This study was aimed to analyse the lower limb kinematics during the change of direction (COD) performance with the dominant (DL) and non-dominant (NDL) leg using linear (traditional kinematics) and nonlinear (Self Organising Map-based cluster analysis) approaches. Three 5-0-5 COD performances with the DL and three with the NDL were performed by 23 (aged 21.6 ± 2.3 years) collegiate athletes. No significant difference was observed between the COD duration, and approach speed of DL and NDL. Significantly greater ankle abductions, knee and hip external rotations were identified in COD with DL, compared to NDL (p < .001, d > 0.8). Self Organising Maps portrayed a completely different coordination pattern profile during change of direction performance with the DL and NDL. The cluster analysis illustrated similar inter-individual coordination patterning when participants turned with their DL or NDL. No visible relationship was observed in the cluster analysis of the lower limb joint angles and angular velocities. Outcomes of this study portrayed that coordination patterning (combination of joint angles and the rate of change of angles) could portray the movement patterning differences in different tasks, while a sole investigation on the joint angles or angular velocities may not reveal the underlying mechanisms of movement patterning.

Alterations in the excitability of dorsal root ganglion (DRG) neurons are critical in the pathogenesis of acute and chronic pain. Neurotransmitter release from the terminals of DRG neurons is regulated by cannabinoid receptor 1 (CB1) and transient receptor potential vanilloid 1 (TRPV1), both activated by anandamide (AEA). In our experiments, the AEA precursor N-arachidonoylphosphatidylethanolamine (20:4-NAPE) was used to study the modulation of nociceptive DRG neurons excitability using K+-evoked Ca2+ transients. Intrathecal administration was used to evaluate in vivo effects. Application of 20:4-NAPE at lower concentrations (10 nM - 1 μM) decreased the excitability of DRG neurons, whereas the higher (10 μM) increased it. Both effects of 20:4-NAPE were blocked by the N-acylphosphatidylethanolamine phospholipase D (NAPE-PLD) inhibitor LEI-401. Similarly, lower concentrations of externally applied AEA (1 nM - 10 nM) inhibited DRG neurons, whereas higher concentration (100 nM) did not change it. High AEA concentration (10 μM) evoked Ca2+ transients dependent on TRPV1 activation in separate experiments. Inhibition of the CB1 receptor by PF514273 (400 nM) prevented the 20:4-NAPE- and AEA-induced inhibition, whereas TRPV1 inhibition by SB366791 (1 μM) prevented the increased DRG neuron excitability. In behavioral tests, lower 20:4-NAPE concentration caused hyposensitivity, while higher evoked mechanical allodynia. Intrathecal LEI-401 prevented both in vivo effects of 20:4-NAPE. These results highlight anti- and pro-nociceptive effects of 20:4-NAPE mediated by CB1 and TRPV1 in concentration-dependent manner. Our study underscores the complexity of endocannabinoid signaling in pain transmission modulation and highlights 20:4-NAPE as a potential therapeutic target, offering new insights for developing analgesic strategies.

The behavior of chemical warfare agents (CWAs) on urban materials, such as concrete, significantly impacts forensic and military responses to chemical incidents. This study examined the persistence and degradation mechanisms of sarin (GB), soman (GD), and sulfur mustard (HD) on three types of commonly used concrete with varying water-cement ratios. Over two months, we evaluated the effects of concrete composition, temperature, and fragment size on CWA behavior. Half-lives and activation energies for CWA dissipation were calculated under various conditions. Results showed that concrete properties and external temperature strongly influenced dissipation rates. G-series agents underwent rapid hydrolysis, forming methylphosphonates, while HD degradation involved elimination, nucleophilic substitution, and oxidation, producing several previously unreported byproducts. Smaller concrete fragments increased recovery values and accelerated degradation due to greater surface area exposure, and higher temperatures further enhanced dissipation rates, particularly for volatile agents. Differences in dissipation among concrete types were linked to their physical and chemical properties, notably water-cement ratios. This study highlights the challenges of detecting CWAs due to their rapid penetration and transformation in concrete and provides insights for improving sampling, identification, and decontamination strategies under realistic conditions.

