Arthroderma is the most diverse genus among dermatophytes encompassing species occurring in soil, caves, animal burrows, clinical material and other environments. In this study, we collected ex-type, reference and authentic strains of all currently accepted Arthroderma species and generated sequences of three highly variable loci (ITS rDNA, β-tubulin, and translation elongation factor 1-α). The number of accepted species was expanded to 27. One novel species, A. melbournense (ex-type strain CCF 6162T = CBS 145858T), is described. This species was isolated from toenail dust collected by a podiatrist in Melbourne, during an epidemiological study of four geographical regions of Eastern Australia. Trichophyton terrestre, Chrysosporium magnisporum, and Chrysosporium oceanitis are transferred to Arthroderma. Typification is provided for T. terrestre that is not conspecific with any of the supposed biological species from the former T. terrestre complex, that is, A. insingulare, A. lenticulare and A. quadrifidum. A multi-gene phylogeny and reference sequences provided in this study should serve as a basis for future phylogenetic studies and facilitate species identification in practice.LAY ABSTRACT: The genus Arthroderma encompasses geophilic dermatophyte species that infrequently cause human and animal superficial infections. Reference sequences from three genetic loci were generated for all currently accepted Arthroderma species and phylogeny was constructed. Several taxonomic novelties are introduced. The newly provided data will facilitate species identification and future taxonomic studies.

Cílem práce bylo zhodnotit molekulárně genetickými metodami (MGM) spektrum původců dermatofytóz na území České republiky s rozdělením podle hlavních klinických forem. Během dvou let (červenec 2011 až červen 2013) bylo na šesti regionálních pracovištích zachyceno 3235 kultivačně pozitivních vzorků s dermatofyty. Nejvyšší počet izolátů pocházel z případů tinea unguium (55,5 %), dále t. corporis (29,2 %), t. pedis (14,6 %) a t. capitis (0,7 %). Druhová identifikace izolátů všech druhů (n = 672) s výjimkou Trichophyton rubrum (n = 2 563) byla provedena MGM (PCR-fingerprinting nebo sekvenace ITS oblasti rDNA). Pro T. rubrum byla molekulárně ověřena jen identifikace morfologicky nejednoznačných izolátů (n = 189). Celkem bylo identifikováno 14 druhů dermatofytů, přičemž nejvýznamnější změnou oproti minulosti bylo vysoké zastoupení zoofilního druhu Arthroderma benhamiae, který působil velkou část případů t. corporis (22,9 %) a t. capitis (29,2 %). Geofilní druhy působily jen 1,3 % všech infekcí, přestože se jednalo o druhově nejpočetnější skupinu, ve které byly odhaleny dva nepopsané druhy a také přehlížené druhy Microsporum persicolor a M. fulvum. The aim of the study was to evaluate the spectrum of causative agents of the main clinical forms of dermatophytoses in the Czech Republic by molecular genetic methods (MGM). During two years (from July 2011 to June 2013), 3255 cultivation specimens were positive for dermatophytes. The highest number of specimens was isolated from tinea unguinum (55,5%), then tinea corporis (29,2%), tinea pedis (14,6%) and tinea capitis (0,7%). The identification of isolated species (n = 672) except for Trichophytom rubrum (n = 2563) was performed by MGM (PCR fingerprinting or sequencing of ITS segments of rDNA). For T. rubrum only morphologically non-clear isolates were identified (n = 189). In total, 14 species of dermatophytes were identified. The most important change noticed compared to previous results was an increased detection of Arthroderma benhamiae in the cases of tinea corporis (22,9%) and tinea capitis (29,2%). Geophilic species caused only 1,3% of all infections even if this group comprised the highest number of species with two newly described ones and also Microsporum persicolor and M. fulvum wich are usually overlooked in the morphological examination. Uvádíme kazuistiku kožní mykotické infekce typu tinea corporis u čtyřicetileté ženy. Ložisko infekce bylo lokalizované na hřbetu levé ruky a nereagovalo na úvodní lokální léčbu ciklopiroxolaminem. Změna léčby na systémovou terapii terbinafinem byla spojena s výskytem nežádoucích účinků a musela být ukončena. Ložisko bylo úspěšně vyléčeno kombinací lokálních antimykotik naftifinu a natamycinu. Jako původce infekce byl izolován zástupce geofilních dermatofytů Microsporum incurvatum, který je poměrně vzácně izolován z klinického materiálu a pravděpodobně žádný případ infekce zatím nebyl popsán v České republice. Identifikace izolátu byla ověřena sekvenací oblasti ITS rDNA. Vzhledem k morfologické podobnosti s druhy M. gypseum a M. fulvum je možné, že část infekcí přisuzovaných těmto druhům je ve skutečnosti působena právě M. incurvatum. Dále se v článku diskutuje o kontroverzní taxonomii druhu spolu s možnostmi, jak je možné ho rozlišit od příbuzných druhů. We present a case of mycotic infection of tinea corporis type in a forty-year old woman. The lesion was localised on the dorsal aspect of the left hand and did not react to ciclopirox olamine. The systemic therapy by terbinafine was stopped because of its side effects. The lesion was treated successfully with local antimycotics naftifine and natamycin. The geophillic dermatophyte Microsporum incurvatum was isolated and verified by the sequencing of ITS rDNA. This dermatophyte is rarely isolated from clinical material and probably no case was described in the Czech Republic. It is possible that some infections caused by M. incurvatum are attributed to M. gypseum and M. fulvum due to morphologic similarity of these species. The controversial taxonomy of this species and possibility of its differentiation from other species is discussed as well. V laboratořích klinické mikrobiologie