A fattening experiment was carried out to analyze and evaluate the effect of ram lamb genotype on the growth, feed conversion (FC), and carcass value of Dhamari ram lambs, F1 crossbreds of Dhamari × Tehami (F1 DhT), and Tehami ram lambs. Genotype of the ram lambs including the experiment (n = 30) had high significant effect on total weight gain (TWG), average daily gain (ADG), and FC (P < 0.01-0.001). Genotype of the lamb had significant effect on hot and cold dressing percentage (P < 0.001). Also, genotype of the lamb had significant effect on the highest leg percentage (P < 0.001). Furthermore, genotype of the ram lambs had significant effect on musculus longissimus dorsi (MLD) area (P < 0.001). Genotype of the ram lamb had significant effect on TWG, ADG, and FC (P < 0.001). The results of moisture percentage, crude protein percentage, and ash percentage of muscle samples were significantly influenced by ram lamb's genotype (P < 0.001). Fasting live weight and other carcass measurements (hot carcass weight, leg weight, loin weight, shoulder weight, first quality cuts weight) have a positive and highest correlations (P < 0.001). The positive and the highest correlations were found between fasting live weight and hot carcass weight, first quality parts weight. In addition, the positive and the highest correlations were found between hot carcass weight and first quality cuts weight. In general, the results of this study documented that F1 DhT was better than Tehami ram lambs in ADG, TWG, and FC. Additionally, the results show that the feed conversion was in F1 DhT crossbreds ram lambs better than pure Dhamari and Tehami ram lambs, mainly in the carcass indicators.

Reactive oxygen species (ROS) are fundamental for intracellular signalling. In spermatozoa, they are involved both to apoptosis and to capacitation, and changes in ROS levels can alter the balance between these two processes. Oestrous sheep serum (OSS) is considered an efficient agent for in vitro capacitation of ram spermatozoa. We have explored the effects of OSS on ram sperm physiology, especially on ROS production, during in vitro capacitation. Semen samples from 15 rams were cryopreserved. After thawing, samples were submitted to four treatments: control (CTL), 10% OSS supplementation for in vitro sperm capacitation, caspase inhibitor (INH, Z-VAD-FMK 100 μM) and OSS (10%) plus caspase inhibitor (I + E). Sperm samples were incubated for 30 min at 38.5°C and 5% CO2 and evaluated motility and kinetic parameters by computer-assisted semen analysis (CASA) and viability (propidium iodide), apoptotic-like membrane changes (YO-PRO-1), acrosomal status (PNA-FITC), intracellular calcium (FLUO-3), membrane fluidity (M540) and ROS production (CM-H2 DCFDA) by flow cytometry. OSS induced changes in kinetic parameters compatible with capacitation, with a decrease in the percentage of progressive motility and linearity, and an increase in the amplitude of the lateral displacement of the sperm head (p < .05). Moreover, OSS increased the proportion of M540+ viable spermatozoa, YO-PRO-1+ and acrosome-reacted spermatozoa (p < .05). After incubation, OSS and I+E achieved lower ROS levels (p < .05). Ca(2+) levels did not change with the incubation, but were slightly higher (p < .05) when both OSS and the inhibitor were present. We suggest that OSS may modulate ROS levels, allowing intracellular signalling for capacitation to occur while preventing higher levels that could trigger apoptosis.

The aim of the study was to examine the effects of insulin-like growth factor I (IGF-I) on ram sperm traits after hypothermic storage. Sperm ejaculates from Lacaune rams were diluted in a Tris extender, pooled, divided into groups of IGF-I doses tested (0, 10, 100 or 200 ng.ml-1) and stored (0-5°C) for 96 h. IGF-I elevated whole sperm motility as measured by a Computer-assisted Sperm Analyser (CASA) system, by 24 h (10 ng.ml-1) and 48 h (200 ng.ml-1) of storage, and by progressive movement on each day of storage. After 72 h the sperm samples were analysed for plasma membrane integrity (peanut agglutinin-fluorescein isothiocyanate), membrane stability (annexin V-Fluos) and apoptosis (Yo-Pro®-1) using fluorescence microscopy. The addition of IGF-I (at 100 or 200 ng.ml-1) reduced the ratio of sperm with disrupted membranes and the ratio of annexin V-labelled sperm. The ratio of apoptotic sperm was reduced by IGF-I given at 10 or 100 ng.ml-1 compared with control. Sperm fertilizing ability, determined at 48 h by an in vitro fertilization (IVF) test on bovine oocytes, was increased by IGF-I given at 100 ng.ml-1 from 47.0 to 67.7%. In conclusion, IGF-I maintained ram sperm functions following cooling storage and its effects were reflected in sperm fertilizing ability in vitro.

The aim of this study was to determine the effect of Camellia sinensis extract as antioxidant supplement and pre-freezing equilibration times in a soybean lecithin extender for freezing ram semen. In this study, a total of 20 ejaculates were collected from four Ghezel rams and diluted with extenders (1.5% soybean lecithin, 7% glycerol) containing no supplements (control) and Camellia sinensis extract (5, 10, and 15 mg/L) and cryopreserved, immediately after thermal equilibrium was reached at 5 °C (0 h), or 4 h after equilibration. Sperm motility characteristics, membrane integrity, abnormal morphology, mitochondria activity, apoptotic status, MDA and antioxidant activities (GPx, SOD and total antioxidant capacity (TAC)) were evaluated following freeze-thawing. Camellia sinensis extract at level 10 mg/L led to the highest total and progressive motilities percentages, in comparison to other treatments (P < 0.05). Our results showed that Camellia sinensis extract at level of 5 and 10 mg/L led to higher plasma membrane integrity, mitochondria activity and Total antioxidant capacity (TAC) in comparison to the level of 15 mg/L and control group (P < 0.05). Camellia sinensis extract at 10 mg/L level produced the highest percentage of live spermatozoa and the lowest apoptotic spermatozoa in comparison to all treatments (P < 0.05). In addition, level of MDA formation significantly decreased at this concentration, 10 mg/L, compared to all treatments (P < 0.05). No differences (P > 0.05) were observed between equilibration times (0 h vs. 4 h) for sperm samples incubated with or without different concentrations of Camellia sinensis extract. In conclusion, addition of Camellia sinensis extract at level of 10 mg/L can improve post-thawing quality of ram semen cryopreserved in a soybean lecithin extender. However, further research is needed to standardize the process of Camellia sinensis extraction and specially for identifying which compounds are responsible of its beneficial effect on ram sperm cryopreservation.

Ve své knize nám Ram Dass pomáhá zkoumat radost, bolest i příležitosti dozrávajících období našeho života. Nabízí nám nový pohled na oblast, která leží před námi. Věří, že nám tento pohled usnadní bouřlivý proces stárnutí, změn a umírání. V naší společnosti je stárnutí hlubokým zdrojem utrpení, a proto začal pořádat semináře zabývající se vědomým stárnutím. Svůj soucit vyjadřoval tím, že lidem ukazoval, co dělat a jak přistupovat k vlastním zkušenostem. Tím otvíral cestu druhým, aby dospěli k objevení svého ducha. Všechno, co prožil, ho činilo neobyčejně šastným, a proto začal pořádat veřejné přednášky, psát knihy a natáčet audio a videokazety. V té době si uvědomil, jaké utrpení vyvolává strach ze smrti. Začal jsem se stýkat s lidmi, kteří umírali – včetně své matky, otce, nevlastní matky, pacientů trpících rakovinou a AIDS a mnoha dalších lidí. Všem těmto lidem nabídl své znalosti vyšších úrovní vědomí.
