Tor barbs, Tor tor (Hamilton), a cyprinid fish collected from Barak Valley, Assam, India were examined for helminth parasites. A nematode species of the genus Rhabdochona Railliet, 1916 (Rhabdochonidae) was found in the intestine of the fish specimens. The morphology of these nematodes was characterised by the presence of a funnel shaped prostom, head with four submedian sublabia, four cephalic papillae, long and slender posterior glandular oesophagus in both sexes, presence of 14 caudal papillae and two unequal spicules with pointed proximal tips in male specimens, and prominent post-equatorial vulval position, posteriorly directed vagina, uterus with fully scattered eggs in female specimens, and a tail pointed with a small mucron in both sexes. This nematode species was subsequently identified as Rhabdochona hospeti Thapar, 1950, reported herein for the first time in the northeastern India. In addition to its morphological description based on light microscopy, partial 18S and 28S rDNA sequences were generated to assess phylogenetic relationships of R. hospeti with other species of Rhabdochona.

IN BRIEF: Understanding the establishment of post-fertilization totipotency has broad implications for modern biotechnologies. This review summarizes the current knowledge of putative egg components governing this process following natural fertilization and after somatic cell nuclear transfer. ABSTRACT: The mammalian oocyte is a unique cell, and comprehending its physiology and biology is essential for understanding fertilization, totipotency and early events of embryogenesis. Consequently, research in these areas influences the outcomes of various technologies, for example, the production and conservation of laboratory and large animals with rare and valuable genotypes, the rescue of the species near extinction, as well as success in human assisted reproduction. Nevertheless, even the most advanced and sophisticated reproductive technologies of today do not always guarantee a favorable outcome. Elucidating the interactions of oocyte components with its natural partner cell - the sperm or an 'unnatural' somatic nucleus, when the somatic cell nucleus transfer is used is essential for understanding how totipotency is established and thus defining the requirements for normal development. One of the crucial aspects is the stoichiometry of different reprogramming and remodeling factors present in the oocyte and their balance. Here, we discuss how these factors, in combination, may lead to the formation of a new organism. We focus on the laboratory mouse and its genetic models, as this species has been instrumental in shaping our understanding of early post-fertilization events.

BACKGROUND: DNA methylation is likely a key mechanism regulating changes in gene transcription in traits that show temporal fluctuations in response to environmental conditions. To understand the transcriptional role of DNA methylation we need simultaneous within-individual assessment of methylation changes and gene expression changes over time. Within-individual repeated sampling of tissues, which are essential for trait expression is, however, unfeasible (e.g. specific brain regions, liver and ovary for reproductive timing). Here, we explore to what extend between-individual changes in DNA methylation in a tissue accessible for repeated sampling (red blood cells (RBCs)) reflect such patterns in a tissue unavailable for repeated sampling (liver) and how these DNA methylation patterns are associated with gene expression in such inaccessible tissues (hypothalamus, ovary and liver). For this, 18 great tit (Parus major) females were sacrificed at three time points (n = 6 per time point) throughout the pre-laying and egg-laying period and their blood, hypothalamus, ovary and liver were sampled. RESULTS: We simultaneously assessed DNA methylation changes (via reduced representation bisulfite sequencing) and changes in gene expression (via RNA-seq and qPCR) over time. In general, we found a positive correlation between changes in CpG site methylation in RBCs and liver across timepoints. For CpG sites in close proximity to the transcription start site, an increase in RBC methylation over time was associated with a decrease in the expression of the associated gene in the ovary. In contrast, no such association with gene expression was found for CpG site methylation within the gene body or the 10 kb up- and downstream regions adjacent to the gene body. CONCLUSION: Temporal changes in DNA methylation are largely tissue-general, indicating that changes in RBC methylation can reflect changes in DNA methylation in other, often less accessible, tissues such as the liver in our case. However, associations between temporal changes in DNA methylation with changes in gene expression are mostly tissue- and genomic location-dependent. The observation that temporal changes in DNA methylation within RBCs can relate to changes in gene expression in less accessible tissues is important for a better understanding of how environmental conditions shape traits that temporally change in expression in wild populations.

