This study was conducted as an in vivo experiment in adult miniature pigs with the aim to test two new biomaterials. An iatrogenic defect was made into the central femoral diaphysis in the experimental animals and subsequently fixated by bridging plate osteosynthesis. Into the defect we implanted a cancellous autograft (control group), a pasty injectable scaffold (EXP A), and a porous 3D cylinder (EXP B). Radiological examination was performed in all animals at 0, 10, 20, 30 weeks after surgical procedure and histological assessment was performed. In the newly formed bone the osteoblastic activity was monitored. In terms of radiology, the most effective method was observed in the control group (completely healed 100%) compared to experimental groups EXP A (70.0%) and EXP B (62.5%). Histological assessment showed a higher cell count in the place of bone defect in the control group compared to experimental groups. Between the experimental groups, a higher count of bone marrow cells was found in group EXP B. Both newly developed biomaterials seem to be suitable as replacements for large bone defects, having good workability and applicability. However, compared to the control group treated with a cancellous autograft, the newly formed bone did not reach the same number of cells settling in and in some cases, full radiological healing was not reached. Nevertheless, the material was found to be grown into the original bone in all cases within the experimental groups. The new biomaterials have a great potential as a substitute in the treatment of large bone defects.

In veterinary medicine, vaginal rings (VRs) are rarely used. However, there are diseases of female dogs' reproductive system which represent a suitable possibility for their usage. An example of such a disease is canine pyometra which can be treated by lipophilic prostaglandin drugs, unfortunately with harmful side effects after systemic administration. The aim of the study was to prove that the matrix VR based on silicone and channel-forming substance can be successfully used as a carrier for a three-day delivery of prostaglandin E2 (PGE2). Based on an in-vitro release study, an optimum channel-forming substance and its concentration were selected. The results were implemented during the construction of VR from the medical grade silicone DDU-4840 with PGE2 (5 mg). Glucose anhydrous in the 30% concentration was chosen as the most functional channel-forming substance due to synergism of osmotic activity and solubility. The DDU-VR containing PGE2 and 30% of glucose anhydrous exhibited excellent mechanical characteristics and ensured 29% drug release through water-filled channels in first-order kinetic manner. This is eight times higher than a sample without glucose where molecular diffusion through the silicone matrix was dominating the release mechanism. Moreover, drug-free VRs were tested for mechanical resistance and the design of removal thread.

The aim of the study was to assess serum hyaluronic acid (HA) and transforming growth factor beta 1 (TGF-β1) concentrations: 1) to differentiate hepatic fibrosis from other forms of liver disease, and 2) for the non-invasive staging of canine liver fibrosis. We also evaluated the association between serum HA concentration and the size of the shunt vessel as an indirect marker of decreased liver clearance in patients with single congenital vascular anomaly. Forty-one healthy client-owned dogs and forty dogs diagnosed with hepatobiliary disease were enrolled in the prospective study. Patients were divided into 4 subgroups: 1) congenital portosystemic shunts (CPSS); 2) parenchymal diseases (a. mild and moderate fibrosis, b. advanced fibrosis and cirrhosis); 3) hepatic neoplasia; 4) biliary tract disorders based on thorough clinical, ultrasound and histopathological examination. Serum HA and TGF-β1 concentrations were measured using ELISA. The HA concentration was significantly increased in patients with advanced liver fibrosis/cirrhosis (P < 0.001) and CPSS (P < 0.001) compared to healthy dogs. Using a cut-off HA concentration of 135.94 ng/ml, the sensitivity and specificity for diagnosis for advanced liver fibrosis/cirrhosis was 100% (95% CI, 50.6–100) and 90.8% (95% CI, 81.6–95.7), respectively. The TGF-β1 levels did not significantly differ among groups (P = 0.180). Negligible correlation was found between serum HA concentration and the size of portosystemic shunt vessel (rs = 0.07; P = 0.831). These findings suggest that serum HA concentration is a potential non-invasive biomarker for advanced liver fibrosis and/or cirrhosis in dogs. The utility of measuring serum concentration of TGF-β1 for diagnosing canine liver fibrosis was not supported.

