The pathogenesis of non-alcoholic fatty liver disease (NAFLD) is associated with abnormalities of liver lipid metabolism. On the contrary, a diet enriched with n-3 polyunsaturated fatty acids (n-3-PUFAs) has been reported to ameliorate the progression of NAFLD. The aim of our study was to investigate the impact of dietary n-3-PUFA enrichment on the development of NAFLD and liver lipidome. Mice were fed for 6 weeks either a high-fat methionine choline-deficient diet (MCD) or standard chow with or without n-3-PUFAs. Liver histology, serum biochemistry, detailed plasma and liver lipidomic analyses, and genome-wide transcriptome analysis were performed. Mice fed an MCD developed histopathological changes characteristic of NAFLD, and these changes were ameliorated with n-3-PUFAs. Simultaneously, n-3-PUFAs decreased serum triacylglycerol and cholesterol concentrations as well as ALT and AST activities. N-3-PUFAs decreased serum concentrations of saturated and monounsaturated free fatty acids (FAs), while increasing serum concentrations of long-chain PUFAs. Furthermore, in the liver, the MCD significantly increased the hepatic triacylglycerol content, while the administration of n-3-PUFAs eliminated this effect. Administration of n-3-PUFAs led to significant beneficial differences in gene expression within biosynthetic pathways of cholesterol, FAs, and pro-inflammatory cytokines (IL-1 and TNF-α). To conclude, n-3-PUFA supplementation appears to represent a promising nutraceutical approach for the restoration of abnormalities in liver lipid metabolism and the prevention and treatment of NAFLD.

Obesity adversely affects bone and fat metabolism in mice and humans. Omega-3 polyunsaturated fatty acids (omega-3 PUFAs) have been shown to improve glucose metabolism and bone homeostasis in obesity. However, the impact of omega-3 PUFAs on bone marrow adipose tissue (BMAT) and bone marrow stromal cell (BMSC) metabolism has not been intensively studied yet. In the present study we demonstrated that omega-3 PUFA supplementation in high fat diet (HFD + F) improved bone parameters, mechanical properties along with decreased BMAT in obese mice when compared to the HFD group. Primary BMSCs isolated from HFD + F mice showed decreased adipocyte and higher osteoblast differentiation with lower senescent phenotype along with decreased osteoclast formation suggesting improved bone marrow microenvironment promoting bone formation in mice. Thus, our study highlights the beneficial effects of omega-3 PUFA-enriched diet on bone and cellular metabolism and its potential use in the treatment of metabolic bone diseases.

