In our previous study, all Pseudomonas strains THP6, THP41, and OHP5 were identified as fluoride-resistant bacteria isolated from Dindigul district, Tamilnadu, India. The selected strains exhibiting a high level of fluoride resistance was determined in Luria broth (LB) medium and LB agar plates. In a further effort, fluoride-resistant organisms were tested for hemolytic activity and showed β-hemolysis on blood agar plates. The virulence factors such as gyrB, toxA, algD and lasB, plcH, rhlC and biofilm response genes (pslA, pelA, ppyR) were detected by PCR analysis. The putative genus-specific and species-specific PCR also confirmed that the selected fluoride-resistant strains were belonging to Pseudomonas aeruginosa species. Fluoride-resistance gene crcB was amplified by gene-specific primers. The crcB gene was cloned in TA vector and transformed into E. coli DH5α. Comparative and blast analysis of THP6, THP41, and OHP5 strains crcB gene sequences were high homology with P. aeruginosa fluoride efflux transporter crcB and P. aeruginosa putative fluoride ion transporter crcB. The recombinants were efficiently growing in the NaF containing LB agar plates. The fluoride tolerance of these strains was also associated with resistance to multiple antibiotics. These results can lead to the use of the fluoride resistance gene of P. aeruginosa for the development of a biosensor for fluoride detection.

Photodynamic therapy (PDT) has garnered immense attention as a minimally invasive clinical treatment modality for malignant cancers. However, its low penetration depth and photodamage of living tissues by UV and visible light, which activate a photosensitizer, limit the application of PDT. In this study, monodisperse NaYF4 :Yb3+ /Er3+ nanospheres 20 nm in diameter, that serve as near-infrared (NIR)-to-visible light converters and activators of a photosensitizer, were synthesized by high-temperature co-precipitation of lanthanide chlorides in a high-boiling organic solvent (octadec-1-ene). The nanoparticles were coated with a thin shell (≈3 nm) of homogenous silica via the hydrolysis and condensation of tetramethyl orthosilicate. The NaYF4 :Yb3+ /Er3+ @SiO2 particles were further functionalized by methacrylate-terminated groups via 3-(trimethoxysilyl)propyl methacrylate. To introduce a large number of reactive amino groups on the particle surface, methacrylate-terminated NaYF4 :Yb3+ /Er3+ @SiO2 nanospheres were modified with a branched polyethyleneimine (PEI) via Michael addition. Aluminum carboxyphthalocyanine (Al Pc-COOH) was then conjugated to NaYF4 :Yb3+ /Er3+ @SiO2 -PEI nanospheres via carbodiimide chemistry. The resulting NaYF4 :Yb3+ /Er3+ @SiO2 -PEI-Pc particles were finally modified with succinimidyl ester of poly(ethylene glycol) (PEG) in order to alleviate their future uptake by the reticuloendothelial system. Upon 980 nm irradiation, the intensive red emission of NaYF4 :Yb3+ /Er3+ @SiO2 -PEI-Pc-PEG nanoparticles completely vanished, indicating efficient energy transfer from the nanoparticles to Al Pc-COOH, which generates singlet oxygen (1 O2 ). Last but not least, NaYF4 :Yb3+ /Er3+ @SiO2 -PEI-Pc-PEG nanospheres were intratumorally administered into mammary carcinoma MDA-MB-231 growing subcutaneously in athymic nude mice. Extensive necrosis developed at the tumor site of all mice 24-48 h after irradiation by laser at 980 nm wavelength. The results demonstrate that the NaYF4 :Yb3+ /Er3+ @SiO2 -PEI-Pc-PEG nanospheres have great potential as a novel NIR-triggered PDT nanoplatform for deep-tissue cancer therapy.

Autism spectrum disorders (ASD) are characterized by impairments in social interaction and communication along with stereotyped patterns of behaviors. Over the past several decades, the prevalence of ASD has increased dramatically. ASD are highly multifactorial, with many risk factors acting together. Our review suggests that most risk factors are connected to immunoexcitotoxicity. Fluoride exposure is common as a result of the artificial fluoridation of drinking water and a dramatic increase in the volume of man-made industrial fluoride compounds released into the environment. Human exposure to environmental aluminum is extensive and appears to be growing. The long-term fluoride and aluminum burden have several health effects with a striking resemblance to the ASD. Moreover, both fluoride and aluminum interfere with a number of glycolytic enzymes, resulting in a significant suppression of cellular energy production. The synergistic interactions of fluoride and aluminum increase the potential neurotoxic effect particularly in children. Aluminofluoride complexes have effects on cell signaling, neurodevelopment, and neuronal function. We suggest that the burden with these new ecotoxicological factors could contribute to an alarming increase in the prevalence of ASD.

Fluorides and fluoroaluminates decrease mouse sperm fertilizing potential by modifying the process of sperm preparation for fertilization, so-called capacitation, followed by acrosome reaction (AR). Capacitation was monitored by protein tyrosine phosphorylation (pTyr), and AR was induced consequently. The aim of this study was to apply kinetic analysis to the previously obtained dependences of pTyr and AR at capacitation times, and propose a mathematical theory for a mechanism when sperm maturation ability is amended by external stimuli. The experimental input data, previously obtained, are consistent with the proposed theory and the results of kinetic analysis show that sperm capacitation runs as two subsequent first-order steps. Firstly, an unstable intermediate is formed and then gradually decomposes. The time corresponding to the maximal production of the unstable intermediate is probably most suitable for sperm obtaining the ability to fertilize the egg. The presented calculations indicate that the application of kinetic analysis can serve as a tool to predict or confirm a course of biological events that are modified by external factors, and therefore the proposed theory shall be of interest to a broad scientific audience.

Responses of adipose cells to adrenoceptor regulation, including that of β-adrenoceptor (AR), and the signalling machinery involved in these responses are not sufficiently understood; information that is helpful for elucidating the adrenoceptor (adrenergic and β-AR)-responsive machinery is insufficient. We examined phospho-Thr-172 AMPK production in mouse-derived 3T3-L1 adipocytes treated with epinephrine or CL316243 (a β3-AR agonist) for 15 min. We also examined MAPK activation or G protein-associated PI3K activation or -associated PI3K p85 complex formation in rat epididymal (white) adipocytes treated with CL316243 for 15 min or aluminum fluoride (a G-protein signalling activator) for 20 min. Furthermore, we examined the effect of PTX (a trimeric G-protein inactivator) on p85 complex formation induced by aluminum fluoride treatment. Western blot analysis revealed that epinephrine or CL316243 treatment increased the phospho- Thr-172 AMPK (an active form of AMPK) level in 3T3-L1 adipocytes. Activated kinase analysis with a specific substrate showed that CL316243 or aluminum fluoride treatment activated MAPK in rat adipocytes. Immunoprecipitation experiments with a G-protein β subunit (Gβ) antibody showed that treatment of rat adipocytes with CL316243 activated PI3K and increased the PI3K p85 level in the Gβ antibody immunoprecipitates. Such an increase in the p85 level was similarly elicited by aluminum fluoride treatment in a PTX-sensitive manner. Our results provide possible clues for clarifying the signalling machinery involved in adrenoceptor responses, including those of β3-AR, in mouse-derived adipocytes and rat white adipocytes. Our findings advance the understanding of responses to adrenoceptor regulation in adipose cells and of the cellular signalling machinery present in the cells.
