Here, we present newly derived in vitro model for modeling Duchenne muscular dystrophy. Our new cell line was derived by reprogramming of peripheral blood mononuclear cells (isolated from blood from pediatric patient) with Sendai virus encoding Yamanaka factors. Derived iPS cells are capable to differentiate in vitro into three germ layers as verified by immunocytochemistry. When differentiated in special medium, our iPSc formed spontaneously beating cardiomyocytes. As cardiomyopathy is the main clinical complication in patients with Duchenne muscular dystrophy, the cell line bearing the dystrophin gene mutation might be of interest to the research community.
Cieľ: Karotický stenting je považovaný za dôležitú minimálne invazívnu metódu liečby stenózy karotických tepien. Použitie stentov prvej generácie bolo v porovnaní s karotickou endarterektómiou spojené s vyššou mierou neurologických komplikácií. S cieľom minimalizácie prolapsu aterosklerotického plátu a následnej embolizácie bola vyvinutá druhá generácia stentov pokrytých mikrosieťkou. Cieľom tohto systematického review je popísať hlavné výhody a prípadné limitácie použitia karotických stentov s dvojvrstvovým dizajnom. Metódy: Články boli vybrané v súlade s odporučením PRISMA. Vyhľadávaním v databázach Scopus, PubMed a Web of Science bolo k analýze vybraných 247 publikácií. Odstránených bolo 128 duplikátov a 81 publikácií na základe vylučovacích kritérií. Preštudovaním zoznamu referencií ostávajúcich 38 článkov bolo dohľadaných ďalších šesť relevantných zdrojov. Do review bolo zahrnutých celkom 44 publikácií. Výsledky: Štúdie ukazujú, že použitie dvojvrstvových stentov redukuje výskyt periprocedurálnej cerebrálnej embolizácie, 30-dňový aj 12-mesačný výskyt ipsilaterálnej cievnej mozgovej príhody u asymptomatických aj symptomatických pacientov. Stenty druhej generácie však vykazujú zásadné rozdiely v konštrukcii stentu a tiež v ich klinických výsledkoch. Záver: Technológia dvojvrstvových stentov môže zlepšiť krátkodobé aj dlhodobé výsledky karotického stentingu. Aj keď chýbajú randomizované štúdie porovnávajúce stenty druhej generácie s chirurgickou liečbou, už v súčasnosti publikované výsledky môžu významne ovplyvniť rozhodovanie v primárnej a sekundárnej prevencii mozgovej príhody
Aim: Carotid artery stenting is considered an important minimally invasive treatment modality in stenotic carotid artery disease. First-generation carotid stents have been associated with a higher rate of neurologic complications than carotid endarterectomy. To minimize atherosclerotic plaque prolapses and subsequent embolization, a second generation of micromesh-covered stents has been developed. The aim of this systematic review is to describe the main advantages and possible limitations of carotid stents with dual-layer design. Methods: The articles were selected in accordance with the PRISMA guidelines. The databases Scopus, PubMed and Web of Science were searched, 247 publications were selected for analysis, and 128 duplicates were removed. Based on the exclusion criteria, 81 publications were omitted. The reference lists of the 38 remaining articles were reviewed and six additional relevant sources were found. A total of 44 publications were included in the review. Results: Studies show that the use of dual-layer stents reduces the incidence of periprocedural cerebral embolism and the incidence of ipsilateral stroke at 30 days and 12 months both in asymptomatic and symptomatic patients. However, second generation stents show fundamental differences in stent construction and also in their clinical outcomes. Conclusion: The dual-layer stent technology may improve short- and long-term clinical results of carotid artery stenting. Although randomized trials comparing second-generation stents with surgical treatment are lacking, currently published results may significantly influence decision-making in primary and secondary stroke prevention.

We present here a new iPS cell line for modeling sporadic form of ALS. Cell line was generated by reprogramming skin fibroblasts isolated with explant culture technology from skin biopsy, donated by ALS patient. For reprogramming, polycistronic self-replicating RNA vector was used and derived iPS cells were characterized by immunocytochemistry and FACS (pluripotent factors expression), karyotyping, STR fingerprinting analysis and in vitro differentiation assay. New cell line showed normal (46, XY) karyotype and differentiated in vitro into cells from three germ layers. STR analysis proved the origin and originality of the cell line.

