While there is substantial research on what people want in their romantic and sexual partners, much of this work focuses on WEIRD, youthful samples, fails to consider the role of undesirable characteristics (i.e., things people do not want in partners) at all, or in conjunction with desirable characteristics (i.e., things people do want in partners), and may be overly reliant on psychometric approaches to pivotal variables in mating psychology like mate value and sociosexuality. In a nationally representative (online) sample of 2280 people from Czechia (aged between 18 and 50 years old), we examined linear and quadratic age, education, and self-perceived mate value (desirability) effects on the desired levels in mate choice of eight undesirable and seven desirable characteristics in men and women in relation to ostensible metrics of mate value. Self-perceived mate value alone explained little variance (men 1%, women 2%), while all mate value and mating strategy indicators together explained little variance of mate preferences and aversions (men 3%, women 5%). Desirable characteristics were better explained by mate value than undesirable ones. Our results are in line with evolutionary predictions suggesting that women are more demanding. Also, more qualities to offer correlate with more expectations in a partner.

To explore the effects and underlying mechanisms of Mdivi-1 on three common clinical models of acute kidney injury (AKI). Three common AKI cell models were constructed, classified into the control group (human renal tubular epithelial cells [HK-2] cells), the Iohexol group (HK-2 cells treated with Iohexol), the Genta group (HK-2 cells treated with Gentamicin), and the Cis group (HK-2 cells treated with Cisplatin). To explore the optimal protective concentration of Mdivi-1 for each AKI cell model, the experimental design consisted of the following seven groups: the control group (HK-2 cells cultured in medium), three injury groups (HK-2 cells subjected to Iohexol, Gentamicin, or Cisplatin), and the corresponding protection groups (with a certain concentration of Mdivi-1 added to each injury group). Cellular survival and apoptosis, reactive oxygen species (ROS) levels, and the expression of recombinant Sirtuin 3 (SIRT3) in each group were measured. Mitochondrial fission and fusion dynamics in cells were observed under an electron microscope. To explore relevant pathways, the changes in relevant pathway proteins were analyzed through Western blotting. The half maximal inhibitory concentration (IC50) values were 150.06 mgI/ml at 6 h in the Iohexol group, 37.88 mg/ml at 24 h in the Gentamicin group, and 13.48 microM at 24 h in the Cisplatin group. Compared with the control group, the three injury groups showed increased cell apoptosis rates and higher expressions of apoptotic proteins in HK-2 cells, with an accompanying decrease in cell migration. After the addition of corresponding concentrations of Mdivi-1, the optimal concentrations were 3 μM in the Iohexo-3 group, 1 microM in the Genta-1 group, and 5 μM in the Cis-5 group, HK-2 cells showed the highest survival rate, reduced apoptosis, decreased mitochondrial ROS and SIRT3 expression, and reduced mitochondrial fission and autophagy when compared with each injury group. Further verification with Western blot analysis after the addition of Mdivi-1 revealed a reduction in the expressions of mitochondrial fission proteins DRP1, Nrf2, SIRT3, Caspase-3, Jun N-terminal Kinase (JNK)/P-JNK, NF-kappaB, Bcl2, and autophagic protein P62, as well as reduced ROS levels. Mdivi-1 had protective effects on the three common AKI cell models by potentially reducing mitochondrial fission in cells and inhibiting the production of ROS through the mediation of the NF- B/JNK/SIRT3 signaling pathway, thereby exerting protective effects. Východiská: V managemente pacientov s kolorektálnym karcinómom (colorectal cancer – CRC) stále existuje priestor pre zlepšenie stratifikácie rizika a tým presnejšie „ušitie“ liečby na mieru. Veľmi sľubným sa v tomto ohľade javia biomarkery získavané prostredníctvom takzvanej tekutej biopsie, čo je neinvazívna metóda odberu telesných tekutín pacienta, najčastejšie periférnej krvi. Analyzujú sa rozličné biomarkery súvisiace s nádorom, ktoré môžu mať ako prognostickú, tak aj prediktívnu hodnotu. Jedným z najviac prebádaných nádorových biomarkerov je práve nádorová cirkulujúca DNA, ktorej spektrum využitia bolo spočiatku len u pokročilých a metastatických karcinómov a spočívalo v molekulárnom profilovaní alebo zisťovaní získanej rezistencie k liečbe. V súčasnosti sa využitie cirkulujúcej nádorovej DNA (ctDNA) posunulo už k skorým štádiám karcinómov, kde okrem iného slúži k identifikácii minimálnej reziduálnej choroby alebo k skorej diagnostike CRC. Doterajšie štúdie ukazujú veľmi sľubný potenciál týchto biomarkerov, ale k využitiu v klinickej praxi bude potrebné získať viac informácií a počkať na výsledky prebiehajúcich výskumov. Cieľ: V tomto prehľadovom článku sa budeme venovať ctDNA, jej aspektom, možnostiam diagnostiky a súčasnému využitiu v rámci CRC.
