Tento článek obsahuje dosavadní poznatky o tradiční amazonské medicíně v České republice. Rovněž je v něm zahrnuta klasifikace pořádaných obřadů a charakteristika jejich pořadatelů a účastníků. Terénní výzkum mezi uživateli tradiční amazonské medicíny byl realizován od listopadu 2015. Výzkumný vzorek tvoří 46 osob (23 žen a 23 mužů), které se v minulosti s tradiční amazonskou medicínou minimálně jednou zúčastnily nějakého typu rituálu. S těmito osobami byly nahrány semi-strukturované rozhovory. Cílem tohoto článku je kategorizovat jejich obsah. Za tímto účelem jsme provedli obsahovou analýzu interview metodou zakotvené teorie. V tomto článku prezentujeme výsledky analýzy prvních 15 rozhovorů. Tyto výsledky potvrzují, že v České republice je nejčastěji užívaným přípravkem z tradiční amazonské medicíny psychoaktivní odvar ayahuasca. V závěru článku jsou uvedena rizika spojená s užíváním psychoaktivních látek.
This article assesses the present situation in traditional Amazonian medicine in the Czech Republic. It also classifies the types of organized rituals and characterizes their organizers and participants in the country. The fieldwork among traditional Amazonian medicine users was conducted since November 2015. A total of 46 respondents (23 women and 23 men) who had attended at least one ritual of traditional Amazonian medicine were selected for the study and we recorded with them semi-structured interviews. The aim of this article is to categorize their contents. In order to fulfill this objective, we performed a content analysis of interviews based on the Grounded Theory method. In this article we publish results of analysis of the first 15 interviews. These results confirm that in the Czech Republic a psychoactive brew ayahuasca is the most used preparation from traditional Amazonian medicine. Finally, there are risks associated with the use of psychoactive substances indicated.

Archaeological research is radically transforming the view that the Amazon basin and surrounding areas witnessed limited societal development before European contact. Nevertheless, uncertainty remains on the nature of the subsistence systems and the role that aquatic resources, terrestrial mammalian game, and plants had in supporting population growth, geographic dispersal, cultural adaptations and political complexity during the later stages of the pre-Columbian era. This is exacerbated by the general paucity of archaeological human remains enabling individual dietary reconstructions. Here we use stable carbon and nitrogen isotope analysis of bone collagen to reconstruct the diets of human individuals from São Luís Island (Brazilian Amazon coast) dated between ca. 1800 and 1000 cal BP and associated with distinct ceramic traditions. We expanded our analysis to include previously published data from Maracá and Marajó Island, in the eastern Amazon. Quantitative estimates of the caloric contributions from food groups and their relative nutrients using a Bayesian Mixing Model revealed distinct subsistence strategies, consisting predominantly of plants and terrestrial mammals and variably complemented with aquatic resources. This study offers novel quantitative information on the extent distinct food categories of polyculture agroforestry systems fulfilled the caloric and protein requirements of Late Holocene pre-Columbian populations in the Amazon basin.

Camu-camu [Myrciaria dubia (Kunth) McVaugh] is currently an important and promising fruit species grown in the Peruvian Amazon, as well as in Brazil, Colombia, and Bolivia. The species is valued for its high content of fruit-based vitamin C. Large plantations have been established only in the last two decades, and a substantial part of the production is still obtained by collecting fruits from the wild. Domestication of the species is at an early stage; most farmers cultivate the plants without any breeding, or only through a simple mass selection process. The main objective of the study was to characterize morphological and genetic variation within and among cultivated and natural populations of camu-camu in the Peruvian Amazon. In total, we sampled 13 populations: ten wild in the Iquitos region, and three cultivated in the Pucallpa region in the Peruvian Amazon. To assess the genetic diversity using seven microsatellite loci, we analyzed samples from ten individual trees per each population (n = 126). Morphological data was collected from five trees from each population (n = 65). The analysis did not reveal statistically significant differences for most of the morphological descriptors. For wild and cultivated populations, the observed heterozygosity was 0.347 and 0.404 (expected 0.516 and 0.506), and the fixation index was 0.328 and 0.200, respectively. Wild populations could be divided into two groups according to the UPGMA and STRUCTURE analysis. In cultivated populations, their approximate origin was determined. Our findings indicate a high genetic diversity among the populations, but also a high degree of inbreeding within the populations. This can be explained by either the isolation of these populations from each other or the low number of individuals in some populations. This high level of genetic diversity can be explored for the selection of superior individuals for further breeding.