A total of 52 specimens of the Pacific seabream Acanthopagrus pacificus Iwatsuki, Kume et Yoshino from the Gulf of Tonkin off Vietnam were examined for monogeneans. Twenty fish were parasitised by 101 individuals of five monogenean species, including two known species Allodiscocotyla diacanthi Unnithan, 1962 and Heterapta chorinemi (Tripathi, 1956), as well as three new species, Polylabroides tienyenensis sp. n., Polylabroides tonkinensis sp. n. and Metacamopia lebedevi sp. n. Polylabroides tienyenensis and P. tonkinensis are morphologically more similar to Polylabroides guangdongensis Zhang et Yang, 2001 in comparison with other species within the genus, based on the absence of small spines on the cirrus. However, P. tonkinensis is distinguished from P. guangdongensis by fewer clamps on the haptor and by the different shapes of the large spines on the cirrus. Similarly, P. tienyenensis differs from P. guangdongensis by vaginal ducts with fewer branches, fewer clamps and smaller egg size. Metacamopia lebedevi is distinguished from Metacamopia chorinemi (Yamaguti, 1953) by the arrangement of testes (one row vs two rows), diverticula absent in the oesophagus, and the number of anchor pairs (one vs two); it differs from Metacamopia oligoplites Takemoto, Amato et Luque, 1996 by the smaller haptor, shape and absence of small sclerotised hooks, the number of ribs in their clamps, and the position of the testes; it can be separated from Metacamopia indica (Unnithan, 1962) by having fewer testes and lacking sclerotised structures in the vagina. The present study also provides the measurements for A. diacanthi, H. chorinemi, and proposes a new key to all species of Polylabroides.

Avian brood parasites lay their eggs in other birds' nests, and hosts can mitigate the fitness cost of raising unrelated offspring by rejecting parasitic eggs. A visually-based cognitive mechanism often thought to be used by hosts to discriminate the foreign egg is to compare it against the hosts' own eggshell by size, shape, maculation, and/or ground coloration (i.e., absolute chromatic contrast). However, hosts may instead discriminate eggs based on their colors along a scale of natural avian eggshell coloration (i.e., directional chromatic contrast). In support of this latter visual process, recent research has found that directional chromatic contrasts can explain some host species' rejection behavior better than absolute chromatic or achromatic contrasts. Here, for the first time, we conducted an experiment in a cavity-nesting host species to test the predictions of these different visual mechanisms. We experimentally parasitized nests of the Common Redstart Phoenicurus phoenicurus, a regular host of a mimetic-egg laying Common Cuckoo Cuculus canorus host-race, using painted, immaculate 3D-printed model eggs in two geographically distant areas (Finland and Czech Republic). We found that directional chromatic contrasts better explained rejection behaviors in both parasitized (Finland) and non-parasitized (Czech Republic) host populations, as hosts rejected eggs that were noticeably browner, but not eggs that were noticeably bluer, than redstart eggs. These results support the paradigm of a single rejection threshold predicted by the directional chromatic contrast model and contribute to a growing generality of these patterns across diverse avian host-brood parasite systems.

1. Selection strategies for broilers must balance rapid growth with the welfare and health of animals, strategies must deal with the trade-off with other vital functions.2. Divergent selection of Japanese quail for high (HG) and low (LG) relative body weight gain between 11 and 28 days of age has been conducted to accelerate linear phase growth without influencing the final adult body weight. Higher body growth rate is often connected with a weakened immune system. Therefore, the present study explored the immunological characterisation of quail from HG and LG lines, which differ substantially in their growth rate.3. The trial evaluated the maternal investment to immunologically active substances, cell-mediated immunity stimulated by phytohaemagglutinin (PHA) injection and the acute phase of the immune response to lipopolysaccharide (LPS) administration in three different phases of early postnatal growth.4. Except for higher lysozyme activity in the LG group when compared to the HG line, the maternal investment did not differ between the two lines. Plasma antibody concentrations responded quickly to any change in growth rate in both lines. Overall, it seems that initial rapid growth of the LG line had long-lasting effects on immune responsiveness, even after the growth rate of the HG line escalated during the linear phase of growth.5. The study indicated that changes in the growth rate caused by the selection for growth in meat-type Japanese quail can influence the acute phase of the immune response and development of the immune system.

