The 3'-5', 3'-5' cyclic dinucleotides (3'3'CDNs) are bacterial second messengers that can also bind to the stimulator of interferon genes (STING) adaptor protein in vertebrates and activate the host innate immunity. Here, we profiled the substrate specificity of four bacterial dinucleotide synthases from Vibrio cholerae (DncV), Bacillus thuringiensis (btDisA), Escherichia coli (dgcZ), and Thermotoga maritima (tDGC) using a library of 33 nucleoside-5'-triphosphate analogues and then employed these enzymes to synthesize 24 3'3'CDNs. The STING affinity of CDNs was evaluated in cell-based and biochemical assays, and their ability to induce cytokines was determined by employing human peripheral blood mononuclear cells. Interestingly, the prepared heterodimeric 3'3'CDNs bound to the STING much better than their homodimeric counterparts and showed similar or better potency than bacterial 3'3'CDNs. We also rationalized the experimental findings by in-depth STING-CDN structure-activity correlations by dissecting computed interaction free energies into a set of well-defined and intuitive terms. To this aim, we employed state-of-the-art methods of computational chemistry, such as quantum mechanics/molecular mechanics (QM/MM) calculations, and complemented the computed results with the {STING:3'3'c-di-ara-AMP} X-ray crystallographic structure. QM/MM identified three outliers (mostly homodimers) for which we have no clear explanation of their impaired binding with respect to their heterodimeric counterparts, whereas the R2 = 0.7 correlation between the computed ΔG'int_rel and experimental ΔTm's for the remaining ligands has been very encouraging.

Článek shrnuje dosavadní poznatky o imunumodulačním efektu psychedelických látek a diskutuje jejich potenciál v terapii některých psychiatrických a neurologických onemocnění, v jejichž patogenezi se uplatňují imunologické vlivy. Jsou nastíněny molekulární mechanis-my účinku psychedelik, jejich interakce s imunitním systémem a neuropsychologické koreláty funkce imunity. Imunomodulační účinek, zejména zvýšená produkce protizánětlivých a snížená produkce prozánětlivých cytokinů, byl doposud prokázán u klasických psychedelik DMT, 5-MeO-DMT a LSD. Dále byl prokázán u disociativního anestetika ketaminu a empatogenu MDMA, které v určitých situacích vy-kazují podobnou fenomenologii jako psychedelika. Limitací je malé množství a převážně preklinická povaha prozatím provedených studií.
The article summarizes the main findings in the recent research of immunomodulatory effects of psychedelics and their potential in the treatment of neuropsychiatric diseases, where immunopathological processes are involved. Their molecular mechanism of action is presented with an emphasis on their interaction with the immune system and neuropsychological correlates of the immune system are also outlined. Increased production of anti-inflamatory and suppression of pro-inflammatory cytokines has been so far demonstrated in classical psychedelics such as DMT, 5-MeO-DMT and LSD. The findings are yet limited by the small amount of studies and by their predominantly preclinical nature.

Growth factors are powerful molecules that regulate cellular growth, proliferation, healing, and cellular differentiation. A delivery matrix that incorporates growth factors with high loading efficiencies, controls their release, and maintains bioactivity would be a powerful tool for regenerative medicine. Alginate has several unique properties that make it an excellent platform for the delivery of proteins. Mild gelling conditions can minimize the risk of protein denaturation; moreover, alginate can serve as protection from degradation until protein release. Various modifications have been proposed to tune alginate binding and release proteins, simultaneously adjusting alginate degradability, mechanical stiffness, swelling, gelation properties and cell affinity. The primary objective of this article is to review the literature related to recent advances in the application of alginate matrices in protein delivery in regenerative medicine. A special emphasis is put on the relevance of delivery of growth factors and chemokine.

This study aims to design an optimal polyelectrolyte multilayer film of poly-l-lysine (PLL) and hyaluronic acid (HA) as an anti-inflammatory cytokine release system in order to decrease the implant failure due to any immune reactions. The chemical modification of the HA with aldehyde moieties allows self-cross-linking of the film and an improvement in the mechanical properties of the film. The cross-linking of the film and the release of immunomodulatory cytokine (IL-4) stimulate the differentiation of primary human monocytes seeded on the films into pro-healing macrophages phenotype. This induces the production of anti-inflammatory cytokines (IL1-RA and CCL18) and the decrease of pro-inflammatory cytokines secreted (IL-12, TNF-α, and IL-1β). Moreover, we demonstrate that cross-linking PLL/HA film using HA-aldehyde is already effective by itself to limit inflammatory processes. Finally, this functionalized self-cross-linked PLL/HA-aldehyde films constitutes an innovative and efficient candidate for immunomodulation of any kind of implants of various architecture and properties.