INTRODUCTION: Loss of consciousness (LOC) is used as a diagnostic feature of mild traumatic brain injury (MTBI). However, only 10% of concussions result in LOC. There are only a limited number of in-vivo studies dealing with unconsciousness and structural and functional integrity of the brainstem in patients with MTBI. The aim of our pilot study was to assess the sensitivity of proton magnetic resonance spectroscopy (1H-MRS) to detect metabolic changes in the brainstem in patients after MTBI with unconscioussness. METHODS: Twenty-four patients (12 with LOC, and 12 without LOC) within 3 days of MTBI and 19 healthy controls were examined. All subjects underwent single-voxel 1H-MRS examination of the upper brainstem. Spectra were evaluated using LCModel software. Ratios of total N-acetylaspartate (tNAA), total choline-containing compounds (tCho) and glutamate plus glutamine (Glx) to total creatine (tCre) were used for calculations. RESULTS: We found a significant decrease in tNAA/tCre and tCho/tCre ratios in the patient group with LOC when compared with the control group of healthy volunteers (P=0.002 and P=0.041, respectively), and a significant decrease in the tNAA/tCre ratio in the LOC group when compared with patients without LOC (P=0.04). Other metabolite ratios in the brainstem did not show any significant group differences. CONCLUSION: Our findings indicate that decrease of tNAA/tCre ratio in the upper brainstem using single-voxel 1H-MRS may provide a potential biomarker for MTBI associated with LOC.
Objev indukované pluripotence v roce 2006 umožnil revoluční způsob získávaní autologních terapeuticky aplikovatelných buněk, a mož‐ nost modelovat jakékoliv onemocnění v in vitro podmínkách. Možnost vrátit libovolnou, finálně diferencovanou buňku „v čase“ zpátky do stádia pluripotence je zajímavé i pro oblast onkologického výzkumu. Tato technologie umožnila studium procesů spojených s roz‐ vojem nádorového fenotypu buňky a taky s přechodem nádorové buňky do stádia s nižší mírou diferenciace. Reprogramování buněk do indukovaných pluripotentních kmenových buněk také pomáhá mnohem lépe studovat raritní populaci buněk, přítomných v nádo‐ rech – tzv. nádorové kmenové buňky. Indukovaná pluripotence některých typů nádorových buněk, spojená s jejich následnou řízenou diferenciací by se zároveň mohla stát jednou z možných terapeutických aplikací v onkologii.
Discovery of technology of induced pluripotency that allows the generation of autologous therapeutically applicable cells and generati‐ on of in vitro cell models for diseases with limited (or highly invasive) access to tested cells has also opened new horizons in the field of oncology research. The unique ability to reprogram the cancer cell into pluripotency with subsequent directed differentiation into cell with no malignant phenotype should be considered as a challenge in the field of new oncotherapy development. Although still conside‐ red to be realistic only on the level of experimental approach, the recent progress in the field of induced pluripotency gives the hope that dedifferentiation‐based therapies connected with the erase of malignant phenotype of original cancer cell will be more realistic in near future. By then, the most important role of induced pluripotency in oncology remains in the field of regenerative therapy as a source of autologous cells for regeneration of tissues or organs damaged by tumor growth or aggressive therapy
S migrénou je spojených viacero komorbidít. V článku sú uvedené najvýznamnejšie ochorenia, ktoré k nim patria. Stručne sú rozobrané vybrané predpokladané patomechanizmy, ako aj špecifiká liečby migrény s ohľadom na prítomnú komorbiditu.
Migraine is associated with several comorbidities. The most important comorbidities are mentioned. Basic supposed pathomechanisms are shortly discussed together with specific migraine treatment with regards on present comorbidity.
Pankreatický duktální adenokarcinom (PDAC) představuje jeden z nejagresivnějších typů lidských malignit. V současnosti je toto zhoubné onemocnění čtvrtou nejčastější příčinou úmrtí na rakovinu. Pětileté přežití pacientů s duktálním adenokarcinomem je méně než 8 %. Nové in vitro a in vivo modely jsou proto nutně potřebné pro vývoj nových terapií. S vlastní technologií pro derivaci nových, unikátních 3D nádorových buněčných linií izolovaných z lidských nádorů a ve spolupráci s Ústavem živočišné fyziologie a genetiky AV ČR v Liběchově představujeme plán vývoje velkého zvířecího modelu pro modelování lidského PDAC. S využitím tohoto modelu a nejmodernějších laboratorních technik provedeme profilovací analýzy (detekce a fenotypování cirkulujících rakovinných buněk, izolace a sekvenování cirkulující DNA, metabolomické profilování a analýzu onkoproteinů, a detekce cytokinů pomocí multiplexních protilátkových čipů za účelem nalezení biomarkerů nádoru pankreatu. Nově vyvinutý model zároveň poskytneme pro potřeby výzkumu spolupracujícím vědeckým pracovištím a farmaceutickým společnostem.
