Over the course of history, human beings have never stopped seeking effective methods for information storage. From rocks to paper, and through the past several decades of using computer disks, USB sticks, and on to the thin silicon "chips" and "cloud" storage of today, it would seem that we have reached an era of efficiency for managing innumerable and ever-expanding data. Astonishingly, when tracing this technological path, one realizes that our ancient methods of informational storage far outlast paper (10,000 vs. 1,000 years, respectively), let alone the computer-based memory devices that only last, on average, 5 to 25 years. During this time of fast-paced information generation, it becomes increasingly difficult for current storage methods to retain such massive amounts of data, and to maintain appropriate speeds with which to retrieve it, especially when in demand by a large number of users. Others have proposed that DNA-based information storage provides a way forward for information retention as a result of its temporal stability. It is now evident that DNA represents a potentially economical and sustainable mechanism for storing information, as demonstrated by its decoding from a 700,000 year-old horse genome. The fact that the human genome is present in a cell, containing also the varied mitochondrial genome, indicates DNA's great potential for large data storage in a 'smaller' space.

In this study, the stability of 124 target analytes in influent and effluent wastewater samples during short-term (4°C) and long-term (-18°C) storage was assessed. The most common storage scenario was considered, in which samples were frozen immediately after sampling without any pre-treatment. During short-term storage more analytes remained stable (concentration during storage was in the range of 60-120% of the initial concentration) at 4°C than at -18°C. During long-term storage (-18°C), three types of behavior were observed: constant concentrations throughout the experimental period, decreasing concentrations with time, and loss of the compound from the sample after freezing. Differences between effluent and influent samples were observed for 50 out of 124 tested PPCPs. The amount of stable analytes decreased with time during long-term storage. 72% and 56% of the target compounds in the effluent and influent wastewater, respectively, remained stable during 60days of storage. The number of stable compounds decreased to 57 (46%) and 46 (37%) in the effluent and influent, respectively, over 120days. 15 Pharmaceuticals were lost after freezing/thawing cycle. The results stress the importance of storage factors during analysis of pharmaceuticals in wastewater. The stability of target compounds in the samples under the planned storage conditions should be checked before starting the experiment to obtain reliable data.

Deficit lyzozomální kyselé lipázy se manifestuje ve dvou prognosticky odlišných klinických variantách: maligní infantilní varianta (Wolmanova nemoc) a relativně benigní forma, která je známa jako nemoc ze střádání esterů cholesterolu (CESD). V kazuistice prezentujeme dívku s hepatomegalií, poruchou růstu, neprospíváním a hyperlipidémií. Podezření na CESD bylo potvrzeno histologickým vyšetřením jater a výrazným snížením aktivity enzymu kyselé lipázy v leukocytech. U pacientky byla ve dvou letech věku zahájena léčba lovastatinem, inhibitorem 3-hydroxy-3-methylglutaryl koenzym A reduktázy. V průběhu pěti let trvající léčby klesla hladina cholesterolu, triglyceridů a LDL cholesterolu. Váha a výška se upravily a odpovídají průměrným hodnotám pro daný věk. Hladina HDL cholesterolu se nezměnila. Sérové hladiny transamináz v průběhu léčby kolísaly a velikost jater se při ultrasonografickém vyšetření nezměnila. Léčba lovastatinem probíhá bez klinických komplikací. Prognóza jaterního postižení zůstává však do budoucna nejasná. CESD patří do diferenciální diagnostiky hepatomegalie od kojeneckého věku až po dospělé pacienty. Léčba lovastatinem je dobře tolerovaná, snižuje hladiny tuků, ale vliv statinů na progresi jaterního postižení zůstává nejasný.
Lysozomal acid lipase deficiency is manifested in two prognostically different clinical variants: malignant infantile form (Wolman's disease) and relatively benign form known as a cholesteryl ester storage disease (CESD). In our case report a girl with liver enlargement, growth retardation, asthenia and hyperlipidaemia is presented. Suspition to CESD was confirmed by liver histology and enormous decreasing activity of acid lipase in white blood cells. In that patient in her second year of age a treatment with lovastatin, which is inhibitor of 3-hydroxy 3-methylglutaryl koenzym A reductase was started. During five years of treatment the serum levels of cholesterol, triglycerides and LDL cholesterol decreased. Her weight and height normalised to mean values for a given age. The level of LDL cholesterol has not changed. Serum transaminases activity varied during treatment and liver enlargement has not changed by ultrasound. The treatment with lovastatin was without any side effect in this time. The prognosis of liver disorder for future is not clear. CESD should be concerned to diferenciál diagnosis of liver enlargement from infant time till adults patients. Treatment with lovastatin has been tolerated well and was able to lower the serum lipid levels, but influence of statins to liver disorder progression is not clear.

