong linear dependences on PM contamination (R(2)=0.60-0.99). PM application on leaves intensified toxic effect of trace elements (As, Pb, Cr and Cd) originating from PM by shading of the leaf surface. The plant accumulation of nitrate nitrogen after PM contamination confirmed to block nitrate assimilation.

The effect of heterozygous duplications of SHOX and associated elements on Lėri-Weill dyschondrosteosis (LWD) and idiopathic short stature (ISS) development is less distinct when compared to reciprocal deletions. The aim of our study was to compare frequency and distribution of duplications within SHOX and associated elements between population sample and LWD (ISS) patients. A preliminary analysis conducted on Czech population sample of 250 individuals compared to our previously reported sample of 352 ISS/LWD Czech patients indicated that rather than the difference in frequency of duplications it is the difference in their distribution. Particularly, there was an increased frequency of duplications residing to the CNE-9 enhancer in our LWD/ISS sample. To see whether the obtained data are consistent across published studies we made a literature survey to get published cases with SHOX or associated elements duplication and formed the merged LWD, the merged ISS, and the merged population samples. Relative frequency of particular region duplication in each of those merged samples were calculated. There was a significant difference in the relative frequency of CNE-9 enhancer duplications (11 vs. 3) and complete SHOX (exon1-6b) duplications (4 vs. 24) (p-value 0.0139 and p-value 0.000014, respectively) between the merged LWD sample and the merged population sample. We thus propose that partial SHOX duplications and small duplications encompassing CNE-9 enhancer could be highly penetrant alleles associated with ISS and LWD development.

We conducted a pot experiment with biochar (BC), wood ash (WA), and humic substances (HS) to investigate their effect on As, Zn, Cu, Cd and Pb mobility in soil, as well as enzyme activities involved in C-, N-, and P-cycles, and Eisenia foetida toxicity in multi-contaminated soils. Amendments were dosed to increase еру soil pH from initial 6.0 to ∼6.5 and ∼7.0. Applying amendments has revealed, that WA significantly immobilized Cu, Zn and Pb, BC - Cu and Zn, and HS decreased solely Cu mobility in soil. The partition indices of Zn, Cu, and Pb, quantitatively describing the bioavailable species of elements in soil, were the lowest for WA. Changes in the water-soluble species of metals were more pronounced than in the exchangeable ones for all amendments. An opposite effect was observed on enzyme activity and earthworm toxicity for the WA and carbonaceous amendments. The BC and HS provided favourable soil conditions to dehydrogenase, β-glucosidase, urease activity and fluorescein diacetate hydrolysis, while WA significantly decreased the activity of all the mentioned enzymes in soil. The results are supported by an enzymes-based weighted mean index, being the highest for BC and HS and the lowest for WA (lower than in the control sample). At the same time, WA was suitable to eliminate the trace elements' stress to earthworms (biomass endpoints and cocoons production). Our data revealed that each amendment has its own advantages and disadvantages. The choice of the most suitable amendment therefore should always be made within an integral approach and based on the purpose of remediation.

The worldwide spread of pathogenic microorganisms poses a significant risk to human health. Electrochemical biosensors have emerged as dependable analytical tools for the point-of-care detection of pathogens and can effectively compensate for the limitations of conventional techniques. Real-time analysis, high throughput, portability, and rapidity make them pioneering tools for on-site detection of pathogens. Herein, this work comprehensively reviews the recent advances in electrochemical biosensors for pathogen detection, focusing on those based on the classification of recognition elements, and summarizes their principles, current challenges, and prospects. This review was conducted by a systematic search of PubMed and Web of Science databases to obtain relevant literature and construct a basic framework. A total of 171 publications were included after online screening and data extraction to obtain information of the research advances in electrochemical biosensors for pathogen detection. According to the findings, the research of electrochemical biosensors in pathogen detection has been increasing yearly in the past 3 years, which has a broad development prospect, but most of the biosensors have performance or economic limitations and are still in the primary stage. Therefore, significant research and funding are required to fuel the rapid development of electrochemical biosensors. The overview comprehensively evaluates the recent advances in different types of electrochemical biosensors utilized in pathogen detection, with a view to providing insights into future research directions in biosensors.

