The prion protein (PrPC) can be structurally shifted to its PrPScisoform causing a wide range of neurodegenerative diseases, which are currently incurable. There is an evidence that metallothioneins (MTs), and especially MT-3, are associated with neurodegenerative diseases. PrPCand MTs play pivotal roles in maintaining metal homeostasis; therefore, it is conceivable that each of them has its own significance in prion diseases. In this paper, we study the nature of interactions between PrPC, MT, and copper ions, Cu(II), using the method of differential pulse voltammetry (DPV) coupled with adsorptive transfer stripping technique (AdTS). Electrochemical properties of PrP itself and its interactions with both the Cu(II) ions and MTs have been found. Based on the results obtained, we hypothesised the formation of the complex in molar ratio 2:1 (PrPC:MT). Surface plasmon resonance imaging (SPRi) was used as a control reference assay to further confirm results obtained by the electrochemical approach, such as the specific interactions between PrPCand MT-3.

Many compounds can interact with DNA leading to changes of DNA structure as point mutation and bases excision, which could trigger some metabolic failures, which leads to the changes in DNA structure resulting in cancer. Oxidation of nucleic acid bases belongs to the one of the mostly occurred type of DNA damaging leading to the above mentioned phenomena. The investigation of processes of DNA oxidation damage is topical and electrochemical methods include a versatile and sensitive tool for these purposes. 8-hydroxydeoxyguanosine (8-OHdG) is the most widely accepted marker of DNA damage. Oxidative damage to DNA by free radicals and exposure to ionizing radiation generate several other products within the double helix besides mentioned oxidation products of nucleic acid bases. The basic electrochemical behaviour of nucleic acids bases on various types of carbon electrodes is reviewed. Further, we address our attention on description of oxidation mechanisms and on detection of the most important products of nucleic bases oxidation. The miniaturization of detector coupled with some microfluidic devices is suggested and discussed. The main aim of this review is to report the advantages and features of the electrochemical detection of guanine oxidation product as 8-OHdG and other similarly produced molecules as markers for DNA damage.

Elektrochemická analýza nukleových kyselin, bílkovin i polysacharidů představuje zajímavou, i když zatím méně používanou alternativu ve srovnání se stávajícími metodami založenými zejména na optické detekci. Nabízí totiž relativně levnou, rychlou a přístrojově nenáročnou možnost paralelní detekce na miniaturizovaných čipech, ideální pro personalizovanou medicínu 21. století. Elektrochemie nukleových kyselin umožňuje např. detekci konkrétních sekvencí DNA (pro určení genů, stanovení přítomnosti bakterií a virů, atd.), analýzu poškození DNA a interakcí s jinými molekulami, DNA metylaci, nebo detekci mikroRNA jako nádorových biomarkerů. V elektrochemii bílkovin je v současnosti kladen důraz zejména na konstrukci tzv. imunosenzorů schopných detekce konkrétních proteinů (antigenů) pomocí jejich vazby na protilátku, s potenciálem pro diagnostiku. Z biofyzikálního hlediska je zajímavý vlastní elektrokatalytický signál bílkovin citlivý k jejich konformačním změnám, který by mohl nalézt uplatnění při rozlišení mutantních forem proteinů (např. u p53) nebo při jejich agregaci (α-synuklein v případě Parkinsonovy nemoci), popřípadě při studiu interakcí s nízkomolekulárními látkami či DNA. Díky zvýšenému zájmu o glykoproteiny se začínají objevovat elektrochemické práce zabývající se detekcí oligo-sacharidů a polysacharidů (tzv. glykanů, pokud jsou součástí bílkovin), a to pomocí elektroaktivních značek specifických pro sacharidy anebo konstrukcí lektinových biosenzorů využívajících lektinů, které se silně váží právě na glykany. Elektrochemická analýza se tak jeví jako zajímavý nástroj v současném výzkumu na poli genomiky, proteomiky i glykomiky, včetně diagnostiky nádorových onemocnění.
Electrochemical analysis of nucleic acids, proteins and polysaccharides represents an interesting, although not widely spread alternative to current methods based predominantly on optical detection because it offers a relatively inexpensive, fast and instrumentally simple detection of parallel samples on miniaturized chips, ideal for personalized medicine of the 21st century. Nucleic acid electrochemistry enables, for example, detection of specific DNA sequences (for determination of genes or presence of bacteria and viruses, etc.), DNA damage analysis and interaction with other molecules, DNA methylation or detection of microRNAs as potential cancer biomarkers. In the electrochemistry of proteins, great emphasis is put on construction of immunosensors for capturing specific proteins (antigens) using antibodies, suitable for diagnostics. From a biophysical point of view, intrinsic electrocatalytic signal of proteins sensitive to conformational changes could be useful in discrimination of mutant proteins (e. g. p53), native and aggregated forms (α-synuclein in Parkinson‘s disease) or for studies of protein interactions with low molecular‑weight ligands and DNA. Due to an increased interest of scientists in glycoproteins, new electrochemical papers emerged aiming at detection of oligosaccharides and polysaccharides (i.e. glycans, when part of the protein). These assays employ for instance electroactive labels specific for saccharides or lectin biosensors using lectins which strongly bind glycans. Electrochemical analysis thus appears as an interesting tool in current genomics, proteomics and glycomics, as well as for cancer diagnostics. Elektrochemická detekce biomolekul se neustále posouvá a má velký potenciál využití v klinické praxi. V posledních letech je kladen důraz na přesné a rychlé stanovení biomarkerů na bázi nukleových kyselin – DNA a RNA. Značná část výzkumných projektů však nereflektuje požadavky na demonstraci vyvinutých metod na klinickém materiálu, což jejich aplikační potenciál značně snižuje. Klinický materiál vnáší do výzkumu oproti modelovým vzorkům větší variabilitu a práce s ním vyžaduje specifické podmínky. Problém získávání klinického materiálu pro výzkumné účely z velké části řeší banky biologického materiálu, které mohou nabídnout vzorky a data vhodné pro konkrétní výzkumný záměr.
Precise diagnostics of cancer or other diseases is crucial when selecting proper treatment. Personalized medicine puts high demands on the accuracy of nucleic acid biomarkers analysis, where subtle differences at the nucleotide level are often involved. Isothermal amplification techniques offer new possibilities of DNA and RNA amplification without using PCR, and their combination with electrochemistry provide a promising fast and cost-effective alternative diagnostic tool. Although electrochemical biosensors are still insufficiently applied to clinical material, thus hindering their development, recent advancements show great promise in translational research. Banks of biological material (biobanks) are specialized workplaces focused on the long-term preservation and processing of clinical material and offer a wide range of expert services, primarily for research purposes, in particular the provision of biological samples and associated pseudonymized data. Their involvement in the field of electrochemical biosensors can facilitate application of electrochemical methods into clinical laboratories and expand the portfolio of currently used diagnostic methods.