Pancreatic ductal adenocarcinoma represents one of the most aggressive type of human malignancy. Currently, this malignancy is the fourth most frequent cause of dead. 5-year survival of patients with ductal adenocarcinoma is less than 8 %. New in vitro and in vivo models are therefore desperately needed for new therapy development. With our own technology for derivation of new, unique 3D cancer cell lines from human tumors and in collaboration with Institute of Animal Physiology and Genetics, AS CR, in Libechov, we present here the plan for the development of large xenograft animal model (pig) bearing human pancreatic tumor. With the use of this model and state -of -the -art lab.techniques, we will perform multiple profiling analyses (circulating cancer cell detection and phenotyping, circulating DNA isolation and sequencing, metabolomic profiling and cancer -related proteins and cytokines detection with multiplex antibody array chips in order to find the hallmarks of pancreatic tumor.
The disease severity of COVID-19, especially in the elderly and patients with co-morbidities, is characterized by hypercytokinemia, an exaggerated immune response associated with an uncontrolled and excessive release of proinflammatory cytokine mediators (cytokine storm). Flavonoids, important secondary metabolites of plants, have long been studied as therapeutic interventions in inflammatory diseases due to their cytokine-modulatory effects. In this review, we discuss the potential role of flavonoids in the modulation of signaling pathways that are crucial for COVID-19 disease, particularly those related to inflammation and immunity. The immunomodulatory ability of flavonoids, carried out by the regulation of inflammatory mediators, the inhibition of endothelial activation, NLRP3 inflammasome, toll-like receptors (TLRs) or bromodomain containing protein 4 (BRD4), and the activation of the nuclear factor erythroid-derived 2-related factor 2 (Nrf2), might be beneficial in regulating the cytokine storm during SARS-CoV-2 infection. Moreover, the ability of flavonoids to inhibit dipeptidyl peptidase 4 (DPP4), neutralize 3-chymotrypsin-like protease (3CLpro) or to affect gut microbiota to maintain immune response, and the dual action of angiotensin-converting enzyme 2 (ACE-2) may potentially also be applied to the exaggerated inflammatory responses induced by SARS-CoV-2. Based on the previously proven effects of flavonoids in other diseases or on the basis of newly published studies associated with COVID-19 (bioinformatics, molecular docking), it is reasonable to assume positive effects of flavonoids on inflammatory changes associated with COVID-19. This review highlights the current state of knowledge of the utility of flavonoids in the management of COVID-19 and also points to the multiple biological effects of flavonoids on signaling pathways associated with the inflammation processes that are deregulated in the pathology induced by SARS-CoV-2. The identification of agents, including naturally occurring substances such as flavonoids, represents great approach potentially utilizable in the management of COVID-19. Although not clinically investigated yet, the applicability of flavonoids against COVID-19 could be a promising strategy due to a broad spectrum of their biological activities.

We generated new in vitro model for sporadic form of amyotrophic lateral sclerosis by reprogramming isolated skin fibroblasts into iPSCs. Fibroblasts were reprogrammed with commercially available synthetic polycistronic, self-replicating RNA vector. As verified by FISH, an early passages of a new iPSC line showed mosaic karyotype (cells with normal and abnormal karyotype 46,XY,t(2;14)(q13;p12) were present), while late passages contained only cells with abnormal karyotype. New iPSCs differentiated into all three germ layers and formed a teratoma in nude mice. Our iPSC line represents a new model for therapy testing and drug development in the field of ALS research.

Objev indukovaných pluripotentních kmenových buněk profesorem Shinya Yamanakem, který byl v roce 2012 oceněn Nobelovou cenou, otevřel nové horizonty v oblasti regenerativní medicíny, in vitro modelování nemocí nebo screeningu léčiv. Unikátní technologie, která umožňuje přípravu eticky přijatelných autologních kmenových buněk, činí reálnou oblast personalizované medicíny. Technologie iPSCs našla využití rovněž při in vitro modelování, kde umožnila přípravu buněk těžko dostupných tkání, např. neuronů nebo kardiocytů, z poměrně lehce dostupného zdroje, kterým jsou např. dermální fibroblasty. Takto připravené in vitro biomedicínské modely jsou aplikovatelné v preklinických studiích resp. při testování léčiv. Technologie iPSCs má široký potenciál využití v různých medicínských oblastech a rovněž již našla upotřebení také v boji proti nové pandemii koronaviru.
Discovery of induced pluripotent stem cells by Dr. Shinya Yamanaka, awarded by Nobel Prize in 2012 opened new horizonts in the field of regenerative medicine, in vitro disease modeling and drug screening. A unique technology that allows preparation of ethical issues-free, autologous therapeutic stem cells is revolutionizing the field of patient -tailored therapies. For in vitro modeling, iPSc technology represents a unique possibility for preparation of hard -to -get cells (neurons,cardiomyocytes etc) from patient skin fibroblasts for further studies and drug testing. iPSc technology has potential and is already used in world research effort in fight against new coronavirus pandemy.