Due to the increasing popularity of unfiltered beer, new methods for its preservation are needed. High-pressure processing (HPP) was applied as a final treatment of packed beer in order to assure storage stability and to retain the desired product quality. Pressures of 250 MPa and 550 MPa for 5 min were used to process unfiltered lager beers. The impact of pressure on basic analytical characteristics was evaluated, and foam stability, the content of carbonyl compounds and sensory properties were monitored during two months of storage. Most of the basic analytical parameters remained unaffected after pressure treatment, and a beneficial effect on foam stability was demonstrated. Changes in the concentration of staling aldehydes were observed during storage. Some features of the sensory profile were affected by HPP as well as by the time of storage. Our study evaluated the suitability of HPP as a novel method for shelf-life extension of unfiltered lager beer.
Dental pulp stem cells (DPSCs) are a type of easily accessible adult mesenchymal stem cell. Due to their ease of access, DPSCs show great promise in regenerative medicine. However, the tooth extractions from which DPSCs can be obtained are usually performed at a period of life when donors would have no therapeutic need of them. For this reason, it is imperative that successful stem cell storage techniques are employed so that these cells remain viable for future use. Any such techniques must result in high post-thaw stem cell recovery without compromising stemness, proliferation, or multipotency. Uncontrolled-rate freezing is not a technically or financially demanding technique compared to expensive and laborious controlled-rate freezing techniques. This study was aimed at observing the effect of uncontrolled-rate freezing on DPSCs stored for 6 and 12 months. Dimethyl sulfoxide at a concentration of 10% was used as a cryoprotective agent. Various features such as shape, proliferation capacity, phenotype, and multipotency were studied after DPSC thawing. The DPSCs did not compromise their stemness, viability, proliferation, or differentiating capabilities, even after one year of cryopreservation at -80 °C. After thawing, they retained their stemness markers and low-level expression of hematopoietic markers. We observed a size reduction in recovery DPSCs after one year of storage. This observation indicates that DPSCs can be successfully used in potential clinical applications, even after a year of uncontrolled cryopreservation.

This review is to summarize and analyze the currently available knowledge concerning the action of oat (Avena sativa L.) consumption on obesity, as well as possible constituents and extra- and intracellular mediators responsible for its anti-obesity effect. The oat constituents could reduce fat storage via several mediatory mechanisms - brain centers regulating appetite, gastrointestinal functions, gut bacteria, fat synthesis and metabolism and maybe via changes in oxidative processes, steroid hormones receptors and adipose tissue vascularization. Several oat constituents (starch, fiber and beta-glucan) could have anti-obesity properties, whilst one oat constituent (starch or fiber) could affect fat storage via several mechanisms of action.

The article deals with some basic approaches to the management of secured access to data on external storage devices in real time. Basic steps of effective, safe and easy-to-use handling of personal and/or confidential data are described. Further, a method to protect logins and passwords during their registration into the various applications is presented in the article. Main features of several most commonly used software products are also described considering data protection when used online and stored on various storage devices. The main reasons to apply data protection in the area of medicine and health care services are stated in this article as well.
The study monitored the effect of various methods of treating pheasant carcasses after killing on the hygienic quality of the venison. Pithed pheasants treated by evisceration (n = 60), drawing (n = 60), or left untreated (n = 60) were stored for a period of 21 d at temperatures of 0, 7, and 15°C. For determination of biogenic amines, samples of breast and thigh muscles were taken on d 1, 7, 14, and 21 after killing of the pheasants. Biogenic amines were separated by reverse-phase liquid chromatography and consequently detected by tandem mass spectrometry. The sum of determined biogenic amine concentrations (cadaverine, putrescine, histamine, tyramine, tryptamine, phenylethylamine) was compared with the value of the index for meat of high hygienic quality (5 mg/kg). At a storage temperature of 0°C, the sum of biogenic amine concentrations did not exceed the value of 5 mg/kg in either breast or thigh muscle at any time during the storage period in untreated and drawn pheasants, and for a period of 14 d in eviscerated pheasants. At a storage temperature of 7°C, values lower than the limit of 5 mg/kg were recorded throughout the storage period in untreated pheasants, for a period of 14 d of storage in drawn pheasants, and for a period of just 7 d of storage in eviscerated birds. At the highest storage temperature (15°C), a value of 5 mg/kg was exceeded in eviscerated and untreated pheasants during the course of the first week of storage, and in drawn pheasants after the first week of storage. Our results indicate that the most suitable method of treatment to ensure high hygienic quality of the meat (assessed according to concentration of biogenic amines) for the longest period during the storage of pithed pheasants is to leave the pheasant carcasses untreated, followed by the drawing, with the least suitable method being the widely recommended method of evisceration.

The aim of this study was to compare hygiene status of wild boar meat (shoulder and leg) stored up to 21days at 0°C, 7°C or 15°C. The microbial counts increased gradually in the expected sequence of increasing storage temperatures, with TVC at the end of storage ranging from approx. 2logCFU/g (0°C) to 5logCFU/g (15°C). The lactic acid bacteria and psychrotrophic microflora didn't exceed 2logCFU/g and 2.5logCFU/g, respectively. Whereas odor of the meat stored at 0°C and 7°C was still acceptable at the end of storage, the odor of the meat stored at 15°C was barely acceptable after only 7d of storage and also the content of ammonia was significantly higher. Game meat obtained from animals hunted in the correct way and stored at low temperatures had good microbiological and hygiene status which could be maintained for more than 15days of storage.