jsou stále častěji využívány molekulárně genetické metody (MGM), a to nejen ke správné identifikaci izolovaného původce onemocnění, ale také k jeho přímé, časné detekci v klinickém materiálu. Také v dermatomykologii MGM vhodně doplňují a někdy dokonce nahrazují klasické, časově a odborně náročné diagnostické postupy, založené zejména na přímé mikroskopii a kultivaci. Výhodou MGM je především vysoká citlivost a rychlost detekce agens, které je možné prokázat v řádu několika hodin až dnů. To může napomoci časnému zahájení cílené léčby, nebo naopak zabránit zbytečnému zatěžování pacienta antimykotiky. MGM určené pro přímou detekci mikromycet nám však i přes značný pokrok stále nezaručují jejich přesnou identifikaci. K tomuto účelu je stále třeba patogen nejprve izolovat, poté je možné využít některých dalších MGM k jeho určení. Cílem tohoto přehledového článku je shrnout možnosti využití MGM pro přímou detekci dermatofytů z klinických vzorků, dále pro identifikaci druhů a typizaci kmenů. The molecular genetic methods (MGM) are used frequently in clinical microbiology to identify the agent of infection and also to its early direct detection in clinical specimens. In dermatomycology MGM supply or even substitute the classic time consuming and sophisticated diagnostic methods based on direct microscopy and cultivation. The high sensitivity and rapid agent detection available in few hours or days is an advantage of MGM that enables to start the early targeted therapy or to avoid an unnecessary antifungal treatment. However MGM designated to direct micromycetes detection does not guarantee their precise diagnostics which still requires the pathogen isolation before MGM are used to its identification. Article reviews the possibilities of MGM use in direct dermatophytes detection from clinical specimens and in species and strains identification. An undescribed Microsporum species was isolated from skin scales recovered from a 40-mm large, annular, scaling lesion on the wrist of a 46-year-old woman. The risk factors for dermatophyte infection in the patient were frequent work in the garden, hunting, and contact with dogs and horses. Direct microscopic examination of the scales revealed the presence of dermatophyte hyphae; when the samples were cultured, a morphologically similar fungus grew on all slants in pure culture. Both of these findings strongly suggested that the isolate was the true causal agent of infection. The possible geophilic nature of the species was based on phylogenetic analysis (internal transcribed spacer region of rDNA and β-tubulin gene) that placed it in between species of the M. gypseum complex. However, its divergencies from all other Microsporum species exceeded 4% base pairs. Based on β-tubulin phylogeny, the isolated species is a sister to M. gypseum. The species produces abundant chlamydospores and clumps of hyphae similar to those of ascomatal primordia but no conidia and ascospores. The species was unable to grow at 37°C and does not grow on T6 basal medium, which is unlike other Microsporum species; hair perforation and urease tests were positive. The addition of histidine to the T6 medium resulted in rapid growth of the fungus. The phylogenetic evidence, morphology, growth parameters, and physiology justified the proposal that the isolate is a new species, M. aenigmaticum, sp. nov.

The effect of ketoconazole (KK) on chosen biochemical indexes was studied in three mutants of dermatophyte Microsporum gypseum (MG-155 wild strain) resistant to KK. Fungistatic concentration of KK added to liquid cultivate medium increases significantly ergosterol pool and nonsignificantly demethylation activity, potassium content, cAMP release in three days old mycelium of mutants when compared to the cultivation without KK. On the contrary, there is a significant suppression of ergosterol level and demethylation activity followed by highly significant increasing of potassium content and cAMP release in the case of wild strain MG-155.
Authors describe the frequency and properties of mutants resistant to ketoconazole, obtained from the dermatophytes Microsporum gypseum and Trichophyton mentagrophytes. Spontaneous mutants occurred with the frequency of 10(-9) to spore and nucleus. Ultraviolet radiation increased the frequency of mutants by two numeric orders. The resistance level was relatively small in both types of mutants (RL within the range of 2-5).
The presented study deals with the properties of two wild strains of dermatophyte Microsporum gypseum and of eleven mutants resistant to ketoconazole. The growth rate of spontaneous mutants is greater than that of the wild strain, the group of UV-induced mutants manifests in general lower growth rate. The resistance level varies in the interval 1.1-1.6 (spontaneous mutants), resp. 2.7-5.5 (UV-induced mutants). The phenomenon of cross-resistance to other imidazole drugs (miconazole, clotrimazole) has been observed in mutants. There is further given the characteristic of ergosterol synthesis influence by the three used antifungal drugs. The data obtained are discussed from the point of view of possible biochemical processes resulting in the formation of resistance to ketoconazole.
The ketoconazole-resistant mutants were isolated from four strains of the dermatophyte Microsporum gypseum. The frequency of spontaneous mutants ranged within 10(-8) to 10(-9) (per spore and nucleus). The method was established for obtaining the spontaneous mutants manifested as the faster growing sectors. By means of mutagenic effect of UV radiation, the frequency of mutants increased by one order. The method used for determination of ergosterol the biosynthesis of which could be associated with the mechanism of resistance to ketoconazole is described and evaluated.
