The aim of this study was to develop and validate methods for the determination of vitamins B2, B9, E and A in serum using liquid chromatography with mass spectrometry (MS) detection. Vitamin analysis was performed using an ultra performance liquid chromatography combined with tandem MS. The compounds were separated on a BEH C18 RP column (2.1 × 100 mm, 1.7 μm) using a gradient elution with an analysis time of 10 min. Sample preparation included protein precipitation with ethanol. The concentration range in human serum was as follows: riboflavin 5-1000 nmol/L, folic acid 2.5-250 nmol/L, α-tocopherol 0.5-100 μmol/L and all-trans-retinol 25-2500 nmol/L. Accuracy and precision were validated according to Food and Drug Administration guidelines, with coefficients of variation ranging from 3.1-11.7% and recoveries from 94.4-107.5%. Routine monitoring of the complex range of vitamins in bariatric medicine is still not common. This is despite the fact that patients are at risk for glitch deficits, especially of a neurological nature. An analytical method that allows for the complex measurement of both water-soluble and fat-soluble vitamins is important and necessary for the clinical monitoring of bariatric patients. The method we have described could benefit both clinical practice and nutritional research.

INTRODUCTION: Our study aim was to assess how the macronutrient intake during total parenteral nutrition (TPN) modulates plasma total free fatty acids (FFAs) levels and individual fatty acids in critically ill patients. METHOD: Adult patients aged 18-80, admitted to the intensive care unit (ICU), who were indicated for TPN, with an expected duration of more than three days, were included in the study. Isoenergetic and isonitrogenous TPN solutions were given with a major non-protein energy source, which was glucose (group G) or glucose and lipid emulsions (Smof lipid; group L). Blood samples were collected on days 0, 1, 3, 6, 9, 14, and 28. RESULTS: A significant decrease (p < 0.001) in total FFAs occurred in both groups with a bigger decrease in group G (p < 0.001) from day 0 (0.41 ± 0.19 mmol∙L-1) to day 28 (0.10 ± 0.07 mmol∙L-1). Increased palmitooleic acid and decreased linoleic and docosahexaenoic acids, with a trend of increased mead acid to arachidonic acid ratio, on day 28 were observed in group G in comparison with group L. Group G had an insignificant increase in leptin with no differences in the concentrations of vitamin E, triacylglycerides, and plasminogen activator inhibitor-1. CONCLUSION: Decreased plasma FFA in critically ill patients who receive TPN may result from increased insulin sensitivity with a better effect in group G, owing to higher insulin and glucose dosing and no lipid emulsions. It is advisable to include a lipid emulsion at the latest from three weeks of TPN to prevent essential fatty acid deficiency.

The diversity in human diets that can be reached by proper use of different crops and varieties, including some underutilized ones, is a potentially powerful strategy to ensure food security and prevent serious health problems caused by current diets that are often not fulfilling nutritional requirements. In the framework of this research, the content of tocopherols and tocotrienols, thiamine, riboflavin, pyridoxine, and superoxide dismutase in nine varieties of quinoa, both colored and nonpigmented, obtained from 4 different countries, was investigated and compared to the content of the same vitamins and antioxidants in barley and wheat, both colored and nonpigmented, cultivated in the same experimental field. The aim of this work was to create a crop diversity strategy and encourage the consumption of underutilized crops to ensure that the human diet fulfills nutritional requirements. The contents of vitamin B1, B2, B6, tocopherol, and tocotrienol isomers and superoxide dismutase were determined via HPLC; imaging techniques were used to evaluate the seed color. Quinoa grains had the greatest concentration of tocopherol isomers and activity, represented mainly by α-tocopherol and γ-tocopherol. Wheat and barley seeds had substantial concentrations of tocopherols and tocotrienols. The concentration of riboflavin was greater in barley and wheat than in quinoa, the concentrations of pyridoxine and thiamine were variety-dependent in all grains. Quinoa grains had greater concentration of superoxide dismutase compared to wheat and barley. The richness of each variety and crop should be recognized and used integrally to improve the diet quality. PRACTICAL APPLICATION: Nutritional potential of crops was evaluated from the viewpoint of selected vitamins and antioxidants to create a well-balanced diet. Combined use of both traditional (wheat, barley) and underutilized crops (quinoa) is recommended. HPLC methods and image analysis were successfully used as viable tools for food quality determination.