A survey of proteocephalidean cestodes found in the firewood catfish Sorubimichthys planiceps (Spix et Agassiz) from the Amazon River is provided. The following taxa parasitic in S. planiceps are redescribed on the basis of their type specimens and material collected recently in the Amazon River, near the type localities in Brazil, and in Iquitos, Peru: Monticellia lenha Woodland, 1933; Nomimoscolex lenha (Woodland, 1933) (syn. Proteocephalus lenha Woodland, 1933); and Monticellia megacephala Woodland, 1934, for which a new genus, Lenhataenia, is proposed, with L. megacephala (Woodland, 1934) comb. n. as its type and only species. The new genus is a member of the Monticelliinae, i.e. has all genital organs in the cortex, and is most similar to Chambriella in possessing biloculate suckers and lacking a metascolex. It differs in the morphology of the cirrus-sac that contains a strongly coiled, thick-walled internal sperm duct (vas deferens) and a muscular cirrus of the appearance typical of most proteocephalideans, whereas that of Chambriella is sigmoid, with voluminous, tightly sinuous thick-walled internal sperm duct. In addition, Lenhataenia possesses a well developed internal musculature, whereas the internal musculature of Chambriella is weakly developed, formed by a low number of muscle fibres. The scolex morphology and distribution of microtriches of Peltidocotyle lenha (Woodland, 1933) (syn. Othinoscolex lenha Woodland, 1933 and Othinoscolex myzofer Woodland, 1933), Chambriella sp. and Choanoscolex sp. are described using scanning electron microscopy. The two latter taxa may be new for science and are reported from S. planiceps for the first time.
The aim of this study was to evaluate the antimicrobial activity of six barks traditionally used in Callería District (Ucayali Department, Peru) for treating conditions likely to be associated with microorganisms. Ethanol extracts of stem barks of Abuta grandifolia (Menispermaceae), Dipteryx micrantha (Leguminosae), Cordia alliodora (Boraginaceae), Naucleopsis glabra (Moraceae), Pterocarpus rohrii (Leguminosae), and root bark of Maytenus macrocarpa (Celastraceae) were tested against nine bacteria and one yeast using the broth microdilution method. All plants possessed significant antimicrobial effect, however, the extract of Naucleopsis glabra exhibited the strongest activity against Gram-positive bacteria (MICs ranging from 62.5 to 125 microg/ml), while the broadest spectrum of action was shown by the extract of Maytenus macrocarpa, which inhibited all the strains tested with MICs ranging from 125 to 250 microg/ml.

An analysis of the population structure and genetic diversity for any organism often depends on one or more molecular marker techniques. Nonetheless, these techniques are not absolutely reliable because of various sources of errors arising during the genotyping process. Thus, a complex analysis of genotyping error was carried out with the AFLP method in 169 samples of the oil seed plant Plukenetia volubilis L. from small isolated subpopulations in the Peruvian Amazon. Samples were collected in nine localities from the region of San Martin. Analysis was done in eight datasets with a genotyping error from 0 to 5%. Using eleven primer combinations, 102 to 275 markers were obtained according to the dataset. It was found that it is only possible to obtain the most reliable and robust results through a multiple-level filtering process. Genotyping error and software set up influence both the estimation of population structure and genetic diversity, where in our case population number (K) varied between 2-9 depending on the dataset and statistical method used. Surprisingly, discrepancies in K number were caused more by statistical approaches than by genotyping errors themselves. However, for estimation of genetic diversity, the degree of genotyping error was critical because descriptive parameters (He, FST, PLP 5%) varied substantially (by at least 25%). Due to low gene flow, P. volubilis mostly consists of small isolated subpopulations (ΦPT = 0.252-0.323) with some degree of admixture given by socio-economic connectivity among the sites; a direct link between the genetic and geographic distances was not confirmed. The study illustrates the successful application of AFLP to infer genetic structure in non-model plants.