Background: Space still exists in the management of patients with colorectal cancer (CRC) for improving risk stratification and thus the precision of treatment tailoring. Quite promising in this regard are biomarkers acquired via liquid biopsy, which is a non-invasive method of body fluid draw, most commonly peripheral blood. A variety of biomarkers associated with the tumor are analyzed, which can have either prognostic or predictive value. Circulating tumor DNA (ctDNA) is one of the most explored tumor biomarkers. Initially, its utility spectrum was only in advanced or metastatic cancers and consisted of molecular profiling and detecting acquired resistance to treatment. Nowadays, the use of circulating tumor DNA has shifted to earlier cancer stages, where it can identify minimal residual disease or diagnose colorectal cancer early. Existing studies show promising potential of these biomarkers, but more information needs to be gathered and information from ongoing studies needs to be obtained in order to use them in everyday practice. Aim: In this review article, we will discuss ctDNA, its aspects, diag- nostic possibilities and current use in CRC.

Fourteen diterpenes were isolated from methanol extracts of the aerial parts ofColeus comosus,Coleus forsteri "Marginatus", and Plectranthus ciliatus. The compounds belong to the abietane (1-4, 9-11, and 13), ent-clerodane (5-8), and ent-kaurane (14, 15) classes. Three new compounds were isolated from C. comosus, including 3-O-acetylornatin G (2), 3,12-di-O-acetylornatin G (3), ornatin B methyl ester (5), and ornatin F (4), for which we proposed a revised structure. The structures of the compounds were determined by comprehensive spectroscopic data analysis. The isolated diterpenes were examined in silico for their physicochemical and early ADME properties. Their antiproliferative effects were determined in vitro using human breast (MDA-MB-231 and MCF-7), cervical (HeLa), and glioblastoma (U-87 MG) cancer cell lines. The royleanone- and hydroquinone-type abietane diterpenes (9-13)exhibited the most potent antiproliferative activity against all cancer cell lines tested, particularly against glioblastoma cells, with IC50 values ranging from 1.1 to 15.6 μM.

Rivalita triatlonistov a snaha posúvať svoje hranice výkonnosti ich núti premyslieť celú svoju prípravu do detailov, aby využívali čas efektívne, pripravili svoje telo nielen na maximálny výkon, ale aj analyzovali výskyt možných zranení. Z daného dôvodu je preto nutné zaradiť diagnostiku v triatlone do tréningového procesu, ktorá slúži pre triatlonistu a trénera ako spätná väzba toho, ako trénujú a na akej výkonnostnej úrovni sú. Na základe týchto poznatkov sme sa rozhodli verifikovať vplyv silovo-vytrvalostného tréningu na zmeny v skóre funkčnej pohybovej diagnostiky u triatlonistov. Vo výskume bolo zahrnutých 8 výkonnostných triatlonistov z triatlonového klubu TRIAN ŠK UMB Banská Bystrica, ktorých vek bol 17,26 ± 0,59 rokov s telesnou výškou 175,8 ± 7,4 cm a telesnou hmotnosťou 63,2 ± 5,8 kg. Na získanie informácií o triatlonistoch sme použili funkčnú pohybovú diagnostiku (Functional movement screen – FMS), ktorú vykonávali bez rozohriatia a rozcvičenia, aby neboli ovplyvnené nadobudnuté hodnoty. Na zabezpečenie objektivity testovania boli oslovení 3 externí examinátori, ktorí sú certifikovaní odborníci funkčnej pohybovej diagnostiky. Zaznamenávanie údajov sme vykonali ručne a boli zapísané do „Functional Movement Screen Score Sheet“. Vstupné a výstupné testovanie prebiehalo v doobedných hodinách za tých istých podmienok. Tréningový program prebiehal 8 týždňov v prípravnom období od 16. 01. 2024 do 07. 03. 2024 a realizovali sme ho 2-krát v týždni. Pri porovnaní vstupných a výstupných údajov zaznamenal experimentálny súbor výraznejšie zlep- šenie priemeru bodového skóre triatlonistov z 12,25 ± 2,17 bodov na 15,75 ± 1,92 bodov, čo predstavuje zlepšenie FMS skóre o 28,6 %, pričom kontrolný súbor zaregistroval taktiež priemerné zlepšenie, a to z 12,25 ± 1,92 na 13 ± 1,22 bodov, čo predstavuje zlepšenie o 6,1 %. Z výsledkov vyplýva, že silovo-vytrvalostný tréningový program bude mať vyšší efekt na zmeny v skóre funkčnej pohybovej diagnostike u triatlonistov ako metóda rovnomerného tréningu. Z hľadiska nižšieho počtu probandov, odporúčame širšie skúmanie danej problematiky.
