NRF2 is a master regulator of the cellular protection against oxidative damage in mammals and of multiple pathways relevant in the mammalian aging process. In the epidermis of the skin NRF2 contributes additionally to the formation of an antioxidant barrier to protect from environmental insults and is involved in the differentiation process of keratinocytes. In chronological aging of skin, the capacity for antioxidant responses and the ability to restore homeostasis after damage are impaired. Surprisingly, in absence of extrinsic stressors, NRF2 deficient mice do not show any obvious skin phenotype, not even at old age. We investigated the differences in chronological epidermal aging of wild type and NRF2-deficient mice to identify the changes in aged epidermis that may compensate for absence of this important transcriptional regulator. While both genotypes showed elevated epidermal senescence markers (increased Lysophospholipids, decreased LaminB1 expression), the aged NRF2 deficient mice displayed disturbed epidermal differentiation manifested in irregular keratin 10 and loricrin expression. The tail skin displayed less age-related epidermal thinning and a less pronounced decline in proliferating basal epidermal cells compared to the wildtype controls. The stratum corneum lipid composition also differed, as we observed elevated production of barrier protective linoleic acid (C18:2) and reduced abundance of longer chain saturated lignoceric acid (C24:0) among the stratum corneum fatty acids in the aged NRF2-deficient mice. Thus, despite epidermal differentiation being disturbed in aged NRF2-deficient animals in homeostasis, adaptations in keratinocyte proliferation and barrier lipid synthesis could explain the lack of a more severe phenotype.

In accordance with the 3 Rs principle (to replace, reduce and refine) animal models in biomedical research, we have developed and applied a new approach for sampling and analyzing hair follicles in various experimental settings. This involves use of a convenient device for non-invasive collection of hair follicles and processing methods that provide sufficient amounts of biological material to replace stressful and painful biopsies. Moreover, the main components of hair follicles are live cells of epithelial origin, which are highly relevant for most types of malignant tumors, so they provide opportunities for studying aging-related pathologies including cancer. Here, we report the successful use of the method to obtain mouse hair follicular cells for genotyping, quantitative PCR, and quantitative immunofluorescence. We present proof of concept data demonstrating its utility for routine genotyping and monitoring changes in quality and expression levels of selected proteins in mice after gamma irradiation and during natural or experimentally induced aging. We also performed pilot translation of animal experiments to human hair follicles irradiated ex vivo. Our results highlight the value of hair follicles as biological material for convenient in vivo sampling and processing in both translational research and routine applications, with a broad range of ethical and logistic advantages over currently used biopsy-based approaches.

Digit ratio is a morphological feature regarded as a biomarker of the balance of sex hormones during early development. The exposure of embryos to a set of sex hormones and the mutual relations between those hormones cause the emergence of individual morphological and/or behavioural characteristics as well as differences between sexes. We have thus hypothesised that differences in one of these morphological traits-digit ratio-may be a proxy representing a tendency towards tail autotomy. The aim of this study is to investigate the digit ratio (2D:3D, 2D:4D, 3D:4D) of the sand lizard, Lacerta agilis, Lacertidae, a species characterised by well-developed sexual dimorphism, whereby females are larger than males. We also tested associations between patterns in digit ratio and caudal autotomy, a common defensive mechanism among lizards. To our knowledge, the relationship between a tendency towards autotomy and digit ratio pattern has never been researched. To date, studies on autotomy have mainly focused on the consequences, costs or evolutionary background of tail loss. Hence, researchers examined mostly the frequency of autotomy in the context of predatory pressure or habitat conditions, omitting an individual's behavioural tendency to shed its tail. However, behavioural traits can affect an individual's exposure to predator attack and consequently the need to use an anti-predator strategy. Thus, following this logic, dropping the tail may be the result of the lizard's intraspecific personality characteristics, resulting from the effect of hormones on behaviour or innate traits. Therefore, we suggest that the inclusion of autotomy as a factor explaining observed digit ratio patterns and their variability between taxa has great potential. We used computerised measurements of photographed limbs to determine the length of digits. We found that the digit ratios for all four limbs were significantly lower in females than in males, excluding the 3D:4D ratio for the right hindlimbs. Therefore, the results confirmed the pattern already observed for most lizards. The novel element in our study is the detection of the relationship between a tendency towards caudal autotomy and digit ratio. Individuals with a tendency towards autotomy have a higher 2D:4D ratio in the right forelimbs and a lower 2D:3D ratio in the right hindlimbs. Obtained results suggest that these morphological characteristics are most likely related to intraspecific differences (between bold and shy individuals) which consequently may determine an individual's reaction or susceptibility to be a prey and escape behaviour. Thus, our results are probably the first attempt to link digit ratio to the susceptibility of lizards to tail autotomy.

