The aim of this study was to assess two protocols for their capacities to simultaneously isolate RNA, mtDNA and ncDNA from mammalian cells. We compared the Invitrogen TRIzol-based method and Qiagen DNeasy columns, using the HepG2 cell line and human primary glioblastoma stem cells. Both methods allowed the isolation of all three types of nucleic acids and provided similar yields in mtDNA. However, the yield in ncDNA was more than tenfold higher on columns, as observed for both cell types. Conversely, the TRIzol method proved more reproducible and was the method of choice for isolating RNA from glioblastoma cells, as demonstrated for the housekeeping genes RPLP0 and RPS9.

Východiska: Monoklonální imunoglobuliny (paraproteiny) jsou produkovány B lymfocyty při lymfoproliferativním onemocnění. Jsou charakteristické svou homogenní strukturou, v séru se mohou vyskytovat ve vysokých koncentracích a mohou být příčinou významné interference při laboratorních stanoveních. Případ: 84letý pacient byl hospitalizován na interním oddělení pro progresi dušnosti. V základním laboratorním vyšetření jsme zaznamenali vysoký konjugovaný bilirubin a nízký celkový bilirubin. Přímý sérový bilirubin byl měřen spektrofotometrickou metodou na principu diazoreakce. Důkladné došetření navzájem si odporujících výsledků vedlo k odhalení paraproteinu jako příčiny laboratorní interference, a tím k diagnostice maligního lymfoproliferativního onemocnění, které u pacienta do té doby nebylo známo. Závěr: Monoklonální imunoglobuliny jsou relativně vzácnou, ale o to klinicky závažnější, příčinou interference. Tato kazuistika ukazuje nutnost bdělého profesionálního přístupu při hodnocení laboratorních výsledků. Nečekaně vysoké, nízké či navzájem si odporující výsledky měření by měly vést ke zvážení přítomnosti monoklonálního imunoglobulinu a vyloučení jeho přítomnosti dalšími metodami (elektroforéza, imunofixace). Důsledkem těchto interferencí mohou být chyby v diagnostice a neadekvátní léčba. Na druhou stranu u pacientů, u nichž je známa přítomnost paraproteinu ve vyšších koncentracích, je nutno myslet na možné interference u rozličných biochemických stanovení a být obezřetný při poskytování péče opírající se o jejich hodnocení.
Background: Monoclonal immunoglobulins (paraproteins) are produced by B lymphocytes in lymphoproliferative disorders. A single monoclonal immunoglobulin is homogeneous in terms of its structure, and it can occur in human serum at high concentration and cause significant interference in laboratory assays. Case: We present a case of an 84-year-old man who was admitted to the hospital for progression of dyspnea. Basic laboratory tests showed a serum concentration of conjugated bilirubin, measured using the diazo spectrophotometric method, which was much higher than that of total bilirubin. The cause of the discrepancy was attributed to analytical interference by monoclonal immunoglobulins, which helped establish a diagnosis of lymphoproliferative disorder. Conclusion: Monoclonal immunoglobulins are relatively rare in serum but are an important cause of analytical interference. Monoclonal immunoglobulins should always be considered a source of interference when unexpectedly high, low, or contradictory data are encountered, and appropriate confirmatory tests (electrophoresis, imunofixation) should be performed in such circumstances. Failure to do so can result in errors in diagnosis and inadequate treatment. Conversely, when samples contain abnormal and especially high monoclonal immunoglobulin levels, the biochemical data should be carefully examined for any discrepancies, such as paraprotein interference, and the results should be taken into consideration in patient management.

MicroRNAs (miRNAs) are evolutionarily conserved small non-coding RNAs that regulate expression of protein-coding genes involved in important biological processes and (patho)physiological states. Circulating miRNAs are protected against degradation, indicating their relevant biological functions. Many studies have demonstrated an association of the specific profile of circulating miRNAs with a wide range of cancers as well as non-malignant diseases. These findings demonstrate the implication of circulating miRNAs in the pathogenesis of diseases and their potential as non-invasive disease biomarkers. However, methods for measurement of circulating miRNAs have critical technical hotspots, resulting in a discrepancy of the reported results and difficult definition of consensus disease biomarkers that may be implicated in clinical use. Here, we review functions of circulating miRNAs and their aberrant expression in particular diseases. Further, we discuss methodological aspects of their detection and quantification as well as our experience with the methods.
Strains of genus Citrobacter (152 isolates from 1950 to 1988 deposited in the Czech National Collection of Type Cultures, Prague) were re-classified using biological and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) methods. One-hundred thirty-six strains (ca. 90 %) were identified to the species level using the biological method with evaluation by Farmer matrix. MALDI-TOF MS exhibited better identification capability, the data being more compact; the method was unambiguously successful in typing 145 (95 %) strains. Comparison of the results of identification by the two methods revealed differences (for 12 samples) in identified species which, considering all biochemical and/or MS characteristics, could be attributed to the natural variability of strains and close relation of the misidentified species (all of them belonged to the Citrobacter freundii complex). Taking into account all the above data, both methods can be considered reliable; however, the MALDI-TOF MS exhibits higher accuracy, efficiency, and rapidity.
Synthesis of base-modified oligonucleotides and DNA by polymerase incorporations of modified nucleoside triphosphates (dNTPs) is summarized. The crosscoupling synthesis of modified dNTPs, methods of their enzymatic incorporations, as well as their applications in bioanalysis and chemical biology are discussed.
