Bacteria have developed different intra- and inter-specific communication mechanisms that involve the production, release, and detection of signaling molecules, because these molecules serve as the autoinducers involved in "quorum sensing" systems. Other communication mechanisms employ volatile signaling molecules that regulate different bacterial processes. The Arthrobacter agilis strain UMCV2 is a plant growth promoting actinobacterium, which induces plant growth and inhibits phytopathogenic fungi by emitting the dimethylhexadecylamine (DMHDA). However, little is known about the effect of this volatile compound on A. agilis UMCV2 itself, as well as on other bacteria. By exposing A. agilis UMCV2 and bacteria of the genus Bacillus and Pseudomonas to different concentrations of DMHDA, this study showed the dose-dependent effects of DMHDA on A. agilis UMCV2 growth, cellular viability, swarming motility, and expression of marker genes of the flagellar apparatus of bacteria. DMHDA was found to also modulate swarming motility of Bacillus sp. ZAP018 and P. fluorescens UM270, but not that of P. aeruginosa PA01. These data indicate that DMHDA is involved in both intra- and inter-specific bacterial interaction.

Akutní infekce močových cest je v dospělé populaci velmi častým onemocněním. Platí zde, že mnohem větší léčebné potíže než infekce akutní, přinášejí chronické a recidivující bakteriální infekce. Tvorba biofilmu je významným faktorem, který umožňuje delší přežívání bakterií a také významně ztěžuje léčbu bakteriálních infekcí. Vzhledem k omezeným možnostem antibakteriální terapie je pochopitelné hledání dalších možností, jak pacientům účinně pomoci. Tyto možnosti již tradičně nabízí fytofarmaka, tedy rostlinné léčivé přípravky. Sdělení se věnuje možnostem registrovaných léčiv rostlinného původu v léčbě akutních infekcí močových cest.
Acute urinary tract infection is a very common disease in the adult population. It holds true that much more significant treatment difficulties are encountered in the case of chronic and recurrent bacterial infections than in acute ones. Biofilm formation is another important factor, allowing prolonged survival of bacteria and, thus, making treatment of bacterial infections more difficult. Given the limited options of antibacterial therapy, other possibilities of helping patients in an effective way are, of course, being sought. These traditionally include the use of phytopharmaceuticals, i.e. herbal medicinal products. The paper deals with the possible use of registered pharmaceutical products of plant origin in treating acute urinary tract infections.

Marine sponges, a well-documented prolific source of natural products, harbor highly diverse microbial communities. Their extracts were previously shown to contain quorum sensing (QS) signal molecules of the N-acyl homoserine lactone (AHL) type, known to orchestrate bacterial gene regulation. Some bacteria and eukaryotic organisms are known to produce molecules that can interfere with QS signaling, thus affecting microbial genetic regulation and function. In the present study, we established the production of both QS signal molecules as well as QS inhibitory (QSI) molecules in the sponge species Sarcotragus spinosulus. A total of eighteen saturated acyl chain AHLs were identified along with six unsaturated acyl chain AHLs. Bioassay-guided purification led to the isolation of two brominated metabolites with QSI activity. The structures of these compounds were elucidated by comparative spectral analysis of 1HNMR and HR-MS data and were identified as 3-bromo-4-methoxyphenethylamine (1) and 5,6-dibromo-N,N-dimethyltryptamine (2). The QSI activity of compounds 1 and 2 was evaluated using reporter gene assays for long- and short-chain AHL signals (Escherichia coli pSB1075 and E. coli pSB401, respectively). QSI activity was further confirmed by measuring dose-dependent inhibition of proteolytic activity and pyocyanin production in Pseudomonas aeruginosa PAO1. The obtained results show the coexistence of QS and QSI in S. spinosulus, a complex signal network that may mediate the orchestrated function of the microbiome within the sponge holobiont.