BACKGROUND: This study qualitatively investigates retirement-age adults' perspectives on engaging in health behaviors such as physical activity or a healthy diet, distinguishing facilitators, barriers, goals, and motivations (the two later in line with Self-Determination Theory). METHODS: Two clinical psychologists conducted four focus groups with Spanish adults around retirement age. We conducted inductive and deductive content analysis. RESULTS: The main facilitators and barriers identified were the presence and absence of social support/social network, mental health, willpower, time, and motivation. Participants reported different types of motivation (e.g., intrinsic motivation in the enjoyment of the activity of exercise or cooking) and goals (intrinsic and extrinsic); except for the goal of health management, which presented both types of motivation, participants regulated intrinsic goals autonomously, and extrinsic ones with controlled motivation. A process of internalizing the source of motivation was identified inductively by participants. CONCLUSIONS: Facilitating social networks and addressing mental health issues could aid engagement in health behaviors among this population. Additionally, health management appeared as a significant goal, where autonomous motivation can develop even if the behavior initially arises from controlled motivation or external triggers, such as medical advice.

In recent years, CM has become increasingly popular in the pharmaceutical industry for the production of OSD forms. Most of the newly developed APIs nowadays are extremely cohesive and sticky with a mean particle size particle of <100 μm, a wide PSD and a high tendency to agglomerate, making them difficult to accurately dose using loss-in-weight equipment during CM. In this research paper, the effect of various glidants on the volumetric and gravimetric feeding of several APIs was assessed. Three challenging API (APAPμ, MPT and SD) and four different glidants (Aerosil® 200, Aerosil® R972, Syloid® 244FP and TRI-CAFOS® 200-7) were selected. For all feeding trials, a GEA CF equipped with 20 mm concave screws was used, in combination with an external catch scale. The volumetric feeding trials showed the ability of each glidant to increase the FFmax and reduce the FFmovRSD40 and the FFdecay for the cohesive APIs (APAPμ and MPT). Although the fumed silica grades showed the highest impact on the previously mentioned feeding parameters, low AE10 values were obtained, negatively affecting the feeding performance at higher glidant concentration. Both Syloid 244FP and TCP were good alternatives. However, to obtain a similar feeding performance a higher concentration of these glidants is required. The volumetric trials showed that glidant addition has no additional benefits for APIs with good flow properties such as SD. The second part of this paper discussed the impact of glidant addition on the gravimetric feeding behavior of the cohesive powders. Both the fumed silica grades (Aerosil® 200 and Aerosil® R972) and Syloid 244FP lowered the deviation on all LC% profiles of the cohesive APIs. In contrast to the volumetric trails, blends with excess fumed silica resulted in low AE10 values which are efficiently dosed by the CF during the gravimetric feeding.
OBJECTIVE: The aim of this qualitative study is to develop a conceptual model for the school life of the children with epilepsy based on grounded theory methodological framework. METHODOLOGY: A total of 20 children with epilepsy participated in the semi-structured interviews. Inclusion criteria were age of 8-15 years, IQ higher than 70, duration of epilepsy of at least of two years and attendance of mainstream schools. 15 children had never seizure in school and 16 of them have not had a seizure in last 12 months. Qualitative research data was processed based on grounded theory with open coding leading consecutively to several main categories, axial coding establishing connections between individual categories, and finally selective coding resulting in one central category which was the core of the developed conceptual model. RESULTS: Eight main categories were identified during open coding: type of epilepsy and its management, sources of stress at school, adaptive responses to stressors, safe school environment in relation to (a) epilepsy and (b) learning, family support, stability of experience and behavior, and positive self-concept. Axial and selective coding resulted in establishment of Adaptive responses to stressors as the central category which is in the developed conceptual model determined by relevant external and internal factors as well as the sources enabling effective adaptation. CONCLUSIONS: The presented research emphasize the importance of adaptive responses to stressors in the school environment among children with epilepsy which may be useful in counselling focused on how to avoid or successfully manage possible traumatizing experience related with epilepsy in this group.

