Endothelial cells (ECs) are primary targets of glucose-induced tissue damage. As a result of hyperglycemia, endothelin-1 (ET-1) is upregulated in organs affected by chronic diabetic complications. The objective of the present study was to identify novel transcriptional mechanisms that influence ET-1 regulation in diabetes. We carried out the investigation in microvascular ECs using multiple approaches. ECs were incubated with 5 mM glucose (NG) or 25 mM glucose (HG) and analyses for DNA methylation, histone methylation, or long non-coding RNA- mediated regulation of ET-1 mRNA were then performed. DNA methylation array analyses demonstrated the presence of hypomethylation in the proximal promoter and 5' UTR/first exon regions of EDN1 following HG culture. Further, globally blocking DNA methylation or histone methylation significantly increased ET-1 mRNA expressions in both NG and HG-treated HRECs. While, knocking down the pathogenetic lncRNAs ANRIL, MALAT1, and ZFAS1 subsequently prevented the glucose-induced upregulation of ET-1 transcripts. Based on our past and present findings, we present a novel paradigm that reveals a complex web of epigenetic mechanisms regulating glucose-induced transcription of ET-1. Improving our understanding of such processes may lead to better targeted therapies.

Diabetes has become one of the major diseases affecting human health. Diabetic cataracts (DCs) are considered a common complication in diabetic patients. The present study investigated differences in lens proteomic profiles between DCs and age-related cataracts (ACs) to determine the mechanism underlying the formation of DCs. Intrasurgical samples were collected from eight DC patients and 12 AC patients, and lens proteins were extracted by lysis and separated using two-dimensional gel electrophoresis (2-DE). The electrophoretic bands were analysed using PD-Quest software 8.0.1. Differentially expressed proteins were identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and peptide mass fingerprinting combined with protein database searching. In the 2-DE maps, the DC and AC lens proteins migrated in the region of pH 5-9 with a relative molecular weight (RMW) of 14-97 kDa, whereas the RMW of more abundant crystallin was 20-31 kDa. Approximately three protein spots with differential intensity were detected. Two crystallin proteins (αB and βB1) were identified using MALDI-TOF-MS. Proteomic analysis of the crystalline humour is feasible, and the proteins can be well separated; moreover, differentially expressed lens proteins can be analysed using 2-DE and mass spectrometry to compare DC and AC. The present results indicate that the αB and βB1 crystallins may accelerate the development of DCs. These techniques offer new avenues for mechanistic evaluation and future prevention or therapy of DCs.

BACKGROUND/AIMS: Calcium-Sensing Receptor (CaSR) significantly affects calcium-phosphate metabolism in kidneys, and it is implicated in the pathogenesis of diabetes mellitus (DM) due to its expression in pancreatic β-cells. The role of CaSR as one of the players in pathogenesis of chronic kidney disease (CKD) has been speculated. METHODS: 158 Type 2 diabetic patients divided into three groups according to occurrence and type of kidney complications, 66 nondiabetic patients CKD, and 93 healthy subjects were enrolled into the study to analyze the role of two CaSR polymorphisms (in the codon 990 and in the intron 4) in ethiopathogenesis of DM and CKD. The Type 2 diabetic groups consisted of 48 patients without any kidney abnormalities, 58 patients with diabetic nephropathy (DN), and 52 patients with nondiabetic renal disease (NDRD). The distribution of genotype and allele frequencies was studied using PCR with the TaqMan Discrimination Assay or followed by the Restriction Fragment Length Polymorphism method, respectively. RESULTS: We have found that the intron 4 polymorphism is a risk factor for the development of DM and CKD, except DN, while the codon 990 does not show any disease association. CONCLUSION: We conclude that CaSR is a general factor in pancreas and kidney pathologies.