Eicosapentaenová kyselina (EPA) a docosahexaenová (DHA) jsou polynenasycené esenciální mastné kyseliny, které se do organismu savců dostávají výhradně zvnějšku při přirozené neschopnosti vlastní syntézy těchto látek. Jejich role je pasuje na něco jako „nutraceutika“, tj. látky přítomné i v běžné lidské stravě. Nejen přímé ovlivnění fungování buněk imunitního systému, ale i další mechanismy v metabolismu membrány buněk, genové transkripce a ovlivnění střevní mikroflory jsou procesy, které se podílejí na jejich farmakologicky definovatelných protizánětlivých a protektivních vlastnostech. Na tyto vlastnosti je pak přímo či nepřímo vázaná jejich antidepresivní účinnost, prokázaná prospektivními i randomizovanými studiemi.
Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) are polyunsaturated essential fatty acids that get in the body of mammals exclusively from the outside sources due to their natural inability to synthesize these substances themselves. Their role promotes them into something like "nutraceuticals", i.e. substances commonly present in the normal human diet. Direct influence on the functioning of cells of the immune system, but also other mechanisms in cell membrane metabolism, gene transcription and influence on the intestinal microflora are processes that participate in their pharmacologically definable anti-inflammatory and protective properties. Their antidepressant efficacy, proven by prospective and randomized studies, is directly or indirectly linked to these properties. Conversely, it is possible to demonstrate the simultaneous occurrence of inflammatory factors, characteristic changes in the metabolic profile and low omega-3 levels in depressive disorders.
Schizofrenie je závažná dlouhodobá psychotická porucha vyznačující se pozitivními i negativními symptomy, závažnými poruchami chování a deficitem kognitivních funkcí. Příčina této poruchy není zcela jasná, ale předpokládá se, že je multifaktoriální a zahrnuje jak dědičné, tak vnější faktory. Vzhledem k tomu, že lidský mozek reguluje veškeré chování, zaměřily se studie schizofrenie na identifikaci změn v neurobiologii a biochemii mozku. Mozek je orgánem nejvíce bohatým na lipidy (lipidy tvoří přibližně 50 % suché hmotnosti mozku). Výsledky četných studií prokázaly významný pokles polynenasycených mastných kyselin (PUFA), zejména kyseliny dokosahexaenové (DHA) v periferní krvi a také v mozku pacientů se schizofrenií v různých vývojových fázích poruchy. Nedostatek PUFA je spojován s psychotickými symptomy a kognitivními deficity u schizofrenie. Tato zjištění vedla k řadě klinických studií zkoumajících, zda by suplementace PUFA ve stravě mohla zlepšit průběh onemocnění u pacientů se schizofrenií. Výsledky jsou nekonzistentní, což lze přičíst zejména heterogenitě sledovaných pacientů. Zda se však, že pacienti s nízkými hladinami PUFA premorbidní, prodromálně nebo v časných stadiích schizofrenie by mohli mít z léčby PUFA prospěch. Každopádně je potřeba více kontrolovaných klinických studií.
Schizophrenia is a serious long-term psychotic disorder marked by positive and negative symptoms, severe behavioral problems and cognitive function deficits. The cause of this disorder is not completely clear, but is suggested to be multifactorial, involving both inherited and environmental factors. Since human brain regulates all behaviour, studies have focused on identifying changes in neurobiology and biochemistry of brain in schizophrenia. Brain is the most lipid rich organ (approximately 50 % of brain dry weight). Results from numerous studies have shown significant decreases of PUFAs, in particular, DHA as well as brain of schizophrenia patients at different developmental phases of the disorder. PUFA deficiency has been associated to psychotic symptoms and cognitive deficits in schizophrenia. These findings have led to a number of clinical trials examining whether dietary PUFA supplementation could improve the course of illness in patients with schizophrenia. Results are inconsistent. Some report beneficial whereas others show not effective. The discrepancy can be attributed to the heterogeneity of patient population. In this review, results from recent experimental and clinical studies, which focus on illustrating the role of PUFAs in the development of schizophrenia were examined. It suggests that patients with low PUFA levels before onset of disease, prodromally, or in the early stages of schizophrenia might benefit from lipid treatments, but more controlled clinical trials are warranted.
Cardiovascular disease (CVD) is a global public health issue due to its high morbidity, mortality, and economic impact. The implementation of innovative therapeutic alternatives for CVD is urgently required. Specialized proresolving lipid mediators (SPMs) are bioactive compounds derived from ω-3 and ω-6 fatty acids, integrated into four families: Lipoxins, Resolvins, Protectins, and Maresins. SPMs have generated interest in recent years due to their ability to promote the resolution of inflammation associated with the pathogeneses of numerous illnesses, particularly CVD. Several preclinical studies in animal models have evidenced their ability to decrease the progression of atherosclerosis, intimal hyperplasia, and reperfusion injury via diverse mechanisms. Large-scale clinical trials are required to determine the effects of SPMs in humans. This review integrates the currently available knowledge of the therapeutic impact of SPMs in CVD from preclinical and clinical studies, along with the implicated molecular pathways. In vitro results have been promising, and as such, SPMs could soon represent a new therapeutic alternative for CVD.