PURPOSE OF THE STUDY The purpose of the study is to compare the repeatability and reproducibility of quantitative and subjective evaluation of elbow humeroulnar incongruity (HUI) using computed tomography (CT) on an in vivo canine model. MATERIAL AND METHODS HUI was evaluated on canine (n = 50) elbow joints (n = 100). The computed tomography of elbow joints was performed under intravenous sedation. Multiplanar reconstructions (MPR) were produced. HUI was evaluated on sagittal MPR images subjectively and by measuring the subluxation index (SI). The SI was defined by measuring the distance between the centres of two circles, the shape of which corresponded the most with the shape of the trochlear notch of the ulna at sagittal crest and the shape of humeral trochlea. This distance was divided by the radius of the circle (r) defining the humeral trochlea. HUI was subjectively evaluated based on the width of the joint space at the greatest caudal convexity of the trochlear notch of the ulna. Three categories of HUI were established: 0 (congruent), 1 (moderately incongruent), 2 (strongly incongruent). Measurement and evaluation of HUI was conducted by two evaluators twice at a one-month interval between the first and second measurement. The statistical analysis was carried out using the repeated measures ANOVA and the Cohen s kappa coefficient. RESULTS The mean SI was 11.14 (SD 8.703). The SI values measured by two evaluators were statistically significantly different (p < 0.05). Contrarily, there was no statistically significant difference between the two measurements of the same evaluator (p > 0.05). The subjective evaluation of HUI done by two evaluators showed a mean to substantial agreement (Kappa = 0.53-0.79). There was a substantial to almost perfect agreement between the results of two evaluations carried out by a single evaluator (Kappa = 0.79-0.83). DISCUSSION The radiographic detection of moderate incongruity is unreliable, especially on account of wrongly positioned elbow joint, superposition of bone structures and due to the evaluation of three-dimensional bone structure through a two-dimensional image. Evaluation of humeroulnar congruity by computed tomography (CT) enables to assess the congruity of joints without the superposition of neighbouring bone structures. The quantification of humeroulnar incongruity using the SI does not show a higher degree of agreement between two evaluators as against the subjective evaluation of HUI. On the contrary, the agreement between two measurements of a single evaluator was high in both the cases. CONCLUSIONS Dog is a suitable model animal for evaluation of HUI of elbow joints due to frequent incidence of elbow dysplasia associated with HUI. The quantification of HUI at the expense of subjective evaluation of HUI is often overrated in radiological studies. Key words: dog, elbow, humeroulnar incongruity.

A prospective study to create radiographic classification of caudal vertebral anomalies in dogs was performed and the prevalence of the disorders estimated. Radiographic examination of the caudal vertebrae was performed in 595 client-owned dogs undergoing routine radiological examination of the hip joints. Anomalies of the caudal vertebrae were divided into four groups: (1) congenital anomalies with aberrant fusion of the vertebral epiphysis, like hemivertebra, block or transitional vertebra; (2) morphological anomalies like vertebra curva and dysplastic vertebral epiphysis; (3) post-traumatic changes, represented by fracture or luxation; and (4) degenerative changes, represented by spondylosis, osteophyte formation and mineralisation of intervertebral spaces. A total of 303 caudal vertebral anomalies were detected in 259 dogs (43.53% of all dogs examined). A single type of vertebral anomaly was present in 215 dogs and a combination of two pathologies was recorded in 44 dogs, which represents 83.01% and 16.99%, respectively, of the affected dogs. Congenital anomaly was present in 11.09% of the dogs examined. Sacrocaudal transitional vertebra was the most common congenital anomaly, present in 12.74% of the affected dogs. Radiographic examination of the caudal vertebrae is recommended as an essential part of any screening process to confirm the presence of congenital anomaly as a cause of kinked tail.
A 4-month-old, 20 kg, intact male, cane corso dog was presented with a slowly growing subcutaneous lesion on the left caudoventral abdominal wall. Ultrasound and computed tomography angiography revealed a subcutaneous plexus of aberrant tortuous vessels directly connected with the superficial branch of the deep circumflex iliac artery and vein. The arteriovenous malformation (AVM) was successfully surgically removed. Early recognition and surgical removal of AVM can have excellent cosmetic results and prevents potential cardiovascular complications.

