Avian eggs contend with omnipresent microorganisms entering the egg interior, where they affect embryo viability and hatchling phenotype. The incubation behaviour and deposition of egg white antimicrobial proteins (AMPs) vary highly across the avian altricial-precocial spectrum. Experimental evidence of how these alterations in avian reproductive strategies affect the antimicrobial properties of the precocial and altricial egg interior is lacking, however. Here, we tested the egg white antimicrobial activity in eggs of two representative model species, from each end of the avian altricial-precocial spectrum, against potentially pathogenic and beneficial probiotic microorganisms. Eggs were experimentally treated to mimic un-incubated eggs in the nest, partial incubation during the egg-laying period, the onset of full incubation and the increased deposition of two main egg white AMPs, lysozyme and ovotransferrin. We moreover assessed to what extent egg antimicrobial components, egg white pH and AMP concentrations varied as a result of different incubation patterns. Fully incubated precocial and altricial eggs decreased their antimicrobial activity against a potentially pathogenic microorganism, whereas partial incubation significantly enhanced the persistence of a beneficial probiotic microorganism in precocial eggs. These effects were most probably conditioned by temperature-dependent alterations in egg white pH and AMP concentrations. While lysozyme concentration and pH decreased in fully incubated precocial but not altricial eggs, egg white ovotransferrin increased along with the intensity of incubation in both precocial and altricial eggs. This study is the first to experimentally demonstrate that different incubation patterns may have selective antimicrobial potential mediated by species-specific effects on antimicrobial components in the egg white.

In birds, spectrum of egg white proteins deposited into the egg during its formation are thought to be essential maternal effects. Particularly, egg white lysozyme (LSM), exhibiting great between and within species variability, is considered to be essential for developing avian embryos due to its physiological, antimicrobial, and innate immune defense functions. However, there have been few studies investigating effects of LSM on early post-hatching phenotype, despite its broad physiological and protective role during embryogenesis. Here, we test how experimentally increased concentrations of egg white LSM affect hatchability in Japanese quail (Coturnix japonica) and chick phenotype immediately after hatching (particularly body weight, tarsus length, plasma LSM concentration, and plasma complement activity). Chicks from eggs with increased LSM concentration displayed reduced tarsus length compared to chicks from control eggs while hatchability, body weight and plasma LSM concentration were unaffected. It is worth noting that no effect of increased in ovo lysozyme on eggs hatchability could be related to pathogen-free environment during artificial incubation of experimental eggs causing minimal pressure on embryo viability. While tangible in vivo mechanisms during avian embryogenesis remain to be tested, our study is the first to document experimentally that egg white LSM appears to have growth-regulation role during embryo development, with possible underlying phenotypic consequences in the early post-hatching period in precocial birds.

Chiroptical methods are widely used in structural and conformational analyses of biopolymers. The application of these methods to investigations of biofluids would provide new avenues for the molecular diagnosis of protein-misfolding diseases. In this work, samples of human blood plasma and hen egg white were analyzed using a combination of conventional and chiroptical methods: ultraviolet absorption/electronic circular dichroism (UV/ECD), Fourier transform infrared absorption/vibrational circular dichroism (FTIR/VCD), and Raman scattering/Raman optical activity (Raman/ROA). For comparison, the main components of these substances--human serum albumin (HSA) and ovalbumin (Ova)--were also analyzed by these methods. The ultraviolet region of the ECD spectrum was analyzed using the CDNN CD software package to evaluate the secondary structures of the proteins. The UV/ECD, FTIR/VCD, and Raman/ROA spectra of the substances were quite similar to those of the corresponding major proteins, while some differences were also detected and explained. The conclusions drawn from the FTIR/VCD and Raman/ROA data were in good agreement with the secondary structures calculated from ECD. The results obtained in this work demonstrate that the chiroptical methods used here can be applied to analyze not only pure protein solutions but also more complex systems, such as biological fluids.

