Structures and formation pathways of compounds responsible for pink discoloration of onion and leek were studied. A procedure was developed for the isolation and purification of the color compounds from various model systems and their identification by HPLC-DAD-MS/MS. In total, structures of 15 major color compounds were tentatively determined. It was found that the pigment is a complex mixture of highly conjugated species composed of two N-substituted 3,4-dimethylpyrrole-derived rings linked by either a methine or a propenylidine bridge. These two-ring units are further modified by various C1- and C3-side chains. Experiments with isotope-labeled thiosulfinates revealed that the methine bridge and C1-side chains originate from the methyl group of methiin, whereas the C3 units are derived from the propenyl group of isoalliin.

Světlobuněčný akantom je vzácný benigní epidermální tumor neznámé etiologie. Vyskytuje se u pacientů středního věku, bez rozdílu pohlaví a s predilekcí na dolních končetinách. Klinicky se projevuje jako solitární pomalu rostoucí papulózní léze růžové až červené barvy. Popisujeme případ, kdy se dermatoskopie ukázala jako velmi užitečná pro diagnostiku léze.
Clear cell acanthoma is a rare benign epidermal tumor of unknown etiology. It occurs in middle aged patients, equally in men and women, with predilection on lower extremities. Clinically it manifests as a solitary slowly growing papular lesion of pink to red colour. We describe a case, in which dermatoscopy turned out to be very useful in the lesion diagnostics.

PURPOSE: The appropriate treatment of pulseless pink supracondylar humerus fractures (SCHF) remains controversial. In this study, the outcomes of two treatment approaches (with and without vascular surgery) were compared. MATERIAL AND METHODS: This was a retrospective multicenter study of patients with pulseless pink SCHFs treated in ten pediatric surgery, trauma, or orthopedics departments in the Czech and Slovak Republic between 2014 and 2018. RESULTS: Of the total 3608 cases of displaced SCHF, 125 had the pulseless pink SCHF. Of those, 91% (114/125) did not undergo vascular surgery and 9% (11/125) underwent vascular surgery. The patients who did undergo vascular surgery had radial artery pulsation restored more frequently in the operating room (73% vs. 36%; p = 0.02), within 6 h (91% vs. 45%; p = 0.004), and within 24 h of surgery (91% vs. 57%; p = 0.05). However, 72 h after surgery, there was no significant difference in palpable radial artery pulsation between the vascular surgery and the non-vascular surgery groups (91% vs. 74%; p = 0.24). Additionally, no significant differences in long-term neurological (9% vs. 22%; p = 0.46) or circulatory (9% vs. 7%; p = 0.57) deficits were found between the two groups. CONCLUSION: While vascular surgery in patients with pulseless pink SCHFs is associated with a more prompt restoration of radial artery pulsation, no statistical significant differences in terms of the restoration of neurological deficits or the risks of long-term neurological or circulatory deficits were found between patients with and without vascular surgery.
There has recently been increasing interest in neutral models of biodiversity and their ability to reproduce the patterns observed in nature, such as species abundance distributions. Here we investigate the ability of a neutral model to predict phenomena observed in single-population time series, a study complementary to most existing work that concentrates on snapshots in time of the whole community. We consider tests for density dependence, the dominant frequencies of population fluctuation (spectral density) and a relationship between the mean and variance of a fluctuating population (Taylor's power law). We simulated an archipelago model of a set of interconnected local communities with variable mortality rate, migration rate, speciation rate, size of local community and number of local communities. Our spectral analysis showed 'pink noise': a departure from a standard random walk dynamics in favor of the higher frequency fluctuations which is partly consistent with empirical data. We detected density dependence in local community time series but not in metacommunity time series. The slope of the Taylor's power law in the model was similar to the slopes observed in natural populations, but the fit to the power law was worse. Our observations of pink noise and density dependence can be attributed to the presence of an upper limit to community sizes and to the effect of migration which distorts temporal autocorrelation in local time series. We conclude that some of the phenomena observed in natural time series can emerge from neutral processes, as a result of random zero-sum birth, death and migration. This suggests the neutral model would be a parsimonious null model for future studies of time series data.
Text reflektuje skutečnost, že preimplantační genetická diagnostika poskytuje naději na zdravé dítě i takovým párům, u kterých více pokusů o zdravého potomka selhalo. V rámci tohoto vyšetření embryí se však dozvídáme také informaci o pohlaví. Tento fakt by neměl být zneužíván k realizaci přání o narození chlapce, nebo děvčete. Nechtěná, přestože zcela zdravá embrya jsou totiž likvidována.
The manuscript reflects the fact that pre-implantation genetic diagnosis provides hope for a healthy child even for couples who have failed several times to have a healthy descendant. However, as part of this examination of the embryos, we also learn information about the sex. This fact should not be abused to realize the wish for the birth of a boy or a girl. Although completely healthy, unwanted embryos are disposed.
Fifteen Amaryllidaceae alkaloids (1-15) of various structural types were isolated by standard chromatographic methods from fresh bulbs of Narcissus poeticus cv. Pink Parasol. The chemical structures were elucidated by MS, and 1D and 2D NMR spectroscopic analyses, and by comparison with literature data. Narcipavline (5) and narcikachnine (6) are reported here for the first time. In their structure are combined two basic structural types of Amaryllidaceae alkaloids (galanthamine- and galanthindole-structural types), which represent a new structural type of these compounds. Alkaloids isolated in sufficient amounts were evaluated for their human erythrocytic acetylcholinesterase, and human serum butyrylcholinesterase (HuBuChE) inhibition activity using Ellman's method. Z-Gly-Pro-p-nitroanilide was used as substrate in the prolyl oligopeptidase (POP) assay. Untested alkaloids were also screened for their cytotoxic activity against a small panel of human cancer cells, which spanned cell lines from different tissue types. In parallel, MRC-5 human fibroblasts were employed to determine overall toxicity against noncancerous cells. Some compounds were evaluated for their antiprotozoal activity. The newly isolated alkaloid narcipavline (5) showed interesting HuBuChE inhibition activity (IC50 = 24.4 ± 1.2 µM), and norlycoramine (11) demonstrated promising POP inhibition (IC50 = 0.21 ± 0.01 mM).