Východiska: Validizační studie v ošetřovatelském procesu umožňují vyjádření míry platnosti komponent NANDA-I pro různé soubory osob a v různých oblastech péče. Bolest jako ošetřovatelská diagnóza v péči o rodící ženu je jedním z nejčastějších problémů, které porodní asistentky v praxi posuzují, řeší a hodnotí. Validované definující charakteristiky ošetřovatelských diagnóz jsou rozhodující pro správnou diagnostiku typu a intenzity bolesti a pro navržení efektivní léčby. Cíl: Cílem práce bylo validizovat definující charakteristiky ošetřovatelské diagnózy 00132 Akutní bolest vybraným souborem expertek – porodních asistentek a zjistit, které definující charakteristiky (určující znaky, související faktory) porodní asistentky považují za hlavní a které definující charakteristiky za vedlejší. Metody: Pro obsahovou validizaci byl využit Fehringův model validity diagnostického obsahu (Diagnostic Content Validity Model). Expertky – porodní asistentky splňovaly kritérium bakalářského stupně vzdělá- ní v oboru porodní asistence a klinické praxe na oddělení porodního sálu. Výzkum probíhal současně ve Fakultní nemocnici Olomouc, v Nemocnici Šternberk a v Nemocnici Uherské Hradiště. Měřící nástroj pro posouzení významnosti diagnostických prvků byl sestrojený pomocí jednotlivých definujících charakteristik ošetřovatelské diagnózy z NANDA-I, Taxonomie II. Výsledky: Z celkového počtu 22 definujících charakteristik (18 určujících znaků, 4 související faktory) považují porodní asistentky za hlavní (majoritní) 9 definujících charakteristik. Mezi hlavní definující charakteristiky (určující znaky) s váženým skóre (VS) vyšším než 0,75 porodní asistentky zařadily: úlevová poloha z důvodu vyhnutí se bolesti (VS 0,82); expresivní chování (VS 0,77); pozorované známky bolesti (VS 0,80); slovní vyjádření bolesti (VS 0,84); změny dýchání (VS 0,80); změny krevního tlaku (VS 0,82); změny srdeční- ho rytmu (VS 0,86). Mezi hlavní související faktory zařadily tyto charakteristiky: biologické faktory – porodní mechanismus (VS 0,94) a faktory psychogenní (VS 0,80). Závěry: Z výsledků tohoto šetření vyplývá, že dotazované porodní asistentky považují z celkového nabízeného počtu 22 definujících charakteristik (18 určujících znaků a 4 související faktory) 9 za hlavní – majoritní pro celkové posouzení stavu klientky – rodičky přicházející na porodní sál k porodu.
Background: Validation studies allow expression of the degree of validity of components NANDA-I for different sets of people and in different areas of care. Pain is one of the most common problems that midwives in practice assess, evaluate and solve. Validated defining characteristics of the nursing diagnoses are crucial for correct diagnosis of the type and intensity of pain, and for proposal of effective treatment. Aim: The aim of the study was to validate defining characteristics of the nursing diagnosis Acute pain 00132 by a selected file of experts-midwives, and see which characteristics they consider major and minor. Methods: The study followed Fehring Diagnostic Content Validity Model. Experts-midwives met the criterion of bachelor degree in midwifery and clinical practice in the delivery room. The research was conducted simultaneously in Olomouc region. Measuring tool for assessing the significance of diagnostic elements was constructed using individual defining characteristics of the nursing diagnoses of NANDA-I Taxonomy II. Results: Out of the 22 defining characteristics considered by experts-midwives nine characteristics were described as major with weighted score (WS) greater than 0,75: antalgic position due to the avoidance of pain (WS 0,82); expressive behavior (WS 0,77); observed signs of pain (WS 0,80); verbal expression of pain (0,84); changes in breathing (0,80); changes in blood pressure (WS 0,82); changes in heart rhythm (WS 0,86). Among the major related factors were selected the following characteristics: biological factors - birth mechanism (WS 0,94) and psychogenic factors (WS 0,80). Conclusion: The results of this study indicate that midwives consider nine defining characteristics and two related factors as major for assesing acute pain of mothers coming to the delivery room for delivery.
Simultaneous electromembrane extraction (EME) of six trace metal cations (Cu2+ , Zn2+ , Co2+ , Ni2+ , Pb2+ , Cd2+ ) from saline samples was investigated. CE with capacitively coupled contactless conductivity detection (C4 D) was used to determine the metals in acceptor solutions due to its excellent compatibility with the minute volumes of acceptor solutions. Bis(2-ethylhexyl)phosphate (DEHPA) was selected as a suitable nonselective modifier for EME transport of target metal cations. Both, the individual effect of each major inorganic cation (Na+ , K+ , Ca2+ , Mg2+ ) and their synergistic effect on EME of the trace metal cations were evaluated. In both cases, a decrease in extraction efficiency was observed when major inorganic cations were present in the sample. This effect was more significant for Ca2+ and Mg2+ . The system was optimized for simultaneous extractions of the six target metals from saline samples (50 mM Na+ , 5 mM Mg2+ , 1 mM K+ , and 1 mM Ca2+ ) and following EME conditions were applied. Organic phase consisted of 1-nonanol containing 1% (v/v) DEHPA, acceptor solution was 1 M acetic acid (HAc) and sample pH was adjusted to 5. Sample was stirred at 750 rpm and EMEs were carried out at extraction potential of 10 V for 20 min. The method presented a repeatability between 8 and 21.8% (n = 5), good linearity in 0.5-10 μM concentration range (R2 = 0.987-0.999) and LOD better than 2.6 nM. Applicability of the EME-CE-C4 D method to the analyses of metal cations in drinking water, seawater, and urine samples was also demonstrated.
BACKGROUND: High-throughput sequencing (HTS) has revolutionized the way in which epigenetic research is conducted. When coupled with fully-sequenced genomes, millions of small RNA (sRNA) reads are mapped to regions of interest and the results scrutinized for clues about epigenetic mechanisms. However, this approach requires careful consideration in regards to experimental design, especially when one investigates repetitive parts of genomes such as transposable elements (TEs), or when such genomes are large, as is often the case in plants. RESULTS: Here, in an attempt to shed light on complications of mapping sRNAs to TEs, we focus on the 2,300 Mb maize genome, 85% of which is derived from TEs, and scrutinize methodological strategies that are commonly employed in TE studies. These include choices for the reference dataset, the normalization of multiply mapping sRNAs, and the selection among sRNA metrics. We further examine how these choices influence the relationship between sRNAs and the critical feature of TE age, and contrast their effect on low copy genomic regions and other popular HTS data. CONCLUSIONS: Based on our analyses, we share a series of take-home messages that may help with the design, implementation, and interpretation of high-throughput TE epigenetic studies specifically, but our conclusions may also apply to any work that involves analysis of HTS data.