The present study aimed to investigate the roles of fibrin deposition and protease activated receptor-1 (PAR-1) in renal cytokine/chemokine production and inflammatory cell infiltration in rats of different ages. Acute inflammation was induced by lipopolysaccharide (LPS) in rats which were then treated with tranexamic acid (TA), TA+urokinase (UK) or TA+low-molecular-weight heparin (HP). Fibrin deposition, inflammatory cells and expressions of PAR-1, monocyte chemoattractant protein-1 (MCP-1) and intercellular adhesion molecule 1 (ICAM-1) were detected. A reduction in fibrin deposition and PAR-1 expression in the LPS+TA+HP group was associated with decreased infiltration of inflammatory cells and down-regulated expressions of MCP-1 and ICAM-1. In the LPS+TA+UK group, the fibrin deposition, but not the PAR-1 expression, was reduced, However, the infiltration of inflammatory cells decreased and the expressions of MCP-1 and ICAM-1 down-regulated. There were significant differences in the fibrin deposition, infiltration of inflammatory cells and expression of PAR-1, MCP-1 and ICAM-1 between young and old rats undergoing the same treatment. These findings demonstrated that fibrin deposition plays more important roles than PAR-1 dose in cytokine/chemokine production and inflammatory cell infiltration in vivo, and ageing may deteriorate the fibrin deposition-induced production of cytokines/chemokines and infiltration of inflammatory cells.

Tento přehled se zabývá obrannými (antimikrobiálními) peptidy. Je zřejmé, že antimikrobiální peptidy aktivně participují na vše ch úrovních imunitní obrany a mají klíčovou modulační úlohu v mechanismech vrozené imunity. Přispívají také k regulaci adaptivní imunity.

The aim of the studies was to ascertain if adenosine is able to co-operate with selected hematopoietic growth factors and cytokines, namely with granulocyte colony-stimulating factor (G-CSF), granulocyte-macrophage colony-stimulating factor (GM-CSF), stem cell factor (SCF), interleukin-3 (IL-3), and interleukin-11 (IL-11), in inducing the growth of colonies from hematopoietic progenitor cells for granulocytes and macrophages (GM-CFC) from normal bone marrow cells in vitro. Adenosine was found not to produce any colonies when present in the cultures as the only potential stimulator. All the tested cytokines and growth factors were observed to induce the growth of distinct numbers of GM-CFC colonies, with the exception of IL-11. When suboptimal concentrations of the evaluated cytokines and growth factors were tested in the cultures in which various concentrations of adenosine were concomitantly present, mutually potentiating effects were found in the case of IL-3 and SCF. These results confirm the role of adenosine in regulation of granulopoiesis and predict IL-3 and SCF as candidates for further in vivo studies of their combined administration with adenosine.

