The objective of the project is to study the binding of platinum cytostatics to DNA of human tumor cells (cell lines including chemoresistant ones, tumor samples obtained from patients) and the influences of metallothionein (MT) on this interaction. Amounts of platinum bound to DNA influence successfulness of their anticancer effect. Newly developed electrochemical methods, will be used as an analytical tool for study of Pt-DNA and Pt-MT complexes. Liposome based transporter carrying platinum cytostatics, as well as carrying MTs and/or zinc ions that can modulate the response of cancer cells to treatment with platinum cytostatics will be prepared and tested in vitro and in vivo. We assume that our results and developed techniques will improve usage of platinum cytostatics and will help to reveal the mechanisms of resistance to those cytostatics. Another benefit will be the introduction of new electrochemical method for determination of Pt and its interaction with DNA and MT that may be helpful for clinical practice.
Cílem projektu je studium interakce platinových cytostatik s DNA získané z lidských nádorových buněk (buněčné linie včetně chemorezistentních linií, vzorky nádorů získané od pacientů) a studium vlivu metalothioneninů (MT) na tuto interakci. Množství platiny vázané na DNA ovlivňuje účinnost protinádorového účinku platinových cytostatik. K analýze komplexů Pt-DNA a Pt-MT plánujeme využít nově vyvinuté elektrochemické metody. Dále připravíme liposomální transportéry nesoucí platinová cytostatika a rovněž MT nebo zinkové ionty, které ovlivňují odpověď nádorových buněk na platinová cytostatika. Tyto transportéry budeme testovat in vitro a in vivo. Předpokládáme, že naše výsledky a nově vyvinuté techniky zlepší použití platinových cytostatik a přispějí k objasnění mechanismů rezistence k těmto cytostatikům. Dalším přínosem bude zavedení nových elektrochemických metod ke stanovení Pt a její interakce s DNA a MT, které bude použitelné pro klinickou praxi.

Cíl: Zhodnotit možnosti diagnosticko-léčebného postupů u sympatické oftalmie (SO) ve srovnání s literárními podklady. Teoretický podklad: SO je oční zánět autoimunitní povahy, charakterizovaný disbalancí subpopulací lymfocytů v rámci IV. typu přecitlivělosti. Metodika: Do souboru vyšetření SO zahrnující humorální a buněčnou imunitu byl zařazen nukleolární test (NT), hodnotící aktuální stav biosyntetické aktivace lymfocytů v organismu podle morfologie jadérka dané aktivitou transkripce RNA. Kromě klasického histologického vyšetření byla jednou provedena i imunohistochemická verifikace SO pro stanovení subpopulací lymfocytů v enukleovaném bulbu. Materiál: V rámci dvou studií v letech 1979 až 1994 a 1999 až 2017 bylo sledováno a léčeno celkově 5 pacientů mužského pohlaví se SO, která vznikla čtyřikrát po perforačním poranění a jednou po lensektomii s vitrektomií. Věk vzniku SO se pohyboval mezi 4 až 24 let (průměr 12 let). SO vznikla za 10 dnů až 3 měsíce (průměr 1,7 měsíce) po inzultu. U všech se objevil relaps onemocnění v rozmezí 3 měsíců až 38 let (průměr 15 let). Další dva pacienti s SO byli jen konsiliárně vyšetřeni, 16letý chlapec pro relaps SO po extrakci katarakty a 71-letá žena pro SO vzniklou po lensektomii s vitrektomii. Současně byla vyšetřena a porovnána skupina 19 nemocných s jinými typy uveitid. U 16 nemocných (z toho u 13 mužů a chlapců) fakoantigenní uveitidy vznikly po úrazech (perforace a kontuze). U dalších třech pacientů (mladé ženy) se po enukleaci dolorózního bulbu s uveitidou II. či. III. typu přecitlivělosti zklidnila sekundární uveitida na druhém oku bez změny celkové imunosupresivní terapie a ovlivnění NT. Výsledky: Z komplexu provedených imunologických vyšetření byly prokázány změny u SO v hodnotách aktivních lymfocytů periferní krve. U SO byl počet aktivních lymfocytů zvýšen na rozdíl od fakoantigenních uveitid. Mezi oběma skupinami uveitid byl zjištěn u NT statisticky významný rozdíl (p = 0,0134). Diagnózy byly histologicky verifikovány (5 bulbů s SO a 7 bulbů s uveitidami nesoucitné etiologie), což bylo pro diagnostickou rozvahu zásadní. Imunohistochemické vyšetření potvrdilo přítomnost jednotlivých subpopulací T-lymfocytů, makrofágů a také B-lymfocytů. Základem imunosupresivní léčby v počátku byla kombinace prednisonu a azathioprinu, později účinnou terapii představoval cyklosporin. Závěr: NT upozorňuje na aktuální zvýšenou aktivaci lymfocytů bez rozdílu na zastoupení subpopulací a jejich absolutní počet v mechanismu uveitidy u IV. typu přecitlivělosti. Imunosupresivní terapie tuto aktivaci zásadně ovlivňuje poklesem hodnot, především u SO, ale i uveitid dalších typů přecitlivělosti. Imunohistochemické vyšetření prokazuje různé zastoupení jednotlivých typů lymfocytů podle stadia SO.