Microorganisms do not live alone, but rather they communicate using diverse „languages“. In general, each bacterial species produces and responds to a unique autoinducer signal. Gram-negative bacteria use N-acylated homoserine lactones and gram-positive bacteria use oligopeptides as autoinducers. Function of autoinducer 2 is bacterial interspecies cell-to-cell communication. The structure and function of two main signaling molecules (farnesol and tyrosol) in the C. albicans quorum sensing (QS) system were also already described. Signaling molecules control the behavior of the whole population (especially virulence factors expression). That’s why QS systems represent a new therapeutic target, especially because of an increasing worldwide antibiotic resistance. Quorum sensing inhibitors are a promising direction in the treatment of infection caused by pathogenic micro¬organisms.

Biofilmy oportunně patogenních mikroorganismů představují zdroj četných obtížně léčitelných onemocnění. Studium přírodních biologicky aktivních látek a jejich vlivu na tvorbu a stabilitu biofilmů hraje významnou roli v současném výzkumu. Přírodní produkty extrahované z nesčetného množství rostlin představují potenciální zdroj nových antimikrobiálních a antibiofilmových činidel. Ze zmíněné kategorie látek práce předkládá cinnamaldehyd a N‐‐acetylcystein.
The biofilms of opportunistic pathogens are a source of numerous difficult‐‐to‐‐treat infections. Exploration of natural biologically active compounds and their influence on the formation and stability of biofilms plays an important role in current research. Natural compounds isolated from plants can be viewed as a potential source of new anti‐‐microbial and anti‐‐biofilm agents. From this category, cinnamaldehyde and N‐‐acetylcysteine are presented.

Brominated furanone and epigallocatechin gallate (EGCG) are naturally occurring polyphenolic compounds that can be derived from sources such as Delisea pulchra algae and green tea, respectively. These compounds may have potential health benefits and antimicrobial properties. Biofilm formation and bacterial motility are virulence factors that seem to be involved in the autoinducer 2 (AI-2)-mediated quorum sensing (QS) response of Campylobacter. In this study, the anti-QS activities of 2(5H)-furanone, EGCG, and a citric-based disinfectant were tested against Campylobacter jejuni. The minimal bactericidal concentration (MBC) was determined by a microdilution method, and the AI-2 activity was measured by bioluminescence. For motility tests, subinhibitory concentrations of each compound were mixed with semisolid Muller Hinton agar. Biofilm formation was quantified in broth-containing microplates after staining with safranin. The MBC of tested compounds ranged from 0.3 to 310 μg/mL. Subinhibitory concentrations of all of the antimicrobial compounds significantly decreased (19 to 62 %) the bacterial motility and reduced biofilm formation. After treatment with EGCG, furanone, and the disinfectant, AI-2 activity was decreased by 60 to 99 % compared to control. In conclusion, 2(5H)-furanone, EGCG, and the disinfectant exert bactericidal effects against C. jejuni and disturb QS activity and reduce motility and biofilm formation. These compounds may be naturally occurring alternatives to control C. jejuni.

Quorum sensing (QS) regulates group behaviors of Candida albicans such as biofilm, hyphal growth, and virulence factors. The sesquiterpene alcohol farnesol, a QS molecule produced by C. albicans, is known to regulate the expression of virulence weapons of this fungus. Fluconazole (FCZ) is a broad-spectrum antifungal drug that is used for the treatment of C. albicans infections. While FCZ can be cytotoxic at high concentrations, our results show that at much lower concentrations, quercetin (QC), a dietary flavonoid isolated from an edible lichen (Usnea longissima), can be implemented as a sensitizing agent for FCZ-resistant C. albicans NBC099, enhancing the efficacy of FCZ. QC enhanced FCZ-mediated cell killing of NBC099 and also induced cell death. These experiments indicated that the combined application of both drugs was FCZ dose dependent rather than QC dose dependent. In addition, we found that QC strongly suppressed the production of virulence weapons-biofilm formation, hyphal development, phospholipase, proteinase, esterase, and hemolytic activity. Treatment with QC also increased FCZ-mediated cell death in NBC099 biofilms. Interestingly, we also found that QC enhances the anticandidal activity of FCZ by inducing apoptotic cell death. We have also established that this sensitization is reliant on the farnesol response generated by QC. Molecular docking studies also support this conclusion and suggest that QC can form hydrogen bonds with Gln969, Thr1105, Ser1108, Arg1109, Asn1110, and Gly1061 in the ATP binding pocket of adenylate cyclase. Thus, this QS-mediated combined sensitizer (QC)-anticandidal agent (FCZ) strategy may be a novel way to enhance the efficacy of FCZ-based therapy of C. albicans infections.