Východisko. Mnohočetný myelom je neobyčejně heterogenní onemocnění s individuálně odlišným průběhem, léčebnou odezvou a prognózou. Významným nedostatkem dosavadních diagnostických a stratifikačních systémů je neuspokojivé zohlednění ukazatelů, vyjadřujících vnitřní biologické vlastnosti myelomových buněk a mikroprostředí kostní dřeně. Náplní studie bylo vyhodnocení vztahu 10 biologických ukazatelů k 6 stěžejním prognostickým faktorům mnohočetného myelomu. Metody a výsledky. Analyzovaný soubor tvořilo 66 jedinců vyšetřených při diagnóze před zahájením chemoterapie. Ke stanovení sérových hladin vyšetřovaných molekul byly použity metody REA, RIA, ELISA a technika kvantitativní sendvičové enzymatické imunoeseje, k vyšetření proliferačních a apoptotických vlastností myelomových buněk byl použit propidium–jodidový (PC-PI) a annexin–V (PC-AI) index vyšetřený s pomocí průtokové cytometrie. Statistická analýza byla provedena s pomocí Pearsonova a Spearmanova testu, a/nebo U-testu dle Manna–Whitneyeho. Vysoký výskyt abnormální sérové hladiny hodnoceného ukazatele byl zaznamenán v případě S-β2-mikroglobulinu (95,5 %), S-tymidinkinázy (57,5 %), S-sVCAM-1 (78,5 %), S-ICTP (87,0 %), S-solubilního osteoprotegerinu (sOPG 76,5 %), S-sSyndecanu-1 (56,5 %) a nízkého indexu apoptózy plazmatických buněk (PC-AI, 78,0 %). Korelační analýza (Pearsonův test) odhalila vzájemný vztah sérových hladin: β2-mikroglobulinu k sVCAM-1 (r=0,39, p=0,002), sICAM-1 (r=0,33, p=0,011), sOPG (r=0,53, p=0,001), sHGF (r=0,34, p=0,006), sSyndecanu-1 (r=0,38, p=0,003) a sFas (r=0,42, p=0,001); S-albuminu k sVCAM-1 (r= -0,29, p=0,036), ICTP (r= -0,33, p=0,016), sOPG (r= -0,63, p=0,000), sHGF (r= -0,39, p=0,003) a sSyndecan-1 (r= -0,29, p=0,042); S-tymidinkinázy k sSyndecanu-1 (r=0,46, p=0,000) a sFas (r=0,29, p=0,019). V žádném případě nebyl zjištěn vztah k některému z hodnocených prognostických faktorů. Bylo zjištěno chybění vztahu všech analyzovaných působků k PC-PI a PC-AI. S pomocí U-testu byl zjištěn vztah sérových hladin sIL-6R (< > 100 IU/l) k β2-mikroglobulinu (p=0,001), albuminu (p=0,002) a k PC-PI (p=0,046). Závěry. Z předložené studie vyplynulo, že tradiční algoritmus ukazatelů, sloužících v klinické praxi k individuální charakteristice MM je možno rozšířit o sOPG, sHGF, sSyndecan-1 a sFas.
Background. Multiple myeloma is an unusually heterogeneous disease with individually different clinical course, sensitivity to therapy and prognosis. The undoubted limitation of contemporary diagnostic stratification systems is the insufficient credit of those parameters that express the intrinsic biological properties of myeloma cells and the microenvironment of the bone marrow. The aim of this study was to evaluate the relationship of 10 biological parameters to 6 fundamental prognostic factors of multiple myeloma. Methods and Results. The analysed group consisted of 66 individuals examined at the time of diagnosis before the start of therapy. For the estimation of serum levels of investigated molecules there were used following methods: REA RIA, ELISA and the technique of quantitative sandwich enzyme immunoassay. For the analysis of proliferative and apoptotic properties of myeloma cells we used propidium iodide (PC-PI) and annexin-V (PC-AI) indices. The statistical estimation was carried out with the help of Pearson and Spearman test, eventually with the help of U-test according to Mann–Whitney. Increased incidence of abnormal serum levels of examined parameter was registered in the case of S-β2-microglobulin (95.5 %), S-thymidinekinase (57.5 %), S-sVCAM-1 (78.5 %), S-ICTP (87.0 %), S-solubile osteoprotegerin (sOPG 76.5 %), S-sSyndecan-1 (56.5 %) and low index of plasma cell apoptosis (PC-AI, 78.0 %). The statistical analysis (Pearson’s test) revealed the mutual relationship of serum levels of: S-β2-microglobulin to sVCAM-1, (r=0.39, p=0.002), sICAM-1 (r=0.33, p=0.011), sOPG (r=0.53, p=0.001), sHGF (r=0.34, p= 0.006), sSyndecan-1 (r=0.38, p=0.003) and sFas (r=0.42, p=0.001); S-albumin to sVCAM-1 (r= -0.29, p=0.036), ICTP (r= -0.33, p=0.016), sOPG (r= -0.63, p=0.000), sHGF (r= -0.39, p=0.003), sSyndecan-1 (r= -0.29, p=0.042); S-thymidinekinase to sSyndecan-1 (r=0.46, p=0.000) and sFas (r=0.29, p=0.019). There was no zjišrelationship of PINP and VEGF to any of the evaluated prognostic factors. There was no relationship found between either of the analysed parameters to PC-PI and PC-AI. With the help of U-test there was found the relationship of sIL-6R to S-β2-microglobulin (p=0.001), albumin (p=0.002) and PC-PI (p=0.046). Conclusions. The presented study implies that the traditional algorithm of parameters used in clinical practice for individual characteristics of MM should be enriched with sOPG, sHGF, sSyndecan-1 and sFas.