The rivalry of triathletes and the effort to push their performance limits forces them to think through their entire preparation in detail, to use time efficiently, to prepare their bodies not only for maximum performance, but also to analyse the occurrence of possible injuries. For this reason, itis necessary to include triathlon diagnostics in the training process which serves as feedback for the triathlete and the coach on how they train and at what performance level they are. Based on this knowledge, the study verifies the impact of strength-endurance training on scorechanges of Functional Movement Screen of triathletes.The research included 8 performance triathletes from TRIAN ŠK UMB triathlon club Banská Bystrica whose age was 17.26 ± 0.59 years with a body height of 175.8 ± 7.4 cm and a body weight of 63.2 ± 5.8 kg. To obtain information about the triathletes, the Functional Movement Screen (FMS) was used and was performed without warm up, so that the obtained values were not affected. 3 external examiners who are certified experts in Functional Movement Screen were included to ensure the testing objectivity. Data recording was done manually and registered into the Functional Movement Screen Score Sheet. Input and output testing took place in the morning hours under the same conditions. The training program ran for 8 weeks in the preparatory period from 16/01/2024 to 07/03/2024 and was implemented 2 times a week.When comparing the input and output data, the experimental group recorded a significant improvement in the average point score of the triathletes from 12.25 ± 2.17 points to 15.75 ± 1.92 points which represents an improvement in the FMS score by 28.6%. The control group also registered an average improvement from 12.25 ± 1.92 to 13 ± 1.22 points which represents an improvement of 6.1%. The results show that a strength-endurance training program will have a higher effect on changes in Functional Movement Screen scores in triathletes than a uniform training method. Due to lower number of the triathletes the further and wider investigation of the issue is recommended.

Aging is a natural process of gradual decrease in physical and mental capacity. Biological age (accumulation of changes and damage) and chronological age (years lived) may differ. Biological age reflects the risk of various types of disease and death from any cause. We selected potential biomarkers of aging - telomerase, AGEs, GDF11 and 15 (growth differentiation factor 11/15), sirtuin 1, NAD+ (nicotinamide adenine dinucleotide), inflammasome NLRP3, DNA/RNA damage, and klotho to investigate changes in their levels depending on age and sex. We included 169 healthy volunteers and divided them into groups according to age (under 35; 35-50; over 50) and sex (male, female; male and female under 35; 35-50, over 50). Markers were analyzed using commercial ELISA kits. We found differences in values depending on age and gender. GDF15 increased with age (under 30 and 35-50 p < 0.002; 35-50 and over 50; p < 0.001; under 35 and over 50; p < 0.001) as well as GDF11 (35-50 and over 50; p < 0.03; under 35 and over 50; p < 0.02), AGEs (under 30 and 35-50; p < 0.005), NLRP3 (under 35 over 50; p < 0.03), sirtuin 1 (35-50 and over 50; p < 0.0001; under 35 and over 50; p < 0.004). AGEs and GDF11 differed between males and females. Correlations were identified between individual markers, markers and age, and markers and sex. Markers that reflect the progression of biological aging vary with age (GDF15, GDF11, AGEs, NLRP3, sirtuin) and sex (AGEs, GDF11). Their levels could be used in clinical practice, determining biological age, risk of age-related diseases and death of all-causes, and initiating or contraindicating a therapy in the elderly based on the patient's health status.

Mate value is an important concept in mate choice research although its operationalization and understanding are limited. Here, we reviewed and evaluated previously established conceptual and methodological approaches measuring mate value and presented original research using individual differences in how people view themselves as a face-valid proxy for mate value in long- and short-term contexts. In data from 41 nations (N = 3895, Mage = 24.71, 63% women, 47% single), we tested sex, age, and relationship status effects on self-perceived mate desirability, along with individual differences in the Dark Triad traits, life history strategies, peer-based comparison of desirability, and self-reported mating success. Both sexes indicated more short-term than long-term mate desirability; however, men reported more long-term mate desirability than women, whereas women reported more short-term mate desirability than men. Further, individuals who were in a committed relationship felt more desirable than those who were not. Concerning the cross-sectional stability of mate desirability across the lifespan, in men, short- and long-term desirability rose to the age of 40 and 50, respectively, and decreased afterward. In women, short-term desirability rose to the age of 38 and decreased afterward, whereas long-term desirability remained stable over time. Our results suggest that measuring long- and short-term self-perceived mate desirability reveals predictable correlates.