Starnutie človeka je prirodzený, nepretržitý a nezvratný proces, pri ktorom dochádza k zmenám organizmu v závislosti od pôsobenia mnohých vonkajších a vnútorných faktorov. Otázky starnutia a postavenia seniorov v spoločnosti sa stávajú trvale aktuálnymi v rovine ekonomickej, psychologickej, etickej, kultúrnej, zdravotno‐sociálnej, ale aj filozofickej. Ide tu o potrebu cieľavedome podporovať zdravie, ale tiež zabezpečiť choré a závislé osoby, o potrebu zlepšiť ich zdravotný stav a možnosti fungovania v spoločnosti, o ponúknutie prozdravotných programov a o snahu predĺžiť ich aktívny vek.
Human aging is a natural, continuous, and irreversible process, during which changes in the organism occur depending on the action of many external and internal factors. The issues of aging and the position of seniors in society are becoming permanently relevant at the economic, psychological, ethical, cultural, health-social, and philosophical levels. This is about the need to promote health purposefully, but also to provide for sick and dependent people, about the need to improve their health status and the possibilities of functioning in society, about offering health programs, and about the effort to extend their active age.
Stereotypie jsou opakující se, rytmické, zdánlivě neúčelné pohybové vzorce. Primární stereotypie jsou typicky popisovány u zdravých dětí s normálním vývojem, stereotypie se spouštěčem v dospělosti jsou výrazně vzácnější. Prevalence primárních stereotypií se udává mezi 3 až 8 %. Sekundární stereotypie se objevují buď v důsledku senzorické deprivace, sociální izolace, nebo jako součást širší symptomatiky u mnoha, především neurovývojových, onemocnění, u psychiatrických onemocnění nebo jako součást polékových syndromů. V genezi stereotypií se zvažují jak neurobiologické mechanismy včetně genetické dispozice, tak vlivy zevního prostředí a sociálních interakcí. Základním terapeutickým postupem je edukace a behaviorální terapie, u farmakologických postupů bohužel scházejí randomizované, dvojitě zaslepené studie.
Stereotypies are repetitive, rhythmic, seemingly purposeless movement patterns. Primary stereotypies are typically described in healthy children with normal development, stereotypies with a trigger in adulthood are significantly rarer. The prevalence of primary stereotypies is reported between 3 and 8 %. Secondary stereotypies appear either as a result of sensory deprivation, social isolation or as part of broader syndroms in many, especially neurodevelopmental diseases, in psychiatric diseases or drug-induced syndromes. In the genesis of stereotypies, both neurobiological mechanisms, including genetic disposition, and the effects of the external environment and social interactions are considered. The basic therapeutic procedure are education and behavioral therapy, unfortunately there are no randomized, double-blind studies for pharmacological procedures.
Antagonistic activity of brain networks likely plays a fundamental role in how the brain optimizes its performance by efficient allocation of computational resources. A prominent example involves externally/internally oriented attention tasks, implicating two anticorrelated, intrinsic brain networks: the default mode network (DMN) and the dorsal attention network (DAN). To elucidate electrophysiological underpinnings and causal interplay during attention switching, we recorded intracranial EEG (iEEG) from 25 epilepsy patients with electrode contacts localized in the DMN and DAN. We show antagonistic network dynamics of activation-related changes in high-frequency (> 50 Hz) and low-frequency (< 30 Hz) power. The temporal profile of information flow between the networks estimated by functional connectivity suggests that the activated network inhibits the other one, gating its activity by increasing the amplitude of the low-frequency oscillations. Insights about inter-network communication may have profound implications for various brain disorders in which these dynamics are compromised.

INTRODUCTION: Loneliness has a rising public health impact, but research involving neuropathology and representative cohorts has been limited. METHODS: Inverse odds of selection weights were generalized from the autopsy sample of Rush Alzheimer's Disease Center cohorts (N = 680; 89 ± 9 years old; 25% dementia) to the US-representative Health and Retirement Study (N = 8469; 76 ± 7 years old; 5% dementia) to extend external validity. Regressions tested cross-sectional associations between loneliness and (1) Alzheimer's disease (AD) and cerebrovascular pathology; (2) five cognitive domains; and (3) relationships between pathology and cognition, adjusting for depression. RESULTS: In weighted models, greater loneliness was associated with microinfarcts, lower episodic and working memory in the absence of AD pathology, lower working memory in the absence of infarcts, a stronger association of infarcts with lower episodic memory, and a stronger association of microinfarcts with lower working and semantic memory. DISCUSSION: Loneliness may relate to AD through multiple pathways involving cerebrovascular pathology and cognitive reserve. HIGHLIGHTS: Loneliness was associated with worse cognition in five domains. Loneliness was associated with the presence of microinfarcts. Loneliness moderated cognition-neuropathology associations. Transportability methods can provide insight into selection bias.
