OBJECTIVE: To identify intracerebral sites activated after correct motor response during cognitive task and to assess associations of this activity with mental processes. METHODS: Intracerebral EEG was recorded from 205 sites of frontal, temporal and parietal lobes in 18 epileptic patients, who responded by button pressing together with mental counting to target stimuli in visual oddball task. RESULTS: Post-movement event-related potentials (ERPs) with mean latency 295 ± 184 ms after movement were found in all subjects in 64% of sites investigated. Generators were consistently observed in mesiotemporal structures, anterior midcingulate, prefrontal, and temporal cortices. Task-variant nonspecific and target specific post-movement ERPs were identified, displaying no significant differences in distribution among generating structures. Both after correct and incorrect performances the post-performance ERPs were observed in frontal and temporal cortices with latency sensitive to error commission in several frontal regions. CONCLUSION: Mesiotemporal structures and regions in anterior midcingulate, prefrontal and temporal cortices seem to represent integral parts of network activated after correct motor response in visual oddball task with mental counting. Our results imply equivalent involvement of these structures in task-variant nonspecific and target specific processes, and suggest existence of common nodes for correct and incorrect responses. SIGNIFICANCE: Our results contribute to better understanding of neural mechanisms underlying goal-directed behavior.

Background/Objective: In the last decade, socio-political violence in Colombia (South America) has created an environment of extreme/chronic stress. In this study, brain imaging technology (fMRI) and behavioral task performance were used to measure potential deficits in executive functioning for emotional processing in Colombian children. Method: Participants (22 Post-Traumatic Stress Disorder, PTSD and 22 neurotypical, NT) were asked to perform a word task with implicit emotional salience, which required them to report the color of the ink in which a positive, negative or neutral word was printed. Results: Mixed design analysis of variance showed no group differences in accuracy for determining ink color when presented as a positive or neutral word. However, PTSD children were significantly less accurate (negative words) and notably slower (both positive and negative words) at determining ink color when presented in the context of an emotional word. PTSD processing of positive and negative words was associated with hypoactivation in the superior and middle frontal gyri of the right hemisphere in comparison to NT children. Conclusions: These results may reflect a deficit in executive functioning for emotionally laden stimuli, perhaps induced as a by-product of their traumatic experiences.

Cyclin-dependent kinases (CDKs) are key regulators of the cell cycle and RNA polymerase II mediated transcription. Several pharmacological CDK inhibitors are currently in clinical trials as potential cancer therapeutics and some of them also exhibit antiviral effects. Olomoucine II and roscovitine, purine-based inhibitors of CDKs, were described as effective antiviral agents that inhibit replication of a broad range of wild type human viruses. Olomoucine II and roscovitine show high selectivity for CDK7 and CDK9, with important functions in the regulation of RNA polymerase II transcription. RNA polymerase II is necessary for viral transcription and following replication in cells. We analyzed the effect of inhibition of CDKs by olomoucine II on gene expression from viral promoters and compared its effect to widely-used roscovitine. We found that both roscovitine and olomoucine II blocked the phosphorylation of RNA polymerase II C-terminal domain. However the repression of genes regulated by viral promoters was strongly dependent on gene localization. Both roscovitine and olomoucine II inhibited expression only when the viral promoter was not integrated into chromosomal DNA. In contrast, treatment of cells with genome-integrated viral promoters increased their expression even though there was decreased phosphorylation of the C-terminal domain of RNA polymerase II. To define the mechanism responsible for decreased gene expression after pharmacological CDK inhibitor treatment, the level of mRNA transcription from extrachromosomal DNA was determined. Interestingly, our results showed that inhibition of RNA polymerase II C-terminal domain phosphorylation increased the number of transcribed mRNAs. However, some of these mRNAs were truncated and lacked polyadenylation, which resulted in decreased translation. These results suggest that phosphorylation of RNA polymerase II C-terminal domain is critical for linking transcription and posttrancriptional processing of mRNA expressed from extrachromosomal DNA.

The quality of data measured in in vivo MR spectroscopy is often insufficient due to a number of limitations such as low concentrations of observed metabolites and restricted measurement time resulting in a low signal-to-noise ratio. However, there are a variety of methods called post-processing techniques which allow the enhancement of the measured signal after measurement. In this review an introduction to the most important post-processing techniques for (1)H MR spectroscopy is given and practical examples are shown. In the first section the concept of FID and spectrum is introduced and the relationship between FID and spectrum is explained. Subsequently, the objectives and description of the following post-processing techniques are provided: eddy current correction, removal of an unwanted component (water), signal filtering for various purposes, zero filling, phase correction and baseline correction.

Collagen is the most abundant protein in the animal and human bodies, and it is not exempt from this aging phenomenon. Some age-related changes may appear on collagen sequences, such as increased surface hydrophobicity, the appearance of post-translational modifications, and amino acids racemization. This study has shown that the protein hydrolysis under deuterium conditions is privileged to limit the natural racemization during the hydrolysis. Indeed, under the deuterium condition, the homochirality of recent collagens is preserved whose amino acids are found in their L-form. However, in aging collagen, a natural amino acid racemization was observed. These results confirmed that the % d-amino acids are progressive according to age. The collagen sequence is degraded over time, and a fifth of the sequence information is lost during aging. Post-translational modifications (PTMs) in aging collagens can be a hypothesis to explain the modification of the hydrophobicity of the protein with the decrease of hydrophilic groups and the increase of hydrophobic groups. Finally, the exact positions of d-amino acids and PTMs have been correlated and elucidated.

Glukokortikoidy jsou protizánětlivá léčiva, která ovlivňují expresi genů, transkripci v jádře, posttranskripční a posttranslační mechanismy v cytoplazmě. Aktivita prozánětlivých proteinů je regulována i posttranskripčními mechanismy, které ovlivňují stabilitu mRNA. Pochopení nových signálních drah regulujících protizánětlivou aktivitu glukokortikoidů je cestou k dosažení maximální léčebné odpovědi v organismu.
Glucocorticoids are anti-inflammatory drugs that regulate gene expression, nuclear transcription and post-transcriptional and post-translational mechanisms in the cytoplasm. The activity of the anti-inflammatory proteins is also regulated by the post-transcriptional mechanisms that control mRNA stability. Understanding new signalling pathways involved in the regulation of anti-inflammatory activity of glucocorticoids is prerequisite for achieving the best possible therapeutic response.

ABSTRACT: In dystonic and spastic movement disorders, abnormalities of motor control and somatosensory processing as well as cortical modulations associated with clinical improvement after botulinum toxin A (BoNT-A) treatment have been reported, but electrophysiological evidence remains controversial. In the present observational study, we aimed to uncover central correlates of post-stroke spasticity (PSS) and BoNT-A-related changes in the sensorimotor cortex by investigating the cortical components of somatosensory evoked potentials (SEPs). Thirty-one chronic stroke patients with PSS of the upper limb were treated with BoNT-A application into the affected muscles and physiotherapy. Clinical and electrophysiological evaluations were performed just before BoNT-A application (W0), then 4 weeks (W4) and 11 weeks (W11) later. PSS was evaluated with the modified Ashworth scale (MAS). Median nerve SEPs were examined in both upper limbs with subsequent statistical analysis of the peak-to-peak amplitudes of precentral P22/N30 and postcentral N20/P23 components. At baseline (W0), postcentral SEPs were significantly lower over the affected cortex. At follow up, cortical SEPs did not show any significant changes attributable to BoNT-A and/or physiotherapy, despite clear clinical improvement. Our results imply that conventional SEPs are of limited value in evaluating cortical changes after BoNT-A treatment and further studies are needed to elucidate its central actions.