Amyloidóza je spektrum onemocnění z extracelulárního hromadění patologických fibrilárních proteinů ve tkáních, což vede k narušení mikroarchitektoniky postižených tkání, lokální toxicitě a aktivaci imunitní reakce. Je popsáno přes dvacet různých amyloidotvorných proteinů, nejčastěji však jde o amyloid lehkých řetězců (AL), nebo amyloid A (AA) v rámci primární, respektive sekundární systémové amyloidózy. Podstatně vzácnější je výskyt fokálních lézí bez systémového postižení. V této kazuistice předkládáme případ fokální amyloidózy s postižením laryngu, trachey a bronchu.
Amyloidosis is a spectrum of diseases caused by extracellular accumulation of pathological fibrillar proteins in the tissues, which leads to a disruption of tissue microarchitecture, local toxicity and activation of immune reaction. Over twenty different amyloidforming proteins have been described, the most frequent being the immunoglobulin light-chain amyloid (AL) or amyloid A (AA) in primary or secondary systemic amyloidosis, respectively. Focal lesions without systemic involvement are a much rarer occurrence. In this case report we present a case of focal amyloidosis involving the larynx, trachea and bronchus.

Oxidative stress may cause extended tyrosine posttranslational modifications of peptides and proteins. The 3-nitro-L-tyrosine (Nit), which is typically formed, affects protein behavior during neurodegenerative processes, such as Alzheimer's and Parkinson's diseases. Such metabolic products may be conveniently detected at very low concentrations by surface enhanced Raman spectroscopy (SERS). Previously, we have explored the SERS detection of the Nit NO2 bending vibrational bands in a presence of hydrogen chloride (Niederhafner et al., Amino Acids 53:517-532, 2021, ibid). In this article, we describe performance of a new SERS substrate, "pink silver", synthesized photochemically. It provides SERS even without the HCl induction, and the acid further decreases the detection limit about 9 times. Strong SERS bands were observed in the asymmetric (1550-1475 cm-1) and symmetric (1360-1290 cm-1) NO stretching in the NO2 group. The bending vibration was relatively weak, but appeared stronger when HCl was added. The band assignments were supported by density functional theory modeling.
