The Purpose of present study was to investigate the effect of riboflavin/ultraviolet-A-induced collagen cross-linking (CXL) on central cornea, limbus and intraocular pressure (IOP). This was an animal experimental study. The right corneas of 10 rabbits were ultraviolet-A irradiated (3 mW/cm2 for 30 minutes) after de-epithelialization and instillation of 0.1% riboflavin / 20% Dextran drops. Left corneas served as controls. Samples were examined histologically one month postoperatively. Before and after treatment, IOP measurements were recorded bilaterally. At central cornea of eyes underwent CXL keratocyte repopulation, normal arrangement of collagen fibres and a statistically significant change in fibres diameter were detected, compared to controls. At limbus area, there were not any significant histological differences after CXL. There was no statistically significant difference between pre- and postoperative IOP in all eyes.

The aim of this study was to detect a spectrum of cytokeratins (CK) present in the adult human cornea, limbus and perilimbal conjunctiva. Cryosections from seven corneo-scleral discs were fixed, and indirect immunofluorescent staining was performed using antibodies directed against CK1-CK10 and CK13-CK20. The percentage of positive cells was calculated in the epithelium of the cornea, limbus and perilimbal conjunctiva. Quantitative real time RT-PCR (qRT-PCR) was used to detect CK6 and CK18 expression in the corneal and conjunctival epithelium. The most intense staining present throughout the cornea was observed for CK3, CK5 and CK14; CK19 was found at the corneal periphery only. CK4 and CK10/13 revealed mild to moderate positivity mostly in the superficial layers of the cornea. The suprabasal cell layers of all examined areas showed a strong positivity for CK16. A heterogeneous staining pattern with a centrifugal decrease in the signal was observed for CK8 and CK18. CK5/6, CK14 and CK19 were present in the limbus, where a positive signal for CK3 was observed in the suprabasal and superficial cells only. In contrast to the cornea, CK15 appeared in the basal and suprabasal layers of the limbus. The perilimbal conjunctiva showed strong immunostaining for CK10/13, CK14 and CK19. A moderate signal for CK7 was detected in the superficial layers of the conjunctiva. qRT-PCR confirmed CK6 and CK18 expression in the corneal and conjunctival epithelium. The detailed characterization of the corneal, limbal and perilimbal conjunctival epithelium under normal circumstances may be useful for characterizing the changes occurring under pathological conditions.

Poznanie konceptu o kmeňových limbálnych bunkách (KLB) otvorilo nové mož- nosti liečby ťažkých poleptaní predného segmentu. Tie spočívajú v transplantácii KLB, perforujúcej keratoplastike, eventuálne v transplantácii amniovej blany. U troch pacientov s ťažkým poleptaním predného segmentu oka sme vykonali transplantáciu KLB, perforujúcu keratoplastiku a u jedného aj transplantáciu amniovej blany. Kde bolo poškodené len jedného oko, KLB pre transplantáciu sme získali zo zdravého oka (autotransplantácia), pri poškodení oboch očí KLB boli v jednom prípade získané od blízkeho príbuzného a v druhomprípade z kadaverózneho oka z rohovkovej banky (allotransplantácia). U pacienta s autotransplantáciou KLB, perforujúcou keratoplastikou a transplantáciou amniovej blany bol výsledok veľmi dobrý. Rohovkový terč 1,5 roka po operácii zostal číry, spojovkový vak bol bez synblerofarónov a vízus sa zlepšil z pohybu ruky na 0,5. U dvoch pacientov s allotransplantáciou KLB a perforujúcou keratoplastikou, KLB sa neprihojili napriek imunosupresívnej liečbe a rohovkový terč sa zkalil. Obnovenie činnosti KLB pri ťažkých poleptaniach predného segmentu oka otvorili nové možnosti liečby.
The concept of limbal stem cells (LSC) opened new possibilities for the treatment of severe caustic damage of the anterior segment. These possibilities include transplantation of LSC, perforating keratoplasty, possibly transplantation of the amniotic membrane. In three patients with severe caustic damage of the anterior segment of the eye the authors implemented transplantation of LSC, perforating keratoplasty and in one patient also transplantation of the amniotic membrane. Where only one eye was damaged LSC for transplantation were obtained from the sound eye (autotransplantation, in case of damage of both eyes LSC were obtained in one case from a close relative in the second case from a cadaverous eye from a corneal bank (Allotransplantation). In the patient with autotransplantation of LSC, perforating keratoplasty and transplantation of the amniotic membrane the result was very good. The corneal disc remained clear 1.5 years after operation, the conjunctival sac was free from synbleropharons and vision improved from moveent of the hand to 0.5. In the two patients with allotransplantation of LCS and perforating keratoplasty, LCS did not become incorporated despite immunosuppressive treatment and the corneal disc became turbid. Restored activity of LCS in severe caustic damage of the anterior segment opened new therapeutic opportunities.

Cieľom tejto práce bolo identifikovať limbálne kmeňové bunky v kultúre ľudskej donorskej rohovky. Po štandardnom kultivačnom postupe sme na identifikáciu kmeňových buniek použili monoklónovú protilátku proti -enoláze ako biochemického markera kmeňových buniek. Imunofluorescenčným mikroskopom sme v zornom poli identifikovali -enoláza pozitívne bunky, ktoré in vitro nie však in situ vykazovali aj

Cieľomtejto experimentálnej práce je overiť významlimbálnych buniek pri hojení poleptanej rohovky. Predmetom experimentu boli angorské králiky. Poleptanie bolo realizované 2,5 N roztokom NaOH, ktorý pôsobil 50 sekúnd a následne bol vypláchnutý prúdom fyziologického roztoku, kontinuálne po dobu 5 minút. Eutanázia králikov bola urobená 24 hodín po poleptaní, tkanivo bolo fixované v 10% formalínovomroztoku. Pre histologické spracovanie bola použitá metódazaliatím do parafínu a vzorky boli ofarbené hematoxilínozínom. Na prezentovaných vzorkách možno pozorovať ložiskové zápalové reakcie s prítomnosťou makrofágov, v limbálnej oblasti s infiltráciou okrúhlojadernými bunkami.
The aim of this experimental study was to evaluate the role of limbal stem cells on corneal wound healing. The experimental objects were New Zealand white rabbits. Damage to the cornea was caused by 2,5 N NaOH solution for 50 sec. and was subsequently washed out by saline solution . The rabbits were euthanased 24 hours after alkaly burn and the tissue was fixated in 10% formalin. The samples wereembeddedinparaffin andstainedwithhaematoxilin andeosin. Thespecimen showed focal inflammatory reaction with the presence of macrophages an round - nucleated cells in the limbal area.