Two new genera and three new species of paramphistomoid digeneans are described in the family Cladorchiidae Fischoeder, 1901 from doradid, heptapterid, pimelodid and pseudopimelodid fishes in the Amazon River in Peru. Goeldamphistomum amazonum n. g., n. sp. (type-species) from Goeldiella eques (Müller & Troschel) (type-host) and Tenellus trimaculatus (Boulenger), and Goeldamphistomum peruanum n. g., n. sp. from Duopalatinus peruanus Eigenmann & Allen (type-host), Calophysus macropterus (Lichtenstein) and Microglanis sp. are placed in the Dadayiinae Fukui, 1929. Both species have an accessory acetabular sucker, which distinguishes the genus from all taxa previously reported from South American freshwater fishes. They differ from each other primarily in that G. amazonum has a prebifurcal genital pore and oblique, separated testes, the levels of which rarely overlap longitudinally, whereas G. peruanum has a postbifurcal genital pore and testes directly to obliquely tandem. Iquitostrema papillatum n. g., n. sp. (Kalitrematinae Travassos, 1933) from the intestine of Hassar orestis (Steindachner) differs from other members of the subfamily in the combination of a massive acetabulum with a papillate luminal surface and symmetrical testes which overlie the caeca close to the caecal arch. These are the first records of paramphistomes from the five host species studied here.

A new nematode species, Philometra mirabilis sp. n. (Philometridae), is described based on a subgravid female specimen recovered from the ovary of the freshwater perciform fish Cichla mirianae Kullander and Ferreira (Cichlidae) in the Juruena River (Amazon River basin), State of Mato Grosso, Brazil. The new species is morphologically very different from congeners parasitizing fishes in South America, being mainly characterized by the markedly elongate, narrow body 171 mm long (maximum width/body length 1:598), the presence of three small cone-shaped oesophageal teeth protruding out of the mouth and an onion-shaped oesophageal inflation distinctly separated from the posterior part of the oesophagus, the relative length of the oesophagus, and the rounded posterior end of the body without any caudal projections. It is the third known valid species of Philometra Costa, 1845 parasitizing a freshwater fish in South America and the second species of this genus reported from fishes of the family Cichlidae.

Plants mentioned in this study have numerous records in traditional Peruvian medicine being used in treatment of cancer and other diseases likely to be associated with oxidative stress. Amongst the eight plant species tested, only Dysphania ambrosioides exhibited combinatory antioxidant and anti-proliferative effect on a broad spectrum of cancer cells (DPPH and ORAC values = 80.6 and 687.3 μg TE/mg extract, respectively; IC50 against Caco-2, HT-29 and Hep-G2 = 129.2, 69.9 and 130.6, respectively). Alkaloids and phenolic compounds might significantly contribute to anticancer/antioxidant activity of this plant. The results justify the traditional medicinal use of this plant. Our findings further suggest that D. ambrosioides might serve as a prospective material for further development of novel plant-based antioxidant and/or anti-proliferative agents. Detailed analysis of chemical composition together with toxicology assessments and in vivo antioxidant/anti-proliferative activity of this plant should be carried out in order to verify its potential practical use.

Investigations into the chemical constituents of the seeds of the neglected tuber crop Pachyrhizus tuberosus (Leguminosae) resulted in the isolation of seven components: five rotenoids [12a-hydroxyerosone (1), 12a-hydroxydolineone (2), erosone (3), 12a-hydroxyrotenone (4) and rotenone (6)], a phenylfuranocoumarin [pachyrrhizine (5)] and an isoflavanone [neotenone (7)]. The compounds were isolated using several chromatography techniques and characterized and verified by NMR and HPLC/MS. The MTT assay was used to examine the selective cytotoxic effects of the methanolic P. tuberosus extract and isolated compounds in two human cancer cell lines [breast (MCF-7) and colorectal (HCT-116)] and in non-transformed human fibroblasts (MRC-5); IC50 values were calculated. The methanolic P. tuberosus extract displayed respectable cytotoxic effects against HCT-116 and MCF-7 cells with IC50 values of 7.3 and 6.3 microg/mL, respectively. Of the compounds, 6 exacted greatest cytotoxicity and selectivity towards the cancer cell lines tested, yielding IC50 values of 0.3 microg/mL against both MCF-7 and HCT-116 cells, and a 6-fold reduced activity against MRC-5 fibroblasts. Compound 4 also demonstrated cytotoxicity against MCF-7 and HCT-116 (1.1 and 1.8 microg/mL, respectively), and reduced cytotoxicity towards MRC-5 cells (7.5 mirog/mL). The results revealed from the in vitro cytotoxic MTT assay are worthy of further antitumor investigation.
