Existuje čoraz viac dôkazov o zachytávaní každodenných zážitkov človeka v podobe intenzívnych longitudinálnych údajov získaných prostredníctvom denných zápiskov alebo metódy Experience sampling. Tieto údaje ponúkajú širokú škálu informácií o dynamike duševných procesov a správania, ich vývoja v čase a v rámci špecifických situácií. Cieľom tohto príspevku je predstaviť metódu zachytávania každodenných zážitkov (Experience Sampling Method – ESM) v kontexte výskumu duševného zdravia a prostredníctvom vybraných príkladov opísať možnosti, ako tieto údaje využiť v rámci nomotetického a idiografického prístupu a robenia záverov. Tento príspevok nie je vyčerpávajúcim opisom témy ESM, ale skôr stručným prehľadom čoraz viac využívanej metódy zachytávania každodenných zážitkov týkajúcich sa psychopatológie, jej limitov a perspektívy do budúcna.
There is growing evidence of capturing human experience in daily life using intensive longitudinal data obtained via daily diaries or experience sampling. These data offer a wide range of information regarding the dynamics of mental processes and behavior and how they unfold within individuals over time and within specific situations. The aim of this paper is to present the Experience Sampling Method (ESM) in the context of mental health research and through selective examples explain the possibilities of how to use data for nomothetic and idiographic inferences and conclusions. This paper is not an exhaustive description of the ESM topic, but rather a brief overview of the emerging methods for capturing daily experience regarding psychopathology, its limitations and future perspective.

The minimization is currently expanding in all fields of human life. The smaller size of analytical instruments and their higher sensitivity enables an analysis of small amounts of a biological material with a very low consumption of chemicals, what is economically and also environmentally benefi cial. A patient friendly and minimal invasive sample collection is therefore more than required. The dried blood spot (DBS) sampling standardly used in the newborn screening (NS) may be an option. The sample collection is simple, non-invasive, does not require trained medical personnel assistance and also storing and transportation of these samples is much easier in comparison with the whole blood samples. The DBS sampling method is used in the therapeutic drug monitoring or in the diagnosis of infectious diseases and its usage is still spreading. In some cases, it is still only a research object, but it shows a big potential for a future use. It could completely replace the whole venous blood collection in some cases, for example in the metabolic screening of diabetic patients or monitoring of treatment response, and it can overall simplify the sample collection and the transportation process (Tab. 1, Ref. 152).

For a better understanding of plant nutrition processes, it is important to study the flux of nutrients within plants. However, existing xylem sap sampling methods are typically destructive and do not allow for repeated, highly frequent measurements of nutrient concentration. In this paper, we present a novel use of microdialysis (MD) for characterizing xylem sap phosphate (PO43-) concentration as a possible alternative to destructive sampling. First, MD probes were tested under laboratory conditions in vitro, in a stirred solution test, and in vivo, using beech tree stem segments. Exponential decline in the relative recovery (RR) with an increasing MD pumping rate allows for determining an optimal sampling interval (i.e., the maximum amount of sample volume with the minimum required concentration). The RR changed only minimally, with a change in the simulated sap flow velocity during the in vivo stem segment test. This suggests that MD can be applied over a range of naturally occurring sap flow velocities. Differences in the ionic strength between the xylem sap and the perfusate pumped through the MD did not influence the RR. Then, MD was successfully applied in a 24 h field campaign in two beech trees of different ages and allowed for in situ assessments of the diurnal variation of PO43- concentration and (together with xylem flow measurements) flux variability in living trees. Both beech trees exhibited the same diurnal pattern in PO43- concentrations with higher concentrations in the younger tree. The xylem PO43- concentration measured with MD was in the same order of magnitude as that received through destructive sampling in the younger tree. The MD probes did not show a decline in RR after the field application. We showed that MD can be applied to capture the PO43- concentration dynamics in the xylem sap with bihourly resolution under field conditions.

Cíl: Vypracování maximálně bezpečné a komfortní techniky odběru plodové vody (dále AMC).Typ studie: Pro praxi.Název a sídlo pracovišť: Gyncentrum s.r.o., Praha 9, Hloubětínská 3, Oddělení lékařské genetikyGENNET, Praha 1, Klimentská 11.Metodika: U 81 pacientky jsme provedli AMC pomocí jehly o vnějším průměru 0,53 mm, za využitíodběrového systému Vacutainer firmy Becton-Dickinson.Závěr: Námi vypracovaná metodika se zdá být perspektivní k použití pro invazivní výkony v ob-lasti prenatální diagnostiky.