In the prevention and treatment of cardiovascular disease, in addition to the already proven effective treatment of dyslipidemia, hypertension and diabetes mellitus, omega-3 polyunsaturated fatty acids (n-3 PUFAs) are considered as substances with additive effects on cardiovascular health. N-3 PUFAs combine their indirect effects on metabolic, inflammatory and thrombogenic parameters with direct effects on the cellular level. Eicosapentaenoic acid (EPA) seems to be more efficient than docosahexaenoic acid (DHA) in the favorable mitigation of atherothrombosis due to its specific molecular properties. The inferred mechanism is a more favorable effect on the cell membrane. In addition, the anti-fibrotic effects of n-3 PUFA were described, with potential impacts on heart failure with a preserved ejection fraction. Furthermore, n-3 PUFA can modify ion channels, with a favorable impact on arrhythmias. However, despite recent evidence in the prevention of cardiovascular disease by a relatively high dose of icosapent ethyl (EPA derivative), there is still a paucity of data describing the exact mechanisms of n-3 PUFAs, including the role of their particular metabolites. The purpose of this review is to discuss the effects of n-3 PUFAs at several levels of the cardiovascular system, including controversies.

Chronic pain (CP) is a severe clinical entity with devastating physical and emotional consequences for patients, which can occur in a myriad of diseases. Often, conventional treatment approaches appear to be insufficient for its management. Moreover, considering the adverse effects of traditional analgesic treatments, specialized pro-resolving lipid mediators (SPMs) have emerged as a promising alternative for CP. These include various bioactive molecules such as resolvins, maresins, and protectins, derived from ω-3 polyunsaturated fatty acids (PUFAs); and lipoxins, produced from ω-6 PUFAs. Indeed, SPMs have been demonstrated to play a central role in the regulation and resolution of the inflammation associated with CP. Furthermore, these molecules can modulate neuroinflammation and thus inhibit central and peripheral sensitizations, as well as long-term potentiation, via immunomodulation and regulation of nociceptor activity and neuronal pathways. In this context, preclinical and clinical studies have evidenced that the use of SPMs is beneficial in CP-related disorders, including rheumatic diseases, migraine, neuropathies, and others. This review integrates current preclinical and clinical knowledge on the role of SPMs as a potential therapeutic tool for the management of patients with CP.

Aging is associated with development of disturbances of lipid and carbohydrate metabolism and with systemic pro-inflammatory state. The studies performed with elderly people suggests that these characteristics may be improved by physical activity and/or nutrition interventions. However, the knowledge of the mechanisms behind these positive changes in target organs (e.g. muscle, liver, or AT) is rather limited. EXODYA project should yield a novel and unique information on the exercise and Omega-3 induced alterations of the molecular characteristics of AT, i.e. adipogenic/lipogenic/lipolytic capacity of preadipocytes/adipocytes, and inflammatory profile of AT that have not been investigated to date. These variables will be analyzed in relation to the effects of exercise/omega-3 supplementation on physical performance, body composition and whole-body metabolic profile. Understanding the benefits of physical activity and diet quality in older age is central to considerations of future public health strategies to promote better physical function and health in later life.
Stárnutí je spojeno s rozvojem poruch metabolismu lipidů a sacharidů a rozvojem systémového prozánětlivého stavu. V jejich patogenezi hraje pravděpodobně významnou roli metabolická a imunitní dysfunkce tukové tkáně (TT). Mechanismy, které k tomuto rozvoji přispívají, však nejsou přesně známy. Projekt EXODYA by měl poskytnout nové unikátní znalosti o změnách v TT, týkajících se adipogeneze/ lipogenese/lipolýzy preadipocytů a adipocytů a zánětlivého profilu TT, v odpovědi na intervence do životního stylu seniorů. Tyto intervence budou sestávat ze 4 měsíčního kombinovaného aerobního plus dynamického silového tréninku a suplementace omega-3 nenasycenými mastnými kyselinami. Sledované molekulární a zánětlivé charakteristiky TT budou korelovány se změnami celkového metabolického stavu organismu, se změnami fyzického výkonu a složení těla. Výsledky ozřejmí roli TT v příznivých metabolických změnách navozených pohybovou aktivitou resp. nutriční suplementací a poskytnou podklad pro nová doporučení intervencí do životního stylu v populaci seniorů.
