Septins, a family of GTP-binding proteins that assemble into higher order structures, interface with the membrane, actin filaments and microtubules, and are thus important regulators of cytoarchitecture. Septin 9 (SEPT9), which is frequently overexpressed in tumors and mutated in hereditary neuralgic amyotrophy (HNA), mediates the binding of septins to microtubules, but the molecular determinants of this interaction remained uncertain. We demonstrate that a short microtubule-associated protein (MAP)-like motif unique to SEPT9 isoform 1 (SEPT9_i1) drives septin octamer-microtubule interaction in cells and in vitro reconstitutions. Septin-microtubule association requires polymerizable septin octamers harboring SEPT9_i1. Although outside of the MAP-like motif, HNA mutations abrogate this association, identifying a putative regulatory domain. Removal of this domain from SEPT9_i1 sequesters septins on microtubules, promotes microtubule stability and alters actomyosin fiber distribution and tension. Thus, we identify key molecular determinants and potential regulatory roles of septin-microtubule interaction, paving the way to deciphering the mechanisms underlying septin-associated pathologies. The obligate intracellular pathogen Anaplasma phagocytophilum infects vertebrate and tick hosts. In this study, a genome-wide search for cytoskeleton components was performed in the tick vector, Ixodes scapularis. The available transcriptomics and proteomics data was then used to characterize the mRNA and protein levels of I. scapularis cytoskeleton components in response to A. phagocytophilum infection. The results showed that cytoskeleton components described in other model organisms were present in the I. scapularis genome. One type of intermediate filaments (lamin), a family of septins that was recently implicated in the cellular response to intracellular pathogens, and several members of motor proteins (kinesins and dyneins) that could be implicated in the cytoplasmic movements of A. phagocytophilum were found. The results showed that levels of tubulin, actin, septin, actin-related proteins and motor proteins were affected by A. phagocytophilum, probably to facilitate infection in I. scapularis. Functional studies demonstrated a role for selected cytoskeleton components in pathogen infection. These results provided a more comprehensive view of the cytoskeletal components involved in the response to A. phagocytophilum infection in ticks.

Rakovina tlustého střeva a konečníku zůstává přes pokrok v diagnostice i terapii nadále celospolečenským problémem. Některá nenaplněná očekávání screeningových programů vedou ke zkoumání možností využití stávajících poznatků molekulární biologie k časné diagnostice. Zprávy o možnostech testování epigenetických známek nositelů rakoviny tlustého střeva nás vedly ke studii proveditelnosti testu Septin 9 pomocí Epi proColon Plasma Quick kit na skupině 108 pacientů podstupujících kolonoskopii, která byla referenční metodou. Zajímala nás senzitivita a specificita testu v našich klinických podmínkách. V souboru 108 osob byl kolonoskopicky zjištěn u 41 osob adenom, u 21 osob karcinom a 46 osob mělo negativní kolonoskopický nález. V kategorii osob s negativním kolonoskopickým nálezem byl Septin 9 u 34 negativní a u dvou pozitivní, u 10 osob byl test hodnocen pro nízkou koncentraci DNA v izobátu jako nezjištěno (ND – non detected). V kategorii pacientů s prokázaným adenomem byl Septin 9 ve 26 případech negativní a v jednom pozitivní a u 14 osob byl výsledek ND. Ve skupině pacientů s karcinomem byl Septin 9 negativní u sedmi osob, pozitivní u sedmi osob, u dalších sedmi osob byl test hodnocen jako ND. Senzitivita testu Septin 9 pro predikci karcinomu nebo adenomu byla 19,5 % a specificita 94,4 %. Pozitivita testu Septin 9 byla zjištěna signifikantně častěji u osob s karcinomem než u osob s adenomem nebo s negativním nálezem při kolonoskopii (p = 0,003). Septin 9 jako test prokázal vysokou specificitu (94,4 %). Zklamáním byla velmi nízká senzitivita u osob s adenomem v testovaném souboru (jen 3,7 %). Celkové výsledky studie jsou ve velké míře ovlivněny testy, ve kterých byla koncentrace DNA v izolátu vyhodnocena jako nedostatečná a výsledek byl stanoven jako ND.