Infekce kloubních implantátů je obtížné nejen diagnostikovat, ale především léčit. Tyto infekce vznikají díky mikrobní adhezi na daný materiál a následné tvorbě biofilmu. Mikroorganismy ve formě biofilmu snadno získávají rezistenci vůči tradičně používaným antibiotikům. Jedinou možností léčby je v současné době další chirurgický zákrok v kombinaci s dlouhodobou léčbou antibiotiky. Byla popsána celá řada přírodních látek, které jsou účinné proti mikrobní adhezi nebo fungují jako inhibitory quorum sensing a zároveň nepodporují vznik mikrobiální rezistence. Použití těchto látek je slibnou možností pro řešení problému infekcí kloubních náhrad.
The infections of prosthetic joints are not only diÇcult to diagnose, but also treat it. These infections occur by microbial adhesion on the surface and the formation of biofilm consequently. Microorganisms in biofilm soon develop a resistance against traditional antibiotics. Entirely one solution is known today – additional surgery in combination with long-term antibiotic therapy. Recently, there were described a wide range of natural products, which are able to decrease microbial adhesion or inhibit quorum sensing system and they do not support the emergence of microbial resistance. The use of these substances is promising way for solving the problem with these infections.

Mikrobiální biofilmy jsou společenství mikroorganismů, která jsou nevratně přichycena k povrchu, rozhraní či k sobě navzájem a jsou obalena v extracelulární polymerní matrix. Tato mikrobiální uskupení jsou zodpovědná za širokou škálu onemocnění a kontaminace lékařských nástrojů či průmyslových zařízení. Protože mikroorganismy žijící ve formě biofimu jsou až tisíckrát odolnější než planktonické buňky, prevence vzniku či eradikace existujících biofilmů je velice obtížná. To je důvod zvýšeného zájmu mnoha vědeckých skupin o studium dané problematiky. Použití vyšších koncentrací antibiotik není vhodným řešením, protože mikroorganismy si vůči nim velmi snadno vytváří multirezistenci. Proto je velice aktuální snaha o využití přírodních látek pro léčbu biofilmových infekcí. Přírodní látky mohou být často použity v nižších koncentracích a u mikroorganismů vůči nim nevzniká rezistence. V nedávné době byla u mnoha přírodních látek produkovaných mikroorganismy či izolovaných z rostlin experimentálně potvrzena jejich antimikrobiální, antifungální či dokonce antibiofilmová aktivita. Jako příklady lze uvést polyfenoly, terpeny, flavonoidy, stilbeny, některé složky běžně používaného koření, biosurfaktanty a mnoho dalších. Tento přehledový článek pojednává o možných aplikacích některých z nich.
Microbial biofilms are communities of microorganisms, which are irreversibly attached to a surface, interface or to each other and are embedded in an extracellular polymeric matrix. They cause wide range of human infections and contaminate medical instruments and industrial devices. Because microorganisms in biofilm are up to 1000 times more resistant than planktonic cells, the prevention or eradication of biofilm is extremely difficult. It is the reason of increased attention to this topic in many groups of researchers. Higher concentrations of antibiotics are not solution because microorganisms soon develop multiresistance. Therefore the use of natural substances represents new promising method for the treatment of biofilm infections. They can be used in low concentrations and no resistance against them is developed. Recently many natural substances with antibacterial, antifungal or antibiofilm activities were isolated from microorganisms or plants. Examples of these substances are groups of polyphenols, flavonoids, stilbenes, terpenes, commonly used spices, biosurfactants and many others. In this review the possible application of selected natural substances is described.