Cíl: Zjistit, jak často se vyskytuje potravinová alergie na vaječné bílkoviny u dospívajících a dospělých pacientů s atopickým ekzémem a zda se podílí na zhoršení ekzémových projevů. Metodika: Bylo zahrnuto 179 pacientů s atopickým ekzémem starších 14 let, u kterých bylo provedeno vyšetření se zaměřením na zhodnocení významu potravinové alergie na vaječné bílkoviny (anamnéza, kožní prick testy, specifické sérové IgE, atopické epikutánní testy). U pacientů s pozitivním výsledkem na žloutek či bílek alespoň v jedné z vyšetřovacích metod byl proveden otevřený expoziční test s vejcem, kterému předcházela diagnostická hypoalergenní eliminační dieta. Potravinová alergie na vejce byla stanovena na základě pozitivního výsledku otevřeného expozičního testu a zhodnocení anamnestických údajů o časné alergické reakci. Výsledky: Výskyt potravinové alergie na vaječné bílkoviny jsme stanovili celkem u 11 pacientů (6 %) - časná reakce (pruritus, orální alergický syndrom, dušnost) byla zaznamenána u 2,7 %, pozdní reakce se zhoršením atopického ekzému u 3,3 % pacientů. Pozitivní výsledek ve specifickém IgE a kožních prick testech na vaječné bílkoviny (u 34 % pacientů) však u většiny z nich (28 %) nekoreloval s klinickými projevy potravinové alergie. Pomocí atopických epikutánních testů bylo možno zachytit potravinovou alergii na vaječné bílkoviny u 1 % pacientů s non IgE reaktivitou. Po eliminaci vejce došlo u pacientů zvláště s pozdní reakcí v otevřeném expozičním testu ke zlepšení kožního nálezu. Závěr: V diagnostice potravinové alergie u pacientů s atopickým ekzémem je vhodný komplexní diagnostický postup s provedením expozičního testu, kterému předchází diagnostická eliminační dieta.
Aim: The aim of our study was to investigate the frequency of egg proteins allergy in adolescent and adult patients with atopic eczema and its possible role in disease flares. Methods: Group of 179 atopic patients older than 14 years was investigated for the egg allergy (history, skin prick tests, serum specific IgE and atopy patch tests were performed) and its significance was evaluated. In patients with at least one positive test of either yolk or egg white the open exposure test with an egg was performed folloving hypoallergenic elimination diet. The egg allergy was diagnosed in case of positive result of the open exposure test and the evaluation of patient’s history. Results: Egg protein allergy was confirmed in 11 patients (6%). Both early reactions (in 2,7% of patients oral allergy syndrome, itching and asthma) and worsening of atopic eczema (in 3, 3% of patients) were noticed. Positivity of serum specific IgE and prick tests to egg proteins (in 34% of patients) in great part of them (in 28%) did not correlate with clinical symptoms of allergy. Atopy patch tests revealed egg allergy in 1% of IgE non-reactive patients. After egg eliminaton the skin symptoms improved especially in those with late reaction in open exposure tests. Conclusion: To diagnose food allergy in atopic patients the complete diagnostic process including exposure test after elimination diet is necessary.

Dva kojenci, chlapci, narození v 11/00 a 12/00 z normálního těhotenství, u jednoho komplikovaný porod zakončen s. C. Rodinná zátěž u obou chlapců, matky polinotičky. Obě děti od narození plně kojeny. Od 2. měsíce života se objevil generalizovaný atopický ekzém, který i přes kožní léčbu přetrvává. Jeden z chlapců prodělal anafylaktickou reakci po 2 ml Sunaru s otoky a dušností. Obě děti byly alergologicky vyšetřeny a byla prokázána senzibilizace na bílkoviny kravského mléka (KM), u jednoho dítěte betalactoglobulin, u druhého kasein, betalactoglobulin i alfalactalbumin, a na vaječný bílek. U 1 pacienta byly provedeny kožní prick testy se stejným výsledkem. Oba chlapci mají vysoké celkové IgE a nápadnou hypoimunoglobulinémii hlavně IgG. Obě matky po porodu pily mléko v normálním, až vyšším množství. Literární údaje o přenosu bílkovin KM mateřským mlékem jsou rozporné, přenos je prokázán, ale v malém množství a možnost senzibilizace touto cestou je zpochybňována. V případech našich pacientů je však senzibilizace cestou mateřského mléka velmi suspektní. Obě děti jsou dále sledovány.
Two infants, boys with positive family history, mothers having pollen allergy. Both were exclusively breast fed. Severe atopic dermatitis started from 2nd month of life, dermatological treatment, even with topical steroids was not effective. One boy had anaphylaxis after 2 ml of cow milk formula with generalised urticaria, edema and dyspnea. Sensitisation to cow milk and egg-white proteins was found in both infants. Specific IgE antibodies were positive in both, in one boy skin prick tests for milk and egg were positive. Both boys have high levels of total IgE and very low levels of IgG and IgA antibodies. Mothers used cow milk and cow milk products in their diet before testing infants, afterwards the cow milk and eggs were eliminated. Two month after elimination skin symptoms in both infants disappeared. We suppose sensitisation via transfer of cow milk and egg proteins by breast milk. Both boys are followed up.

Elektrotermokoagulace pulpózního jádra meziobratlové ploténky, která způsobí exsikaci a svraštění tkáně, by se mohla stát jednoduchou alternativou minimálně invazivní terapie hemie disku. Autoři použili v pokusech na vaječném bílku a na sekčnim materiálu termistorovou elektrodu o průměru 2 mm a ploše 28,3 mm2, kterou na tkáň působili teplotou 95 *C po dobu 3 minut a dosahovali ,jednotkovou lézi'' o obsahu 1/3 ml v bílku a 1/2 ml v ne. pulposus. Posunem elektrody lze tuto Jednotkovou lézi " znásobit.
Electrothermocoagulation of the pulpous nucleus of the intervertebral disc, which causes exsiccation and wrinkling of tissue, could become a simple alternative of minimally invasive therapy of disc hernias. The authors used in experhnents on egg white and necroptic material a thermistor electrode with a diameter of 2 mm and area of 28.3 mm2 by means of which they acted on the tissue using a temperature of 95 'C for three minutes and obtained a „unit lesion" with a volume of 1/3 ml in egg white and 1/2 ml in the nc. pulposus. By shifting the electrode this „unit lesion" can be extended.