Colorectal cancer still persists despite advances in diagnosis and therapy. The unfulfilled expectations of screening programs has led to the development of new tests based on molecular biology for early diagnosis. Reports over the past 15 years concerning the possibility of using epigenetic markers of colon cancer for colon cancer diagnosis led us to study the feasibility of using the Septin 9 test comprised in the Epi proColon Plasma Quick Kit on a group of 108 patients undergoing colonoscopy, the reference method. We were interested in the sensitivity and specificity of the test under our clinical conditions. Among the 108 patients, colonoscopy detected adenomas in 41, carcinomas in 21, and no findings in 46 persons. In the patients with negative colonoscopic findings, the Septin 9 test was negative for 34 and positive for two, with 10 people were classified as “not detected” (ND) because the DNA concentration in the isobath was below the required level. In patients with a confirmed adenoma, the Septin 9 test was negative in 26 and positive in one, and 14 people were classified as ND. In patients with cancer, the Septin 9 test was negative in seven and positive in seven, and seven were classified as ND. The sensitivity of the Septin 9 assay was 19.5% and the specificity was 94.4%. Septin 9 test positivity was found significantly more often in people with cancer than in those with an adenoma or with negative findings on colonoscopy (p = 0.003). The Septin 9 test showed a high degree of specificity (94.4%). It was disappointing that the test showed very low sensitivity in patients with adenoma (only 3.7%). The overall results of the study were affected by the high proportion of tests in which the concentration of DNA in the isolate was insufficient, necessitating that these patients be classified as ND.

Kolorektální karcinom představuje svou vysokou incidencí a mortalitou veliký medicínský problém. Přitom je prevencí velmi dobře ovlivnitelný. Je nutné detekovat časná stadia tohoto nádoru ke snížení úmrtnosti na tuto neoplazii. O to se snaží screeningové programy. Ty se opírají hlavně o záchyt skrytého krvácení pomocí různých laboratorních technik, sigmoideoskopii a hlavně kolonoskopii. Nicméně screeningové programy zatím nedosahují požadované účinnosti. Proto se vyvíjejí nové techniky, jako jsou např. detekce krevních biomarkerů. Do této skupiny patří detekce SEPT9 v krvi. Pomocí tohoto testu jsme vyšetřili 57 pacientů. Rozdělili jsme skupinu do 2 částí, v jedné bylo 33 asymptomatických jedinců. V této skupině jsme zachytili jenom 1 SEPT9 pozitivní výsledek, přičemž následné kolonoskopie byly vždy negativní. V další skupině bylo 24 prokázaných karcinomů a u 2 byl SEPT9 negativní výsledek. U dalších 22 pacientů byl výsledek na SEPT9 pozitivní. Test se v budoucnu, po ověření jeho účinnosti dalšími studiemi, může použít zvláště u jedinců s malou compliencí. Pro nemocného totiž vyžaduje pouze rutinní krevní odběr.
Due to its high incidence and mortality rates, the colorectal carcinoma represents a crucial medical issue. However, when it is detected in early stage there is high rate of successful treatment. That's why, early stage cancer screening programmes were introduced into the clinical practice. They focus on the finding of hidden bleeding, using various laboratory techniques, sigmoidoscopy, and, primarily, colonoscopy. However, screening programmes have not yet reached the effect required. New techniques are therefore being developed, such as the detection of blood biomarkers. This group includes also methylated SEPT9 (mSEPT9) detection in blood. We applied this test on 57 patients; we divided the group into two parts. There were 33 asymptomatic individuals in the first group. In this group, we were got only one positive mSEPT9 result. The consequent colonoscopies were negative. The other group had 24 proven carcinomas. Of them, two had negative mSEPT9 results. The remaining in all 22 patients was tested mSEPT9 positive. After its efficiency is tested by further studies, this test may be used especially for patients with low compliance, as it only requires routine blood drawing.