Aim: To evaluate options of diagnostic and therapeutic procedures of sympathetic ophthalmia (SO) compared with literature data. Backgroud: SO is an ocular autoimmune disease. It is characterized by disbalance in the imunoregulatory T-subsets within cell mediated immune response. Methods: File examination of SO by evaluation humoral and cellular immunity was ranked nukleolar test (NT). It evaulates the current status of lymphocyte activation based on the nucleolar morphology and RNA transcription aktivity. The classical histological examination was supported by immunohistochemical analysis of lymphocytic subpopulations in the eyeball enucleated for. SO in one case. Material: Five boys and men overall were monitored and treated in two studies from 1979 to 1994 and from 1999 to 2017 with SO In four cases it was subsequent after penetrating injuries and once after lensectomy with vitrectomy. The age of patients in the time of onset of SO was between 4 and 24 years (average 12 years). The time interval between insult and onset of SO varied between 10 days and 3 months (average 1.7 months). The relaps of disease appeared in the all cases in the time interval from 3 months to 38 years (average 15 years). Another two patients with SO were examined only in consultation: 16 years old boy with relaps of SO after cataract extraction and 71 years old women with SO subsequent after lensectomy and vitrectomy. There was examined and compared group of 19 patients with other types of uveitis in the same time. The lens-associated uveitis were caused after ocular contusion and penetrating eye injury in 16 patients (13 male patients). In another three cases (young women) with uveitid underlined by II. or III. type of hypersenzitivity the enucleation of dolorous eyeball calmed down the secondary uveitis on their second eye without any change of immunosupressive treatment and without change in NT. Results: There were changes in the complex immunological laboratory tests results in the SO cases in the counts of activated lymphocytes in the peripheral blood. The count of activated lymphocytes was increased in SO cases unlike in lens-associated uveitis. There was detected in NT statistically significant difference (p = 0,0134) between the two groups of uveitis. The histological examination (5 eyes with SO and 7 eyes without sympathetic uveitis) confirmed the diagnoses, supporting basically the clinical diagnosis. The immunohistochemical examination corfirmed the presence of populations of T-lymphocytes, macrophages and also B-lymphocytes. A basis of immunosuppressive therapy was the combination of prednisone and azathioprin at the first time. Effective therapy featured cyklosporine later. Conclusion: The nucleolar test of lymphocytes draws attention of their up-to-day increased activity without the diferentiation of subpopulations and their absolute number increase related to the current activation of type IV. hypersensitivity (cell-mediated) in uveitis mechanism. The immunosupressive therapy calmes down this activation predominantly in SO, but also in other case sof uveitis with different types of hypersensivity. The immunohistochemical examination illustrates different presence of lymphocytic types according to the stage of SO.

Cancer despite the introduction of new targeted therapy remains for many patients a fatal disease. Nanotechnology in cancer medicine has emerged as a promising approach to defeat cancer. Targeted delivery of anti-cancer drugs by different nanosystems promises enhanced drug efficacy, selectivity, better safety profile and reduced systemic toxicity. The article presents an overview of recent developments in cancer nanomedicine. We focus on approved anti-cancer medical products and on the results of clinical studies, highlighting that liposomal and micellar cytostatics or albumin-based nanoparticles have less side effects and are more efficient than "free" drugs. In addition, we discuss results of in vitro and in vivo preclinical studies with lipid, inorganic and polymer nanosystems loaded by anticancer drugs which according to our meaning are important for development of new nanodrugs. Pharmacokinetic characteristics of nanodrugs are discussed and characterization of major nanotechnology systems used for cancer nanomedicine is presented.