Vertebral column pathologies requiring surgical intervention have been described in pet ferrets, however little information is available on the normal vertebral formula and congenital variants in this species. The purpose of this retrospective study was to describe vertebral formulas and prevalence of congenital vertebral anomalies in a sample of pet ferrets. Radiographs of 172 pet ferrets (96 males and 76 females) were included in this retrospective study. In 143 ferrets (83.14%), five different formulas of the vertebral column were recorded with normal morphology of vertebrae (rib attachment included) but with a variable number of thoracic (Th), lumbar (L), and sacral (S) vertebrae. The number of cervical (C) vertebrae was constant in all examined animals. Observed vertebral formulas were C7/Th14/L6/S3 (51.74%), C7/Th14/L6/S4 (22.10%), C7/Th14/L7/S3 (6.98%), C7/Th15/L6/S3 (1.74%), and C7/Th15/L6/S4 (0.58%). Formula C7/Th14/L6/S4 was significantly more common in males than in females (P < 0.05). Congenital spinal abnormalities were found in 29 ferrets (16.86%), mostly localized in the thoracolumbar and lumbosacral regions. The cervical region was affected in only one case. Transitional vertebrae represented the most common congenital abnormalities (26 ferrets) in the thoracolumbar (13 ferrets) and lumbosacral regions (10 ferrets) or simultaneously in both regions (three ferrets). Other vertebral anomalies included block (two ferrets) and wedge vertebra (one ferret). Spina bifida was not detected. Findings from the current study indicated that vertebral formulas may vary in ferrets and congenital abnormalities are common. This should be taken into consideration for surgical planning.

CÍL: Kostní defekty jsou problematikou zejména traumatologie, ortopedie a onkologie. Tradiční metodou léčby je autospongioplastika, v popředí zájmu je dnes použití resorbovatelných materiálů s osteoindukčními vlastnostmi (trikalciumfosfát - ChronOS). Cílem předkládané studie je ověření použití samotného trikalciumfosfátu v léčbě velkého diafyzárního defektu na modelu miniaturního prasete. Pozitivní výsledek by mohl omezit nutnost provádění spongioplastiky a zjednodušit celý proces léčby. Získané výsledky budou také sloužit jako technologická východiska pro studium možnosti náhrady kostních defektů mezenchymovými kmenovými buňkami. MATERIÁL A METODIKA: Experimentální skupinu tvořilo 25 miniaturních prasat z certifikovaného chovu Ústavu živočišné fyziologie a genetiky Akademie věd. U 12 prasat byl použit k náhradě defektu Trikaciumfosfát a u 13 prasat autogenní spongiózní štěp. Do vytvořeného diafyzárního defektu byl vložen Trikalciufosfát či autoštěp a zafixován. V obou skupinách byla fixace provedena LCP dlahou a intramedulárně Kirschnerovým drátem. Po zhojení byla tato oblast histologicky zpracována a hodnocena stran tvorby nové kostní tkáně a přihojení k původní kosti v místě okrajů defektu. VÝSLEDKY: Z výsledků vyplývá, že ve skupině s použitím autoštěpů došlu k výraznější novotvorbě zralé kostní tkáně. Při hodnocení okrajů defektu stran přihojení k původní kosti byly výsledky u skupiny s autoštěpem výrazně lepší. ZÁVĚR: Očekávané osteoindukční vlastnosti samotného trikalciumfosfátu se u rozsáhlého defektu diafýzy stehenní kosti v provedeném experimentu nepotvrdily. Náhradu autospongioplastiky jinou neinvazivní metodou v léčbě velkých kostních defektů je třeba nadále experimentálně hledat. Východiskem může být použitá operační metodika a využití vhodného biologického nosiče poskytujícího mechanickou oporu v kombinaci s trikalciumfosfátem nebo mezenchymovými kmenovými buňkami.
AIM: Bone defects are, in particular, the problematics of traumatology, orthopedics and oncology. The traditional method of treatment is autospongioplastics, nowadays the use of absorbable materials with osteoinductive properties (tricalcium phosphate - ChronOS)1,2 is in the forefront. The aim of the submitted study is to verify the application of tricalcium phosphate itself as a treatment of a large diaphyseal bone defect in a model of a miniature pig. A positive result could reduce the necessity of bonegrafting and thus simplify the whole process of treatment. The results obtained will serve as technological starting points for further studies regarding the possibility of replacement of bone defects by mesenchymal stem cells. MATERIALS AND METHODS: The experimental group was made up of 25 miniature pigs from the certified breeding of the Institute of Animal Physiology and Genetics Academy of Sciences. In 12 pigs the tricalcium phosphate as a replacement of the defect was used and in 13 pigs autogennous cancellous graft was used. The tricalcium phosphate, or autograft, was applied and fixed into the diaphyseal bone defect, which was formed. In both groups the fixation was carried out by means of LCP and application of intramedullary Kirschner wire. After healing, this area was processed from the histological point of view and evaluated with respect to the new formation of bone tissue and engraftment to the original bone, in the area of the edges of the defect. RESULTS: The results demonstrate that in the group with the application of autografts, more distinctive new formation of the mature bone tissue occurred. While evaluating the edges of the defect with regard to the engraftment to the original bone, the results in the group with the application of autograft were distinctively better. CONCLUSION: In the experiment conducted, the expected osteoinductive properties of the tricalcium phosphate itself have not been confirmed for the extensive defect of the femoral diaphysis. It is necessary to go on searching for the replacement of autospongioplastics by another noninvasive method for the treatment of extensive bone defects, by means of conducting experiments. The starting point might be the applied surgery method and the use of an appropriate biological carrier that will provide a mechanical support in combination with the tricalcium phosphate or mesenchymal stem cells.
OBJECTIVE: To assess the effects of intravenous (IV) medetomidine-butorphanol and IV dexmedetomidine-butorphanol on intraocular pressure (IOP). STUDY DESIGN: Prospective, randomized, blinded clinical study. ANIMALS: Forty healthy dogs. Mean ± SD body mass 37.6 ± 6.6 kg and age 1.9 ± 1.3 years. METHODS: Dogs were allocated randomly to receive an IV combination of dexmedetomidine, 0.3 mg m(-2), combined with butorphanol, 6 mg m(-2), (group DEX) or medetomidine 0.3 mg m(-2), combined with butorphanol 6 mg m(-2), (group MED). IOP and pulse (PR) and respiratory (f(R) ) rates were measured prior to (baseline) and at 10 (T10), 20 (T20), 30 (T30) and 40 (T40) minutes after drug administration. Oxygen saturation of hemoglobin (SpO(2)) was monitored following sedation. Data were analyzed by anova followed by Dunnett's tests for multiple comparisons. Changes were considered significant when p < 0.05. RESULTS: Following drug administration, PR and f(R) were decreased significantly at all time points but did not differ significantly between groups. Baseline IOP in mmHg was 14 ± 2 for DEX and 13 ± 2 for MED. With both treatments, at T10, IOP increased significantly (p < 0.001), reaching 20 ± 3 and 17 ± 2 for DEX and MED respectively. This value for DEX was significantly higher than for MED. There were no significant differences in IOP values between groups at any other time points. At T30 and T40, IOP in both groups was below baseline (DEX, 12 ± 2 and 11 ± 2: MED 12 ± 2 and 11 ± 2) and this was statistically significant, for DEX. CONCLUSIONS AND CLINICAL RELEVANCE: At the documented doses, both sedative combinations induced a transient increase and subsequent decrease of IOP relative to baseline, which must be taken into consideration when planning sedation of animals in which marked changes in IOP would be undesirable.