The aim of the study was to establish and optimize an electrochemical method for the assay of low-molecular-weight antioxidants (LMWA). We used square-wave vol¬tammetry (at 01.1 V) on screen-printed sensors with C working electrodes. Optimum parameters were: potential step and amplitude 5 mV and frequency 1 Hz. Ascorbic acid, trolox, N-acetylcysteine and melatonin were used as standard antioxidants to test the linearity and sensitivity of the method. Limits of detection were 0.09 mmol l1 for ascorbic acid, 0.04 mmol l1 for melatonin, 0.03 mmol l1 for trolox, and 0.07 mmol l1 for N-acetylcysteine. 20 ??????l samples were used. The suitability of the method for the assay was confirmed by the standard ferric reducing ability of plasma test. Both methods correlated well (correlation coefficient 0.9778).

From the rediscovery of graphene in 2004, the interest in layered graphene analogs has been exponentially growing through various fields of science. Due to their unique properties, novel two-dimensional family of materials and especially transition metal dichalcogenides are promising for development of advanced materials of unprecedented functions. Progress in 2D materials synthesis paved the way for the studies on their hybridization with other materials to create functional composites, whose electronic, physical or chemical properties can be engineered for special applications. In this review we focused on recent progress in graphene-based and MoS2 hybrid nanostructures. We summarized and discussed various fabrication approaches and mentioned different 2D and 3D structures of composite materials with emphasis on their advances for electroanalytical chemistry. The major part of this review provides a comprehensive overview of the application of graphene-based materials and MoS2 composites in the fields of electrochemical sensors and biosensors.
Ellipticine (5,11-dimethyl-6H-pyrido[4,3-b]carbazole) is an alkaloid that has been isolated from plants of an Apocynaceae family. It is one of the simplest naturally occurring alkaloids with a planar structure. Over the past decades, ellipticine became a very promising antitumor agent. Interaction with DNA is one of the most studied ellipticine effects on cell division. This phenomenon is not clearly explained so far. In our experiments we studied interaction of ellipticine with single-stranded and double-stranded oligonucleotides by electrochemical methods on mercury electrode. Differential pulse voltammetry was applied for ellipticine (Elli) and CA peak detection. Square wave voltammetry was applied for G peak detection. The effect of the interaction time and ellipticine concentrations on interactions of ellipticine with single- and double-stranded oligonucleotides was tested too.