One of the most effective defenses against avian brood parasitism is the rejection of the foreign egg from the host's nest. Until recently, most studies have tested whether hosts discriminate between own and foreign eggs based on the absolute differences in avian-perceivable eggshell coloration and maculation. However, recent studies suggest that hosts may instead contrast egg appearances across a directional eggshell color gradient. We assessed which discrimination rule best explained egg rejection by great reed warblers Acrocephalus arundinaceus, a frequent host to an egg-mimetic race of common cuckoos Cuculus canorus. We deployed 3D-printed model eggs varying in blue-green to brown coloration and in the presence of maculation. Using visual modeling, we calculated the absolute chromatic and achromatic just-noticeable differences (JNDs), as well as directional JNDs across a blue-green to brown egg color gradient, between host and model eggs. While most model eggs were rejected by great reed warblers, browner eggs were rejected with higher probability than more blue-green eggs, and the rejection probability did not depend on maculation. Directional egg color discrimination shown here and in a suite of recent studies on other host species may shape the cognitive decision rules that hosts use to recognize foreign eggs and affect the course of evolution in parasitic egg mimicry.

Necropsy of two free-ranging common blackbirds (Turdus merula) found dead in central Italy revealed the presence of a high number of cyclocoelid flukes in the coelomatic cavity. Cyclocoelid flukes primarily infect avian respiratory system. Histologically, air sac walls were covered with a fibrinous exudate containing degenerate heterophils, many trematodes and some colonies of Gram-positive cocci. The superficial bronchi and parabronchi were markedly distended, and the adjacent pulmonary parenchyma was congested and collapsed. Trematodes, surrounded by a mild suppurative to pyogranulomatous inflammatory reaction, were also observed on the pericardial, intestinal, kidney and hepatic serosal surfaces. The death of the two examined birds was likely due to the high parasite load and associated severe lesions. At parasitological examination, flukes showed a tongue-shaped elongate body, tapered anteriorly and rounded posteriorly, of 2,088-2,314 μm in width and 8,268-11,830 μm in length. The mouth was slightly oval and sub-terminal, with a small oral sucker. The oval pharynx measured 250-309 μm, and the two caeca joined posteriorly. Two large (550-702 μm × 450-520 μm) globular testes were situated obliquely to each other, whereas an oval (250 × 300 μm in mean) or round (about 334 μm in diameter) intertesticular ovary was placed in a longitudinal straight line with the testes. The ootype was about 110 μm in diameter, while the brown-yellow eggs measured 131.5 × 73.9 μm in mean. The genital pore was post-pharyngeal, while the symmetrically arranged vitelline glands were not confluent posteriorly. Morphoflogical diagnosis led to the identification of Morishitium polonicum, a cyclocoelid fluke species that typically inhabits the air sacs of blackbirds. The morphological diagnosis was corroborated by molecular phylogenetic analysis of the mitochondrial (CO1, ND1) DNA loci. The present study provides the first report of pathological lesions and death caused by M. polonicum in birds.

Based on light and scanning electron microscopical studies, a new nematode parasite, Capillaria appendigera n. sp. (Capillariidae), is described from the intestine of the goldbanded jobfish Pristipomoides multidens (Day) (Perciformes, Lutjanidae) from the Arafura Sea West, off the northern coast of Australia. The new species, belonging to the subgenus Procapillaria Moravec, 1987, differs from other congeneric species from fishes mainly in the length (0.92-1.13 mm), shape and structure of the spicule, obtuse spines on the spicule sheath and the structure of eggs. It is characterised, in the male, by the presence of two well-developed dorsolateral caudal lobes, a pair of lateral papillae, a heavily sclerotised spicule with many rough transverse grooves covering almost the entice spicule surface (except for spicule ends), a spinose spicule sheath, and in the female, by a subterminal anus, mostly the presence of a large vulval appendage and by eggs (size 54-69 × 27-33 µm) encapsulated by a conspicuous light-coloured superficial layer. Capillaria appendigera n. sp. is the 12th nominal species of capillariids recorded from fishes in Australian waters and the second known capillariid species parasitising fishes of the perciform family Lutjanidae. In addition, four unidentifiable, morphologically different types of capillariid females, probably representing undescribed species, were recorded from the intestines of marine fishes off the northern coast of Australia: Capillariidae gen. sp. 1 and Capillariidae gen. sp. 2 from Lutjanus johnii (Bloch) and L. malabaricus (Bloch & Schneider), respectively (both Lutjanidae), Capillariidae gen. sp. 3 from Protonibea diacanthus (Lacépède) (Sciaenidae) and Capillariidae gen. sp. 4 from Rachycentron canadum (Linnaeus) (Rachycentridae).