OBJECTIVE: To quantify humeroulnar incongruity on elbow radiographs in Labrador Retrievers with or without medial coronoid disease (MCD). STUDY DESIGN: Retrospective study of 92 elbows. SAMPLE POPULATION: Radiographic projections of elbow joints from Labrador Retrievers with MCD (n = 42 elbows; 26 dogs) and without MCD (n = 50 elbows; 25 dogs). PROCEDURE: The congruity of the humeroulnar joint was measured using an index of subluxation (SI) for each elbow. SI was defined as the distance between the centers of 2 circles drawn along the margins of the incisura trochlearis and the trochlea of humerus on mediolateral digital radiographic projections, normalized by the radius of the circle circumscribing the humeral trochlea. SI was compared between right and left elbows with and without pathology using a Wilcoxon test for paired data, and between normal and abnormal groups with a Wilcoxon test for unpaired data. Mismatch between ulnar curvature and curvature of humeral trochlea and radioulnar incongruency were also noted (Wilcoxon test). The intraobserver repeatability, correlation between SI and radioulnar incongruency, and between SI and mismatch elbow curvature were estimated with a Pearson's correlation coefficient. RESULTS: Intraobserver repeatability of SI measurement was high (r = 0.97). Mean ± SD humeroulnar incongruity (SI) was greater in elbows with MCD (18.5 ± 6.6) than in the normal elbows (1.7 ± 2.0, P < 0.001). The difference between the diameters of the curvatures of the ulnar and humeral trochlea was greater in elbows with MCD (12.5 ± 4.4) than in the normal group (10.7 ± 4.1, P < 0.05). A moderate correlation was found between the degree of humeroulnar incongruity and a radioulnar step (r = 0.63); however, no correlation was identified between SI and the difference between the diameters of the curvatures of the ulnar and humeral trochleae (r = 0.14). CONCLUSION: We propose a radiographic index to measure humeroulnar incongruity on mediolateral digital radiographic projections. This index (SI) supports the presence of humeroulnar incongruity in Labrador Retrievers with MCD. Further evaluation of its reproducibility and clinical importance are warranted. Although there is a moderate correlation between humeroulnar incongruity and radioulnar incongruency, causation has not been established.