Lysine deacetylases, like histone deacetylases (HDACs) and sirtuins (SIRTs), are involved in many regulatory processes such as control of metabolic pathways, DNA repair, and stress responses. Besides robust deacetylase activity, sirtuin isoforms SIRT2 and SIRT3 also show demyristoylase activity. Interestingly, most of the inhibitors described so far for SIRT2 are not active if myristoylated substrates are used. Activity assays with myristoylated substrates are either complex because of coupling to enzymatic reactions or time-consuming because of discontinuous assay formats. Here we describe sirtuin substrates enabling direct recording of fluorescence changes in a continuous format. Fluorescence of the fatty acylated substrate is different when compared to the deacylated peptide product. Additionally, the dynamic range of the assay could be improved by the addition of bovine serum albumin, which binds the fatty acylated substrate and quenches its fluorescence. The main advantage of the developed activity assay is the native myristoyl residue at the lysine side chain avoiding artifacts resulting from the modified fatty acyl residues used so far for direct fluorescence-based assays. Due to the extraordinary kinetic constants of the new substrates (KM values in the low nM range, specificity constants between 175,000 and 697,000 M-1s-1) it was possible to reliably determine the IC50 and Ki values for different inhibitors in the presence of only 50 pM of SIRT2 using different microtiter plate formats.
Súhrn Úvod: Potravinová alergia je chronické ochorenie, ktoré si vyžaduje veľké úsilie a môže mať za následok nielen zníženú kvalitu života, ale aj zvýšený pocit úzkosti a stresu. Štúdia skúma súvislosť medzi potravinovou alergiou, kvalitou života (HRQL) a úrovňou úzkostlivosti u dospelých jedincov s potravinovou alergiou. Sekundárnym cieľom bolo overiť spoľahlivosť slovenskej verzie dotazníka FAQLQ-AF, ktorý bol použitý pre slovenskú populáciu po prvýkrát. Metódy: Výskumnú vzorku tvorilo 167 dospelých osôb s potravinovou alergiou vo veku 19-69 rokov (M vek = 37). Na meranie úzkostlivosti bol použitý dotazník STAI X2 a na meranie kvality života súvisiacej so zdravím bol použitý dotazník FAQLQ-AF, ktorý dosiahol vysoké hodnoty reliability. Výsledky: Vo výsledkoch sa preukázal mierny, pozitívny vzťah medzi úzkostlivosťou a HRQL meraný dotazníkom FAQLQ-AF. Jednoduchá lineárna regresná analýza preukázala, že úroveň črty úzkosti je prediktorom úrovne HRQL a nárast úrovne úzkostlivosti znižuje úroveň HRQL. Faktory ako tetanický syndróm, komorbidity, predpísané lieky a odporúčané voľnopredajné lieky sú spojené s úrovňou úzkostlivosti. Súvisiace ochorenia a zoskupenia symptómov súvisia s úrovňou HRQL. Závery: Dospelí jedinci s potravinovou alergiou, ktorí sú viac úzkostliví, majú vyššiu pravdepodobnosť horšej kvality života. Včasná intervencia a prevencia, optimalizácia manažmentu liečby by mali byť nutnou súčasťou psychologickej starostlivosti o pacientov s potravinovou alergiou.
Introduction: Food allergy is a chronic disease that requires a great deal of effort. It can result not only in a reduced quality of life, but also increased feelings of anxiety and stress. This study examines the association between health-related quality of life (HRQL) and trait anxiety levels in adults with food allergy. Methods: The research sample consisted of 167 adults with food allergy aged 19-69 years, arithmetic average (M) age 37. The STAI X2 questionnaire was used to measure trait anxiety. The food allergy quality of life questionnaire for adult population (FAQLQ-AF) was used to measure the health-related quality of life - and this achieved high reliability values. Results: A moderate, positive relationship was demonstrated between anxiety and the FAQLQ-AF. Simple linear regression analysis demonstrated that trait anxiety level is a predictor of HRQL level, and an increase in anxiety level decreases HRQL. Factors such as tetanus syndrome, comorbidities, prescribed medications and recommended over-the-counter medications are associated with trait anxiety levels. Related illnesses and symptom clusters are related to HRQL levels. Conclusions: Adults with food allergy who are more anxious are more likely to experience a poorer quality of life. Early intervention, prevention and optimization of treatment management should be a necessary part of the psychological care of patients with food allergy.