Tail autotomy is a crucial antipredatory lizard response, which greatly increases individual survival, but at the same time also compromises locomotor performance, sacrifices energy stores and induces a higher burden due to the ensuing response of regenerating the lost body part. The potential costs of tail autotomy include shifts in energy allocation and metabolic rates, especially in juveniles, which invest their energy primarily in somatic growth. We compared the metabolic rates and followed the growth of juvenile males with and without regenerating tails in the Madagascar ground gecko (Paroedura picta), a nocturnal ground-dwelling lizard. Geckos with intact tails and those that were regrowing them grew in snout-vent-length at similar rates for 22weeks after autotomy. Tail regeneration had a negligible influence on body mass-corrected metabolic rate measured at regular intervals throughout the regenerative process. We conclude that fast-growing juveniles under the conditions of unrestricted food can largely compensate for costs of tail loss and regeneration in their somatic growth without a significant impact on the total individual body mass-corrected metabolic rate.

A prospective study to create radiographic classification of caudal vertebral anomalies in dogs was performed and the prevalence of the disorders estimated. Radiographic examination of the caudal vertebrae was performed in 595 client-owned dogs undergoing routine radiological examination of the hip joints. Anomalies of the caudal vertebrae were divided into four groups: (1) congenital anomalies with aberrant fusion of the vertebral epiphysis, like hemivertebra, block or transitional vertebra; (2) morphological anomalies like vertebra curva and dysplastic vertebral epiphysis; (3) post-traumatic changes, represented by fracture or luxation; and (4) degenerative changes, represented by spondylosis, osteophyte formation and mineralisation of intervertebral spaces. A total of 303 caudal vertebral anomalies were detected in 259 dogs (43.53% of all dogs examined). A single type of vertebral anomaly was present in 215 dogs and a combination of two pathologies was recorded in 44 dogs, which represents 83.01% and 16.99%, respectively, of the affected dogs. Congenital anomaly was present in 11.09% of the dogs examined. Sacrocaudal transitional vertebra was the most common congenital anomaly, present in 12.74% of the affected dogs. Radiographic examination of the caudal vertebrae is recommended as an essential part of any screening process to confirm the presence of congenital anomaly as a cause of kinked tail.
Recently, it has been found that spontaneous mutation Lx (polydactyly-luxate syndrome) in the rat is determined by deletion of a conserved intronic sequence of the Plzf (Promyelocytic leukemia zinc finger protein) gene. In addition, Plzf is a prominent candidate gene for quantitative trait loci (QTLs) associated with cardiac hypertrophy and fibrosis in the spontaneously hypertensive rat (SHR). In the current study, we tested the effects of Plzf gene targeting in the SHR using TALENs (transcription activator-like effector nucleases). SHR ova were microinjected with constructs pTAL438/439 coding for a sequence-specific endonuclease that binds to target sequence in the first coding exon of the Plzf gene. Out of 43 animals born after microinjection, we detected a single male founder. Sequence analysis revealed a deletion of G that resulted in frame shift mutation starting in codon 31 and causing a premature stop codon at position of amino acid 58. The Plzftm1Ipcv allele is semi-lethal since approximately 95% of newborn homozygous animals died perinatally. All homozygous animals exhibited manifestations of a caudal regression syndrome including tail anomalies and serious size reduction and deformities of long bones, and oligo- or polydactyly on the hindlimbs. The heterozygous animals only exhibited the tail anomalies. Impaired development of the urinary tract was also revealed: one homozygous and one heterozygous rat exhibited a vesico-ureteric reflux with enormous dilatation of ureters and renal pelvis. In the homozygote, this was combined with a hypoplastic kidney. These results provide evidence for the important role of Plzf gene during development of the caudal part of a body-column vertebrae, hindlimbs and urinary system in the rat.