A high-throughput miniaturized liquid-liquid extraction procedure followed by a simple ultra-high performance liquid chromatography method coupled with fluorescence detection for bioanalytical analysis of all tocopherol isomers and retinol in human serum has been developed and validated. In the extraction procedure, a synthetic internal standard tocol was used, which does not occur in the human body. The separation of structurally related vitamins was achieved using a new generation of pentafluorophenyl propyl core-shell stationary phase with elution using methanol and an aqueous solution of ammonium acetate. The fluorescence of retinol and tocopherol isomers was detected at λex = 325, 295 nm and λem = 480, 325 nm, respectively. The rapid baseline separation of all analytes was accomplished within 4.0 min. The sensitivity of method was demonstrated with lower limits of quantification: retinol 0.01 μM, α-tocopherol 0.38 μM, β-tocopherol 0.18 μM, γ-tocopherol 0.14 μM, and δ-tocopherol 0.01 μM. Possible application of this method in clinical practice was confirmed by the analysis of human serum samples from healthy volunteers. Finally, the simultaneous determination of retinol and all tocopherol isomers in human serum can enable the clarification of their role in metabolism and in diseases such as cancer.
AIM: To study inflammatory response and nutritional biomarkers in operated endometrial cancer (EC) patients. MATERIALS AND METHODS: A total 109 consecutive EC patients undergoing open laparotomy (LT), laparoscopic (LS) or robot-assisted surgery (RS) were studied. Twenty four patients served as controls. Pre- and postoperative levels of inflammatory and nutritional biomarkers were analyzed prospectively. RESULTS: The estimated blood loss was significantly lower in RS compared to all other groups. C-reactive protein (CRP) and interleukin-6 (IL-6) correlated with each other and exhibited positive correlation with age, body-mass index (BMI), leukocyte count, platelet count, kynurenine, kynurenine/tryptophan ratio and urinary neopterin and a negative correlation with vitamin D and retinol. Hemoglobin, retinol, alpha-tocopherol, vitamin D and citrulline concentrations decreased and inflammatory biomarkers increased after surgery to a different extent in LT, LS, RS and control groups. CONCLUSION: The present data demonstrate a differential response to surgical trauma in patients with endometrial carcinoma.

Dosažení patologické kompletní odpovědi po neoadjuvantní chemoterapii je ukazatelem dlouhodobé kontroly onemocnění. V předchozím projektu byla patologická kompletní odpověď pozorována významně častěji u nemocných s nádory expresí HER-2 a bez exprese HER-2 i hormonálních receptorů. Cílem předkládaného projektu je studovat biologickou odpověď na neoadjuvantní chemoterapii. Použity budou standardní režimy chemoterapie používané v jednotlivých centrech a volbu léčby projekt neovlivní. V retrospektivní studii na materiálu nemocných z 2 center budou vyhodnoceny rozdíly tumor infiltrujících lymfocytů u nemocných s různým fenotypem nádoru, bude také studován vliv exprese indoleamin 2,3-dioxygenázy na přítomnost tumor infiltrujících lymfocytů a bude provedeno imunohistochemické zhodnocení fenotypu nádoru. Budou rovněž sledovány změny metabolismu železa a citrulin, ukazatel toxicity léčby.; Pathologic complete response is an indicator of long-term disease control after neoadjuvant chemotherapy in breast carcinoma. In our earlier study, patologic complete response was observed significantly more often in patients with tumors expressing HER-2 or triple negative tumors. The aim of the present study is to study biologic response to neoadjuvant chemotherapy in patients treated with standard regimens that will not be affected by the project. In a retrospective study on material from patients in 2 centers difference in tumor infiltrating lymphocytes in patients with different tumor phenotype as well as the association between indoleamine 2,3-dioxygenase expression and the presence of tumor infiltrating lymphocytes will be evaluated together with tumor phenotype using immunohistochemistry. Alterations of iron metabolism and citrulline, an indicator of intestinal toxicity, will also be assessed.

Vitamin E comprises eight related compounds: α-, β-, γ-, δ-tocopherols and α-, β-, γ-, δ-tocotrienols. In the past, α-tocopherol has been the isomer that was studied most, and its anti-inflammatory and anti-proliferative effects have been described. Therefore, many prevention trials have investigated the effect of α-tocopherol on human health. Current research studies have also defined the important roles of other tocopherols, such as anti-inflammatory, anti-proliferative and cancer preventative effects. Knowledge of the individual tocopherols could help to understand their roles in various metabolic pathways. This review summarizes the recent trends in sample pretreatment, liquid chromatography and selected applications of the determination of tocopherols in various biological materials. The relationship between tocopherol isomers and serious diseases is also described. Graphical Abstract Article structure.

Oxidative stress has been proposed as one of the potential causes for infertility in men. Retinol and α-tocopherol have an important role in the spermatozoa defences against oxidative stress. A method is described here for the simultaneous determination of retinol and α-tocopherol in human seminal plasma with a suitable sample preparation procedure to prevent retinol and α-tocopherol degradation. After adequate sample preparation, the samples were determined by reversed-phase column chromatography with UV detection. The analytical performance of this method was satisfactory. The intra-assay and inter-assay coefficients of variation were below 10%. The recoveries were as follows: 90.7% (CV 8.1%) for retinol and 98.2% (CV 4.8%) for α-tocopherol. No significant differences in both retinol and α-tocopherol concentration between the smokers and nonsmokers (15 ± 7 nm and 1.86 ± 0.29 μm versus 15 ± 6 nm and 1.93 ± 0.45 μm) were found. A selective high-performance liquid chromatography method for the determination of retinol and α-tocopherol in human seminal plasma was developed.

The aim of the study was to establish and optimize an electrochemical method for the assay of low-molecular-weight antioxidants (LMWA). We used square-wave vol¬tammetry (at 01.1 V) on screen-printed sensors with C working electrodes. Optimum parameters were: potential step and amplitude 5 mV and frequency 1 Hz. Ascorbic acid, trolox, N-acetylcysteine and melatonin were used as standard antioxidants to test the linearity and sensitivity of the method. Limits of detection were 0.09 mmol l1 for ascorbic acid, 0.04 mmol l1 for melatonin, 0.03 mmol l1 for trolox, and 0.07 mmol l1 for N-acetylcysteine. 20 ??????l samples were used. The suitability of the method for the assay was confirmed by the standard ferric reducing ability of plasma test. Both methods correlated well (correlation coefficient 0.9778).