Monogénový diabetes je typom cukrovky, kde genetická porucha je rozhodujúca pre vznik ochorenia. Podľa klinických príznakov sa rozdeľuje na cukrovku so skorým začiatkom a rodinným výskytom, familiárnu hyperglykémiu nalačno, diabetes s extrapankreatickými príznakmi a neonatálny diabetes. V posledných rokoch kontinuálne rastie počet génov, ktorých mutácie spôsobujú monogénový diabetes. Ich klinický obraz je veľmi heterogénny, preto je nutná DNA analýza na identifikáciu príčiny ochorenia, od ktorej sa odvíja aj liečba. Náš článok prináša prehľad súčasným poznatkov o monogénovej cukrovke so zameraním na klinicky a epidemiologicky najzávažnejšie formy.
Monogenic diabetes mellitus is a type of diabetes, where genetics without any other factors is strong enough to cause the disease. According to the clinical features monogenic diabetes can be divided to the mild familial early onset diabetes, familial fasting hyperglycemia, diabetes with extrapancreatic features and neonatal diabetes mellitus. During the last several years the number of genes causing monogenic diabetes has continuously increased. The clinical picture of the monogenic diabetes is very heterogeneous, thus DNA analysis is required for identification of the diabetes etiology, which influences also the choice of treatment. This article is an overview of current knowledge on monogenic diabetes, focusing at the clinically and epidemiologically most important forms.
Tuková tkáň je dnes považována za endokrinní orgán produkující řadu hormonů. Hormony tukové tkáně se významnou měrou podílejí na regulaci energetické homeostázy, inzulínové senzitivity a řady dalších procesů včetně podílu na vzniku diabetes mellitus 2. typu. Jedním z těchto hormonů je fatty acid binding protein-4 (FABP-4). Jde o cytoplazmatický protein o molekulové hmotnosti 14,7 kDa, který se podílí na transportu, skladování a metabolizmu lipidů. Mezi další funkce FABP-4 patří modulace signálních regulačních drah a buněčných enzymatických procesů. Jde o faktor, který je primárně produkován makrofágy a adipocyty. Experimentální a klinická data svědčí pro možnost, že změny sérových koncentrací FABP-4 mohou být významným etiopatogenetickým faktorem v rozvoji inzulínové rezistence, diabetu 2. typu a aterosklerózy. Cílem tohoto článku je podat aktuální přehled o FABP-4, jeho vztahu k výše uvedeným onemocněním a perspektivách využití modulace hladin tohoto hormonu v prevenci a léčbě diabetu 2. typu a jeho komplikací.
It is now generally accepted that adipose tissue is an endocrine organ producing numerous of hormones. These hormones play an important role in the regulation of energy homeostasis, insulin sensitivity and many other processes including its possible role in the development of type 2 diabetes mellitus. Fatty acid binding protein-4 (FABP-4) is a 14,7 kDa cytoplasmic protein that plays a role in lipid transport, storage and metabolism. Other functions of FABP-4 include modulation of signalling pathways and cell enzymatic proccesses. FABP-4 is primarily produced by macrophages and adipocytes. There is a growing number of experimental and clinical evidence suggesting that fatty acid binding protein-4 is an important ethiopathogenic factor in the developement of insulin resistance, type 2 diabetes and atherosclerosis. The aim of this article is to review the current state of knowledge about fatty acid binding protein-4 and its relationship to above mentioned diseases including perspectives of modulation of FABP-4 levels in the prevention and treatment of type 2 diabetes mellitus.

The project aims at determining the role of variability of connexin 37 gene in development of micro- and macroangiopathy in females with type 1 or type 2 diabetes mellitus. A secondary target is to determine the effect of menopause on the complications.Female patients (300-350) with type 1 diabetes mellitus and the same number of type 2 diabetes mellitus female patients will be examined. A cohort of 1 000 women from a representative sample of Czech population will be used as a control group. Clinical data and the presence of micro- or macroangiopathy will be assessed. The measurement of ankle-arm, toe-arm blood pressure ratios and intima/media thickness will determine the presence of subclinical atherosclerosis.
Cílem našeho projektu je stanovit úlohu variabity genu pro connexin 37 při vzniku mikro- a makroangiopatie u žen s diabetes mellitus 1.a 2. typu. Vedlejší cíl je stanovení vlivu menopauzy na tyto komplikace. Budou vyšetřeny pacientky s diabetes mellitus1. typu (300-350 osob) a diabetes mellitus 2. typu (300-350 osob). Kontrolní skupinou bude přibližně již 1 000 žen z reprezentativního vzorku české populace. U všech vyšetřených budou zjištěna základní klinická data (trvání a kompenzace diabetes mellitus, přítomnost mikro- či makroangiopatie), ultrazvukem a změřením poměru tlaků kotník/paže či palec/paže bude stanovena přítomnost subklinické aterosklerózy.