Several taxonomic studies of the entomopathogenic nematode family Steinernematidae have reported on the minute tail projections on the tail tips of females of the first generation; however, the occurrence and morphology of these traits in other steinernematid species and their taxonomic significance are still unclear. In the present study, the tail projections in females of seven steinernematid species belonging to four phylogenetic clades were examined. The projections are present in all of the tested species and their number and shape is consistent within species but significantly differs among species. It thus seems that these structures may have taxonomic significance at the species level. Based on these results, we suggest that every description of a new species in the family Steinernematidae should contain a precise morphology of the female tail, focused on these projections, using both light and scanning electron microscopy.

BACKGROUND: Many animals rely on their escape performance during predator encounters. Because of its dependence on body size and temperature, escape velocity is fully characterized by three measures, absolute value, size-corrected value, and its response to temperature (thermal sensitivity). The primary target of the selection imposed by predators is poorly understood. We examined predator (dragonfly larva)-imposed selection on prey (newt larvae) body size and characteristics of escape velocity using replicated and controlled predation experiments under seminatural conditions. Specifically, because these species experience a wide range of temperatures throughout their larval phases, we predict that larvae achieving high swimming velocities across temperatures will have a selective advantage over more thermally sensitive individuals. RESULTS: Nonzero selection differentials indicated that predators selected for prey body size and both absolute and size-corrected maximum swimming velocity. Comparison of selection differentials with control confirmed selection only on body size, i.e., dragonfly larvae preferably preyed on small newt larvae. Maximum swimming velocity and its thermal sensitivity showed low group repeatability, which contributed to non-detectable selection on both characteristics of escape performance. CONCLUSIONS: In the newt-dragonfly larvae interaction, body size plays a more important role than maximum values and thermal sensitivity of swimming velocity during predator escape. This corroborates the general importance of body size in predator-prey interactions. The absence of an appropriate control in predation experiments may lead to potentially misleading conclusions about the primary target of predator-imposed selection. Insights from predation experiments contribute to our understanding of the link between performance and fitness, and further improve mechanistic models of predator-prey interactions and food web dynamics.

The effects of blocking ventromedial hypothalamic nucleus (VMH) muscarinic cholinoceptors on cardiovascular responses were investigated in running rats. Animals were anesthetized with pentobarbital sodium and fitted with bilateral cannulae into the VMH. After recovering from surgery, the rats were familiarized to running on a treadmill. The animals then had a polyethylene catheter implanted into the left carotid artery to measure blood pressure. Tail skin temperature (Ttail), heart rate, and systolic, diastolic and mean arterial pressure were measured after bilateral injections of 0.2 µl of 5 × 10-9 mol methylatropine or 0.15 M NaCl solution into the hypothalamus. Cholinergic blockade of the VMH reduced time to fatigue by 31% and modified the temporal profile of cardiovascular and Ttail adjustments without altering their maximal responses. Mean arterial pressure peak was achieved earlier in methylatropine-treated rats, which also showed a 2-min delay in induction of tail skin vasodilation, suggesting a higher sympathetic tonus to peripheral vessels. In conclusion, muscarinic cholinoceptors within the VMH are involved in a neuronal pathway that controls exercise-induced cardiovascular adjustments. Furthermore, blocking of cholinergic transmission increases sympathetic outflow during the initial minutes of exercise, and this higher sympathetic activity may be responsible for the decreased performance.
